Ticks are vectors of bacterial, viral, and protozoan pathogens and represent a growing public health concern in the United States and worldwide. In California, vector surveillance has largely focused on Ixodes pacificus, the major vector of the spirochete, Borrelia burgdorferi. However, the presence of multiple medically important Dermacentor species may harbor additional, understudied pathogens. To address this gap, broad pathogen screening was conducted on ticks collected from Siskiyou, Humboldt, Trinity, and Mendocino counties in Northern California, using multiplex quantitative PCR targeti

Colorado tick fever virus (CTFV) is a double-stranded RNA virus in the family Reoviridae. CTFV is the causative agent of Colorado tick fever (CTF), a febrile illness that can have mild to severe manifestations. CTFV is an underreported and underrecognized virus; similarly, the literature concerning neglected tick-borne viruses lacks standard protocols for viral research. Here we describe protocols and procedures for CTFV studies, including the quantification of the virus through viral plaque assay, generation and storage of CTFV stocks, and detection with reverse transcription-quantitative PCR

The Rocky Mountain wood tick, Dermacentor andersoni Stiles, is the most common human biting tick in the Rocky Mountain region of the USA. Although Rickettsia spp. and Colorado tick fever virus have been studied in D. andersoni in certain areas of their distribution, investigations of most other tickborne pathogens are lacking. Despite the history of Rocky Mountain spotted fever in the Bitterroot Mountains of western Montana, no comprehensive surveys of large numbers of ticks for Rickettsia spp. have been conducted there for >40 years. For the present study, host-seeking adult D. andersoni were

Sandflies are vectors for several tropical diseases such as leishmaniasis, bartonellosis, and sandfly fever. Moreover, sandflies exhibit species-specificity in transmitting particular pathogen species, with females being responsible for disease transmission. Thus, effective classification of sandfly species and the corresponding sex identification are important for disease surveillance and control, managing breeding/populations, research and development, and conducting epidemiological studies. This is typically performed manually by observing internal morphological features, which maybe an err

Toscana virus (TOSV) is an emerging but neglected human pathogen currently circulating around the Mediterranean basin including North Africa. Human illness ranges from asymptomatic or mild flu-like syndromes to severe neurological diseases such as meningitis or meningoencephalitis. Despite its significant impact, understanding of TOSV transmission and epidemiology remains limited. Sand flies (Diptera: Phlebotominae), specifically Phlebotomus perniciosus and Phlebotomus perfiliewi, are believed to be the primary vectors of TOSV. However, the spread of TOSV to new geographical areas and its dete

BACKGROUND: Sand fly-borne phleboviruses (SbPV) are globally distributed and pose potential public health risks. Despite increased detection in recent decades, detailed knowledge of their ecology, characteristics and clinical relevance remains limited. Many cases of SbPV infection likely go unreported or misdiagnosed due to limited awareness and the lack of standardized screening. The External Quality Assessment (EQA) reported herein was organized within the framework of the European Union CLIMOS (EU Climate Monitoring and Decision Support Framework for Sand Fly-borne Diseases Detection and Mi

Toscana virus (TOSV) and Schmallenberg virus (SBV) are arthropod-borne viruses from the Bunyaviricetes class, posing significant human and animal health threats. TOSV, endemic to the Mediterranean region, is a notable human pathogen detected in various animals, suggesting potential zoonotic reservoirs. SBV emerged in Europe in 2011, affecting ruminants and causing reproductive issues, with substantial economic implications. The rapid spread of both viruses underscores the need for novel antiviral strategies. Host defense peptides (HDPs), particularly those derived from scorpion venom, are gain

OBJECTIVES: Toscana virus (TOSV) is an emerging sandfly-borne virus, causing meningitis and encephalitis in Mediterranean and Middle Eastern regions. Only sporadic cases have been diagnosed in Austria over the past 20 years. We aim to identify and describe TOSV-associated aseptic meningitis (TOSV-AM) cases in Austria between 2006 and 2023.
METHODS: We report a series of four imported cases of TOSV-AM diagnosed in Salzburg between July and September 2023, after travel to Italy. In addition, we analyzed all TOSV cases identified by the Austrian National Reference Center (2006-2018). Diagnosis w

Determining the incubation period of Oropouche virus disease can inform clinical and public health practice. We analyzed data from 97 travel-associated cases identified by the Centers for Disease Control and Prevention (n = 74) or the GeoSentinel Network (n = 13) and 10 cases from published literature. Using log-normal interval-censored survival analysis, we estimated the median incubation period to be 3.2 (95% CI 2.5-3.9) days. Symptoms developed by 1.1 (95% CI 0.6-1.5) days for 5% of patients, 9.7 (95% CI 6.9-12.5) days for 95% of patients, and 15.4 (95% CI 9.6-21.3) days for 99% of patients

BACKGROUND: The 2024 Oropouche virus (OROV) outbreak in Brazil raised public health concerns due to its unprecedented rapid spread, high incidence, and potential neurological complications. OROV symptoms overlap with locally endemic arbovirus diseases, like dengue virus (DENV), complicating diagnosis. The study aimed to compare clinical, laboratory, and immunological profiles in OROV and DENV cases, crucial for improving diagnosis and management.
METHODS: This study analyzed 51 OROV and 78 of DENV cases consecutively enrolled in Manaus, Amazonas, Brazil, and monitored for 28 days. OROV diagno

Oropouche virus (OROV) is an emerging arbovirus responsible for Oropouche fever, also known as sloth fever, a febrile illness that can lead to recurrent outbreaks in affected regions. Endemic to parts of South and Central America, OROV is primarily transmitted by biting midges (Culicoides paraensis), although mounting evidence implicates mosquitoes, particularly the Culex and Aedes species, as additional vectors. Recent ecological disturbances-such as deforestation, urbanization, and climate change-have driven significant shifts in vector population dynamics, contributing to the expanded geogr

BACKGROUND: Toscana virus (TOSV) is a sand fly-borne phlebovirus causing central nervous system (CNS) infection in Mediterranean countries, during summer season. However, clinical aspects of the disease caused by this virus are poorly known by clinicians, so that its prevalence is probably underestimated due to a lack of diagnosis.
STUDY DESIGN: The data was gathered from all available case series and retrospective studies identifying TOSV as the causative viral agent. The informations of age, sex, clinical characteristics, laboratory findings, imaging results and clinical outcomes of TOSV inf

Toscana virus (TOSV), a Phlebovirus transmitted by sandflies, is a leading cause of aseptic meningitis in the Mediterranean region. Despite its clinical significance, underreporting and limited availability of complete genomic data hinder a thorough understanding of its genetic diversity and evolution. This study presents a novel amplicon-based whole-genome sequencing (WGS) method using Illumina library preparation kits and proprietary software to optimize workflows and enhance bioinformatic analyses. Primers targeting TOSV lineage A genomes were designed with PrimalScheme to generate 400 bp a

Toscana virus (TOSV), belonging to Phenuiviridae family, is circulating in most Mediterranean countries and is transmitted to humans by infected female sand flies. While most infections are asymptomatic, TOSV is considered as a leading cause of meningitis and encephalitis in humans during summer. Three TOSV genotypes (named A, B, and C) have been identified, although no virus strain belonging to lineage C has been isolated so far. To date, the relationship between TOSV genetic diversity and viral pathogenicity or replication capacity remains unknown. This study aimed to compare two TOSV strain
