Expert consensus on the diagnosis and management of tooth developmental anomalies.

Tooth developmental anomalies are a group of disorders caused by unfavorable factors affecting the tooth development process, resulting in abnormalities in tooth number, structure, and morphology. These anomalies typically manifest during childhood, impairing dental function, maxillofacial development, and facial aesthetics, while also potentially impacting overall physical and mental health. The complex etiology and diverse clinical phenotypes of these anomalies pose significant challenges for prevention, early diagnosis, and treatment. As they usually emerge early in life, long-term manageme

Huntingtin Aggregate-Responsive Autophagy Gene Circuit Mitigates Disease Pathology in R6/2 Mice.

The accumulation of mutant huntingtin (mHTT) aggregates drives the pathology of Huntington's disease (HD), yet therapies capable of distinguishing toxic species from wild-type proteins remain elusive. Here, a synthetic gene circuit, termed ARAA, was engineered to couple the preferential recognition of aggregated polyQ species to the on-demand activation of autophagy. Utilizing a repurposed bacterial NarX-NarL system fused with a conformation-sensitive intrabody, the circuit detects pathological polyQ conformers and triggers the transcriptional expression of the master autophagy regulator TFEB.

Finite Element Analysis in Cleft Lip Nasal Deformity: A Narrative Review from Biomechanical Insights to Clinical Translation.

Cleft lip nasal deformity poses significant surgical challenges due to its multidimensional anatomical distortions and dynamic biomechanical interactions. Conventional approaches relying on two-dimensional imaging and empirical judgment fail to address the three-dimensional tissue mechanics governing nasal symmetry and airway function. This review demonstrates how finite element analysis (FEA) transforms cleft nasal reconstruction by integrating biomechanical precision with clinical translation. Patient-specific modeling based on CT/MRI data enables FEA to quantify stress-strain distributions

Effect of Nasoalveolar Molding on Skeletal Growth in Individuals With Bilateral Cleft Lip and Palate During the Mixed Dentition Stage.

BACKGROUND: Nasoalveolar molding (NAM) is widely used to improve surgical outcomes in cleft lip and palate by reshaping nasal cartilage and aligning the alveolar cleft. However, its lasting impacts on skeletal growth in individuals with bilateral cleft lip and palate (BCLP) remain unclear. METHODS: This retrospective study reviewed individuals with nonsyndromic BCLP born between 1997 and 2017. Two groups were compared: NAM-treated (n=33) and control (n=45). Lateral cephalometric x-rays taken in the mixed dentition before orthodontic intervention or alveolar bone grafting were compared between
