Broad-spectrum antiviral activity of the synthetic rocaglate zotatifin against Mayaro virus and other viruses.

Viruses pose a significant threat to global public health, yet therapeutic options remain limited. Zotatifin, a synthetic rocaglate targeting eukaryotic initiation factor 4A (eIF4A), inhibits viral protein synthesis and potentially triggers interferon responses; however, its broad-spectrum antiviral potential remains unclear. We evaluated zotatifin's cytotoxicity using MTT assays and assessed its antiviral activity through plaque-forming assays, Western blot analysis, immunofluorescence, and flow cytometry across multiple cell lines and virus strains. Zotatifin demonstrated superior tolerabili

Evaluation of the Antimicrobial Effect of Silver Nanoparticles Incorporated Into Heat-Cured Acrylic Denture Base Resin: A Clinical Study.

INTRODUCTION: Denture-associated candidal stomatitis is one of the most common inflammatory conditions affecting complete denture wearers and is primarily associated with Candida albicans colonization of the denture base. Conventional antifungal therapies are often associated with recurrent infections and patient non-compliance. The incorporation of silver nanoparticles (AgNPs) into polymethyl methacrylate (PMMA) denture base resin has emerged as a promising approach to provide sustained antimicrobial activity and reduce fungal biofilm formation. This study aimed to evaluate the antimicrobial

Exosomal miR-375-3p mediated lipid metabolism, ferritinophagy and CoQ-dependent pathway contributes to the ferroptosis of keratinocyte in SJS/TEN.

Stevens-Johnson syndrome (SJS) and toxic epidermal necrolysis (TEN) manifest life-threatening cutaneous adverse drug reactions characterized by keratinocyte death. Previous studies have indicated that apoptosis and necroptosis are implicated in SJS/TEN pathogeneses. However, other forms of cell death involved in this process remain unidentified. Ferroptosis, cell death driven by iron-dependent lipid peroxidation, has been implicated in various human diseases. In this study, the identification of ferroptosis and the potential effects of ferroptosis on SJS/TEN were investigated. We demonstrated

Short 5' UTRs serve as a marker for viral mRNA translation inhibition by the IFIT2-IFIT3 antiviral complex.

Recognition of "non-self" nucleic acids, including cytoplasmic dsDNA, dsRNA, or mRNAs lacking proper 5' cap structures, is critical for the innate immune response to viruses. Here, we demonstrate that short 5' untranslated regions (UTRs), a characteristic of many viral mRNAs, can also serve as a molecular pattern for innate immune recognition via the interferon-induced proteins IFIT2 and IFIT3. The IFIT2-IFIT3 heterodimer, formed through an intricate domain swap structure resolved by cryo-EM, mediates viral mRNA 5' end recognition, translation inhibition, and ultimately antiviral activity. Tuning the tropism and infectivity of SARS-CoV-2 virus-like particles for mRNA delivery.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) virus-like particles (VLPs) are ∼100-nm-sized bioinspired mimetics of the authentic virus. We undertook molecular engineering to optimize the VLP platform for messenger RNA (mRNA) delivery. Cloning the nucleocapsid protein upstream of M-IRES-E resulted in a three-plasmid (3P) VLP system that displayed ∼7-fold higher viral entry efficiency compared with VLPs formed by co-transfection with four plasmids. More than 90% of human ACE2-expressing cells could be transduced using these 3P VLPs. Viral tropism could be programmed by switching

Evaluating the Efficacy of Rose Bengal as a Photosensitizer in Antimicrobial Photodynamic Therapy Against Candida albicans: A Systematic Review.

Candida albicans is a significant pathogen in various fungal infections, including oral candidiasis and denture stomatitis. As antifungal resistance rises globally, there is an urgent need for alternative treatment strategies. Antimicrobial photodynamic therapy (aPDT), utilizing a photosensitizer and light to produce reactive oxygen species (ROS), has emerged as a promising approach. Rose Bengal (RB), a xanthene dye, exhibits a high singlet oxygen quantum yield, making it a candidate for aPDT. However, its efficacy in C. albicans treatment has been inconsistent, particularly against biofilm-as

Synergistic exacerbation of oral mucositis caused by IL-23 deficiency and oral Candida albicans exposure.

Targeted head and neck irradiation (HNI) used for cancer therapy causes mucosal damage and immune dysregulation, leaving cancer patients highly susceptible to infections of the oral mucosa. Oropharyngeal candidiasis (OPC) is an opportunistic infection caused by Candida albicans, a commensal fungus found in up to 80% of the population at any given time. High colonization levels of C. albicans are known to worsen damage caused by HNI. The interleukin-23 (IL-23)-T-helper 17 (Th17) axis is a central and non-redundant mediator of immunity to OPC, and anti-cytokine biologics targeting IL-23 have com

Long-term antifungal drug-delivery denture materials with plasma-initiated surface functionalization.

Conventional polymethylmethacrylate (PMMA)-based denture materials are susceptible to colonization and biofilm-formation by Candida species, leading to Candida-associated denture stomatitis (CADS), a recurring fungal infection that affects a large patient population. In this report, we developed a two-step, plasma-initiated surface functionalization approach to covalently bond poly (methacrylic acid) (PMAA) onto denture resins. Various reaction conditions were investigated and the effects of PMAA grafting yields on the physical properties of the resulting denture materials were determined. The
