ASH2L-mediated H3K4me3 methylation governs palatal development by regulating WNT5B.

BACKGROUND: Orofacial clefts, particularly cleft palate, are among the most common congenital anomalies, imposing substantial functional impairment and psychosocial burden on affected individuals. Emerging evidence implicates epigenetic modifications, especially histone lysine methylation, as critical regulators of embryonic development, yet their precise roles in palatogenesis remain poorly defined. The H3K4 methyltransferase complexes, which require the core subunit ASH2L for full enzymatic activity, govern transcriptional activation during development; however, whether ASH2L-mediated H3K4me

Denosumab Therapy Duration and Osteonecrosis of the Jaw and Skeletal Events in Bone-Metastatic Prostate Cancer.

IMPORTANCE: Denosumab reduces skeletal-related events in bone-metastatic prostate cancer but carries a cumulative risk of medication-related osteonecrosis of the jaw (ONJ). The treatment duration that best balances these risks has not been established in clinical settings. OBJECTIVE: To evaluate the association between duration of denosumab treatment and the 3-year cumulative incidence of ONJ and skeletal events. DESIGN, SETTING, AND PARTICIPANTS: This nationwide retrospective cohort study emulated a target trial using Korean Health Insurance Review and Assessment Service data from January 1

Central nervous system, cranial, and cerebrovascular conditions beyond the Kimonis criteria in patients with Gorlin syndrome: a retrospective cohort study.

BACKGROUND: Gorlin syndrome is a multisystem developmental and cancer-predisposing disorder commonly caused by germline PTCH1 variants. The Kimonis clinical criteria capture the canonical phenotype; however, central nervous system (CNS), cranial, and cerebrovascular conditions documented beyond these criteria remain poorly defined. METHODS: We conducted a retrospective descriptive cohort study of 32 patients who fulfilled the Kimonis clinical criteria and received care at either or both hospitals in Japan. Clinical information through March 31, 2026, including sufficiently documented care els

CBCT features of florid cemento-osseous dysplasia with coexisting lesions at different maturation stages: a case report and literature review.

BACKGROUND: Florid cemento-osseous dysplasia (FLCOD) is a begin fibro-osseous lesion occurring in the tooth-bearing regions of the jaws and is characterized by replacement of normal bone with fibrous connective tissue and cementum-like mineralized material. Most patients are asymptomatic and are diagnosed incidentally during radiographic examinations, and the condition may be misdiagnosed because of overlapping imaging features with other jaw lesions. Because the disease is usually asymptomatic and exhibits imaging features that overlap with those of several odontogenic and inflammatory lesion

Radiographic Archetypes of Periapical Lesions from Deep Embeddings: A Baseline Weighted ResNet-18 Explainability Study on 3,924 Annotated Periapical Images.

AIMS AND BACKGROUND: Here, we present a simple endodontic imaging procedure that leverages expert extensible markup language (XML) annotations of publicly available periapical radiographs. MATERIALS AND METHODS: To develop a machine-learning model for automated severity classification, this work used a publicly accessible dataset of periapical radiographs with labeled lesions. After building a lesion-level index from XML annotations and aggregating bounding boxes into image-level features, 3,924 radiographs were split into stratified training, validation, and test sets. A weighted residual ne

Mendelian randomization of plasma lipidome and temporomandibular joint disorders.

Mendelian randomization (MR) was used to investigate the possible causal association between lipids (LP) and temporomandibular joint disorders (TMD). To investigate the causal relationship between circulating LP and TMD, we conducted a 2-sample MR analysis. Exposure data comprised summary statistics for 179 LP from the GeneRISK genome-wide association study (N = 7174 samples). Outcome data were derived from the FinnGen genome-wide association study for TMD (N = 185169 samples). Primary causal estimates were generated using inverse-variance weighted regression, with sensitivity analyses includi

Atypical Stafne bone defect: a systematic review.

OBJECTIVES: Atypically located Stafne bone defects (SBDs) may be challenging to identify because they deviate from the typical posterior mandibular presentation. This systematic review aims to analyze and synthesize the radiographic, anatomical, and histological characteristics of atypically located SBDs reported in the literature and to assist clinicians in orienting the diagnostic hypothesis. MATERIALS AND METHODS: A comprehensive literature search was conducted in the PubMed, Scopus, Embase, and Web of Science databases up to January 2026. Eligibility criteria included English-language cas

Association between malocclusion and temporomandibular joint disorders: evidence from a Mendelian randomization study.

