Top tips for the management of patients with cleft lip and palate in the primary care dental practice.

Cleft lip and/or palate (CL/P), affecting about 1.35 per 1,000 live births globally and 650 annually in the UK, requires a multidisciplinary hub-and-spoke approach. This marginalised group suffer disproportionately elevated levels of dental decay. General dental practitioners (GDPs) are crucial in preventive care, early intervention, and ongoing oral health management. This article provides guidance for GDPs on early patient registration, specialist communication, and tailored preventive strategies targeting caries and oral hygiene. Key measures include dietary advice, specialised feeding tool

NCOA4-mediated ferritinophagy: emerging role and novel therapeutic target in precision oncology.

Iron is vital for life but can be toxic in excess by forming reactive oxygen species. Ferroptosis, a type of regulated cell death, relies on iron-dependent lipid peroxidation and requires a labile iron pool (LIP) in cells. Ferritin stores iron safely, and its degradation increases the LIP. Ferritinophagy, the autophagic breakdown of ferritin, is crucial for releasing stored iron to trigger ferroptosis. This review examines ferritinophagy's molecular mechanisms, highlighting NCOA4 (nuclear receptor coactivator 4) as the main receptor targeting ferritin for lysosomal degradation. It also discuss

Effect of age and sex on cardiovascular outcomes among patients with chronic lymphocytic leukemia treated with Bruton tyrosine kinase inhibitors: a propensity matched analysis.

The combined effect of age and sex on the risk of atrial fibrillation (AF) in patients receiving Bruton tyrosine kinase inhibitors (BTKi) is unclear. To address this knowledge gap, propensity score matching was applied to the TriNetX database to generate matched cohorts of males and females with chronic lymphocytic leukemia (CLL) who received ibrutinib (n = 5312) or acalabrutinib (n = 3110). AF events during the first three years of BTKi therapy increased with age and were more common in males compared to females. For ibrutinib, nearly one in three males aged ≥75 developed AF compared with few

Temporally Programmed and Laser-Triggered Immunomodulatory Hydrogel Enables Synergistic Wound Healing and Prevention of Postoperative Tumor Recurrence.

Surgery remains the predominant therapeutic option for patients with colorectal cancer. However, residual microscopic lesions and the immunosuppressive microenvironment considerably elevate the risk of postoperative tumor recurrence. Additionally, intestinal fistulas are the most common complication of surgery and severely impair the quality of life for patients. Current postoperative adjuvant therapies are limited by their systemic toxicity, inadequate immunomodulatory potential, and poor tumor targeting capacity, resulting in limited therapeutic efficacy. To address these challenges, a multi

Advanced Glycation End Products Induce Cuproptosis and Then Promote the Activation and Migration of Cardiac Fibroblasts.

Excessive advanced glycation end products (AGEs) can lead to cardiovascular diseases such as myocardial fibrosis (MF). Although studies have found that AGEs induce cuproptosis, and cuproptosis promotes fibrosis, it has not been confirmed whether AGEs promote MF through cuproptosis. AGEs increased intracellular copper levels, promoted the expression of solute carrier family 31 member 1 (SLC31A1), and downregulated the expression of ferredoxin 1 (FDX1), lipoic acid synthetase (LIAS), lipoylated (Lip)-dihydrolipoamide S-succinyltransferase (DLST), and Lip-dihydrolipoamide S-acetyltransferase (DLA

Targeting keratin 6 A overcomes gemcitabine resistance by restoring equilibrative nucleoside transporter 1 and TAM-mediated metabolic compensation in pancreatic cancer.

