Crystal structure and functional characterization of the microalgal carotenoprotein AstaP-pink1.

The recently discovered family of microalgal water-soluble astaxanthin-binding proteins (AstaPs) functions in carotenoid sequestration and transfer. We present the crystal structure and characterization of AstaP-pink1, a homolog from Scenedesmus sp. We show that despite low sequence identity with the known AstaP-orange1, AstaP-pink1 also binds various xanthophylls, but in contrast to AstaP-orange1, induces a pronounced bathochromic shift and a near-UV spectral bump upon binding ketocarotenoids, which is reminiscent of photoactivated Orange Carotenoid Protein (OCP). Mutagenesis and domain-swap

Clinical efficacy of orifice barriers in endodontics: A systematic review.

OBJECTIVES: Orifice barriers (OBs) are commonly placed as sub-seals following root canal treatment, to resist microbial leakage if the primary coronal seal fails. However, despite numerous in vitro studies, clinical evidence supporting their efficacy is limited. This is the first systematic review of the in vivo endodontic literature evaluating OB effectiveness in root filled teeth (RFT), addressing a critical research gap. DATA SOURCES: MEDLINE, Embase, CENTRAL, Web of Science and Scopus were systematically searched from inception through 16th May 2025. Articles were screened using Covidence

Integrative Computational and Experimental Approaches Reveal the Protective Role of Moderate Caffeine Intake Against Apical Periodontitis Induced Bone Loss.

AIM: To investigate whether moderate systemic caffeine intake modulates the progression of apical periodontitis (AP) and associated alveolar bone loss, combining in vivo rat experiments with in silico molecular docking to explore potential mechanisms. METHODOLOGY: Male Wistar rats were randomly assigned to four groups (n = 8 per group): control, caffeine, AP, AP + caffeine. AP was induced by pulp exposure of mandibular first molars and allowed to develop for 28 days. Animals in caffeine groups received 10 mg/kg/day by orogastric gavage during the experimental period. The antioxidant capacity

Microbiome of Apical Intracanal and Extraradicular Biofilms From the Same Roots of Teeth With Persistent Apical Periodontitis: An Observational Study.

AIM: Bacterial biofilms around the apex are crucial in disease progression and persistence of apical periodontitis. While intracanal biofilms initiate infection, extraradicular biofilms contribute to treatment resistance and persistence. Thus, a comprehensive understanding of these biofilms may help elucidate mechanisms underlying persistent apical periodontitis. Therefore, in this study, we aimed to compare the microbiome and predicted functional profiles in matched apical root canals with those of extraradicular biofilms associated with persistent apical periodontitis. METHODOLOGY: Seventee

The Global Periapical Health Study: A Big Data CBCT Analysis of Periapical Pathology across 54 Countries.

INTRODUCTION: Periapical pathology, a common outcome of pulpal infection or failed endodontic therapy, remains underexplored globally. Most studies are small-scale and based on two-dimensional radiographs. This study aimed to provide a standardized worldwide estimate of its prevalence using cone-beam computed tomography (CBCT). METHODS: A cross-sectional, multicenter study was conducted across 54 countries, each represented by one calibrated examiner. Examiners consecutively assessed 3,500 roots from pre-existing CBCT scans, yielding data from 189,000 roots (138,536 teeth) of 6,688 patients.

A Retrospective Analysis of the Diagnostic Accuracy in Diagnosing OPMD and Benign Oral Lesions.

OBJECTIVE: The clinical suspicion of a neoplastic lesion necessitates prompt diagnostic assessment through oral biopsy. However, the failure to accurately recognize an oral potentially malignant disorder (OPMD) or the misdiagnosis of a benign lesion can lead to diagnostic delays with significant impact on the patient's health. This study aimed to evaluate the diagnostic accuracy of clinical diagnosis compared to the histopathological diagnosis, focusing on OPMDs and suspected benign lesions in an Italian university hospital. METHODS: A retrospective analysis was conducted on patients with a c

DNA tetrahedron-mediated vascular targeted delivery of astragaloside IV enhances distraction osteogenesis via PI3K/AKT/FOXO pathway.

Bone regeneration during distraction osteogenesis (DO) remains limited by insufficient vascularization, which delays consolidation and compromises clinical outcomes. Here, we developed a vascular-targeted DNA nanoplatform (TDN@Ast-AptCD34) for the delivery of Astragaloside IV (Ast), a natural compound with pro-angiogenic and osteogenic properties. The nanoplatform integrated a DNA tetrahedron carrier with a vascular-targeting aptamer (AptCD34) and was further incorporated into a biocompatible mHA-PVA hydrogel for localized and sustained release. Physicochemical analyses confirmed its stable st

Ultrastable, Magnet-Insensitive Perfluoropolyether-Based Oleogel for Reliable Sensing in Magnetic Resonance Imaging.

In a strong magnetic field environment like Magnetic Resonance Imaging (MRI), traditional electronic conductors may interfere with imaging, whereas ionic conductors exhibit superior interference resistance, because the ion migration is minimally affected by the Lorentz force. Here, we developed a perfluoropolyether-based oleogel (PFOG) with ionic conductivity. As the major component in the network, perfluoropolyether endows PFOG with excellent biological, physical, and chemical stability, ensuring its compatibility in diverse environments. Additionally, the PFOG-based sensor is capable of effe

DNA Supramolecular Hydrogel Alters Exosomal microRNA Atlas and Paracrine Secretory Profile of MSCs to Promote Bone Remodeling and Ameliorate Osteoradionecrosis of the Jaw.

Osteoradionecrosis of the jaw (ORNJ) is a severe complication of head and neck radiotherapy, characterized by a hostile tissue microenvironment that hinders healing. To address the limitations of mesenchymal stem cell (MSC) therapy, such as rapid dispersion and clearance, we developed an injectable, shear-thinning DNA supramolecular hydrogel (DNASH) as a 3D scaffold. The DNASH matrix significantly prolonged the local retention of MSCs and their exosomes for over 3 weeks. Crucially, the scaffold actively reprogrammed the MSC secretome by modulating the FAK‑Integrin and Hippo‑YAP mechanotransduc

Consensus recommendations in the management of jaw (gnathic) osteosarcoma.

Gnathic osteosarcoma (OS), which includes mandibular and maxillary jaw OSs, account for 6%-9% of OS. Single-institutional and multi-institutional retrospective studies, as well as population-level databases, suggest that clinical differences exist among gnathic OS and OS of other sides, including other craniofacial OS. To date, no specific prospective studies of gnathic OS have been reported, and aspects of clinical management are controversial. Some elements of care are aligned with extragnathic OS, e.g., margin negative (R0) surgery, whereas others, such as chemotherapy and radiation, are no