BACKGROUND: Although malocclusion and temporomandibular joint disorders (TMD) frequently co-occur in clinical practice, controversy persists regarding whether this overlap reflects a pathophysiological causal relationship or incidental concurrence. This study aimed to investigate the potential association between malocclusion and TMD using a bidirectional two-sample Mendelian randomization (MR) approach. METHODS: Bidirectional two-sample MR was performed to evaluate the potential causal relationship between malocclusion and TMD. Summary statistics for malocclusion were obtained from the FinnG

Recognizing a Forgotten Disease: A Case Report of Tetanus in an Unvaccinated Adolescent.

Tetanus is a rare but potentially life-threatening vaccine-preventable disease in the United States. Because tetanus is now rarely encountered in the United States, diagnosis may be delayed despite classic clinical findings. We report the case of an unvaccinated sixteen-year-old Amish male who presented with progressive trismus and cervical rigidity thirteen days after tympanoplasty with mastoidectomy and incus debridement. Initial symptoms were attributed to a postoperative infectious process and treated with oral antibiotics. Progressive jaw stiffness and neck rigidity prompted emergency eva

Comparative efficacy of two novel bioceramic root canal sealers in the treatment of chronic apical periodontitis.

Chronic apical periodontitis (CAP) is a persistent periradicular inflammatory condition driven by intraradicular infection and remains a common indication for primary nonsurgical root canal therapy. This retrospective cohort study compared 2 calcium silicate-based bioceramic root canal sealers (C-Root SP vs iRoot SP) used during obturation in adults treated for CAP between January 2022 and December 2024. CAP was confirmed by compatible clinical findings and radiographic evidence of periapical radiolucency; cone-beam computed tomography (CBCT) was obtained when clinically indicated. The primary

Regulatory T Cells Across Oral Diseases: A Narrative Review on a Functional Perspective.

Regulatory T cells (Tregs) maintain peripheral immune tolerance and are involved in various oral diseases with markedly different characteristics. In the oral field, Tregs have largely been discussed through reports on their distribution and numbers, and interpretations of whether their role is protective or pathogenic. Current identification markers do not directly assess the suppressive function, and reports on this function remain limited. This review focuses on the suppressive function of Tregs in the organization of oral diseases. Cells may express FOXP3 without exerting stable suppressiv

Extracellular Matrix Remodeling and Glycoprotein Biomarkers Across Osteoarthritis, Rheumatoid Arthritis, and Temporomandibular Disorders: A Systematic Review and Integrative Analysis.

Osteoarthritis (OA), rheumatoid arthritis (RA), and temporomandibular disorders (TMDs) are heterogeneous joint diseases whose pathophysiology involves inflammation, tissue degeneration, and progressive functional decline. Although their etiologies differ, these conditions display overlapping molecular mechanisms related to extracellular matrix (ECM) remodeling, proteolytic activity, and immune signaling. This systematic review synthesizes current evidence on glycoprotein-associated and proteomic biomarkers identified in synovial fluid and saliva, predominantly derived from protein-level studie

[Application of sample-efficient neural network in predicting postoperative facial profiles in skeletal class Ⅱ orthognathic patients].

OBJECTIVE: Soft tissue prediction is of great importance for the surgical design and patients ' satisfaction in orthognathic surgery. Previous studies either rely on elastic mechanical models, such as finite element which involves time-consuming computations, or using artificial intelligence method which require insatiable large patient data. Therefore, in this study, we proposed a more efficient and lightweight method for facial morphology prediction and validated its accuracy. METHODS: A total of 85 skeletal class Ⅱ patients who underwent double jaw surgery performed by the same senior surg

High-order chromatin structures mediated by phase separation in cell senescence.

Liquid-liquid phase separation (LLPS) coordinates biomolecular condensates to establish three-dimensional chromatin architecture and epigenetic regulation. The progressive loss of epigenetic information is a fundamental, reversible driver of cellular senescence. In this review, we systematically synthesize emerging evidence on the spatiotemporal dynamics of LLPS-mediated chromatin compartmentalization during the aging process. We highlight how age-dependent alterations in histone, RNA, and DNA modifications disrupt multivalent interactions, leading to constitutive heterochromatin dissolution,