BACKGROUND & AIMS: Gemcitabine remains a cornerstone chemotherapy for pancreatic cancer, yet intrinsic and acquired resistance severely limits its efficacy. This study aimed to identify key drivers of gemcitabine resistance and develop a targeted strategy to restore chemosensitivity. METHODS: Public datasets of gemcitabine-resistant pancreatic cancer cells were integrated to identify resistance-associated genes, followed by RT-qPCR validation in resistant cell models. Clinical relevance was evaluated using pancreatic cancer cohorts and tissue microarrays with survival and multivariable analys

Prevalence of orofacial cleft and its associated factors among newborns in Kembata zone, central Ethiopia 2024.

INTRODUCTION: Orofacial clefts are common congenital malformations that impair structural, functional, and psychosocial outcomes. They may be identified at birth or later in life, often requiring surgical correction, which is caused by defects in the integrity and unity of fetal facial structures. These disorders occur between the medial nasal and maxillary processes (cleft lip) and between the maxillary processes (cleft palate). It affects nearly every aspect of life for the exposed newborns, as they involve both structural and functional abnormalities of the facial region. These defects sign

The Evolution of Gene Sequencing Technologies: Unveiling Genetic Architecture of Nonsyndromic Orofacial Clefts.

Nonsyndromic orofacial clefts (NSOC) are common congenital craniofacial developmental defects. Current evidence suggests that genetic factors, environmental exposures, and their interactions jointly contribute to the development of the disease. Owing to the high heritability of NSOC, identifying susceptibility genes and loci is a central focus of etiological research. This review summarizes key findings in the identification of NSOC susceptibility genes and loci across successive stages of sequencing technology development. With the evolution of sequencing approaches, from Sanger sequencing to

Stem cell-derived in vitro models of human cerebellar development.

Understanding human cerebellar development and pathology remains challenging due to its complexity, unique protracted developmental timeline, and notable species-specific differences. Advances in human stem cell technologies have opened new windows of opportunity for exploring these processes through improved in vitro models, providing insights into human-specific mechanisms that traditional animal models may fail to recapitulate. Here, we review recent advancements in modelling physiological and pathological processes of human cerebellar development. We first outline key features of cerebella

Adulthood stressful life events as predictors of incident cardiovascular disease: insights from two prospective cohorts.

BACKGROUND: Stressful life events (SLEs) in adulthood are potential risk factors for cardiovascular disease (CVD), yet evidence remains inconsistent. This study examined the association between adulthood SLEs and incident CVD and evaluated mediation by depression, physical inactivity, and smoking. METHODS: We analyzed harmonized data from two nationally representative aging cohorts: the US Health and Retirement Study and the English Longitudinal Study of Ageing. SLEs were assessed at baseline, and incident CVD was physician-diagnosed. We used Cox and restricted mean survival time (RMST) regres

Resorbable plate as a novel method for tip derotation in intermediate rhinoplasty for bilateral cleft lip nasal deformity.

BACKGROUND: Bilateral complete cleft lip and nasal deformity are characterized by a flattened nasal tip, splayed nostrils, and a shortened columella. Efforts to improve nasal tip projection during intermediate rhinoplasty may inadvertently induce cephalic rotation. We selectively applied a resorbable plate secured to the nasal dorsum in a derotation configuration for patients with a preoperative nasal tip angle ≥120° and evaluated its effect on postoperative cephalic rotation of the nasal tip. METHODS: A retrospective analysis was performed in 15 patients with bilateral complete cleft lip nas

Association Between Soft Tissue Facial Profile and Dentoskeletal Changes in High-Angle Skeletal Class II Patients After Orthodontic Treatment: A Retrospective Cephalometric Study.

Orthodontic treatment of high-angle skeletal Class II patients often leads to unsatisfactory facial profiles, highlighting the need to understand the relationship between dentoskeletal changes and soft tissue profile alterations. This retrospective study examined female Southern Chinese patients (n = 21, mean age 24.7 ± 4.8) with high-angle skeletal class II. Pre- and post-treatment lateral cephalograms were analysed for linear, angular, and proportional changes. Correlations between facial profile changes and dentoskeletal modifications were assessed using Pearson and Spearman's coefficients.
