Selecting the optimal method for assessment of hypertension-mediated organ damage: a personalized approach.

AIMS: Susceptibility to hypertension-mediated organ damage (HMOD) is influenced by genetics, age, gender, and additional cardiovascular risk factors and comorbidities in the individual patient. All major hypertension guidelines recommend assessment of HMOD to identify high cardiovascular risk. However, in clinical practice, it may be difficult to choose the optimal strategy and diagnostic tools for the individual patient. METHODS AND RESULTS: We reviewed recommendations on HMOD assessment in the 2024 European Society of Cardiology, the 2023 European Society of Hypertension, the 2025 American

Integrated Structural and Glycoproteomic Profiling Reveals Protein Conformational Remodeling and Biomarkers Across Alzheimer's Disease Progression.

Alzheimer's disease (AD) is characterized by progressive neurodegeneration and protein misfolding, yet the structural dynamics of proteins and their post-translational modifications during disease progression remain poorly understood. Here, we present an integrated structural and glycoproteomic analysis of paired serum and cerebrospinal fluid (CSF) samples from individuals across three clinical stages: normal cognition, mild cognitive impairment, and AD. Using limited proteolysis mass spectrometry (LiP-MS) combined with high-field asymmetric waveform ion mobility spectrometry and data-independ

AI-based multiomics profiling reveals complementary omics contributions to personalized prediction of cardiovascular disease.

Genomics, metabolomics, and proteomics offer complementary insights into cardiovascular disease (CVD) risk. Leveraging UK Biobank data, we introduce the CardiOmicScore, a multitask deep learning framework, to learn disease-specific proteomic (ProScore) and metabolomic (MetScore) risk scores for the six most common CVDs by profiling 2920 proteins and 168 metabolites. Experiments demonstrate that ProScore and MetScore are strong sole CVD risk predictors (C-index range: 0.69-0.82 for ProScore and 0.64-0.74 for MetScore), and can significantly enhance risk prediction across CVDs up to 15 years pri

Sentinel lymph node biopsy in cT1-2N0 lip squamous cell carcinoma: site-specific diagnostic accuracy, anatomic mapping, and survival impact.

BACKGROUND: Optimal management of the clinically node-negative (cN0) neck in early-stage (cT1-2) lip squamous cell carcinoma (SCC) remains controversial. Sentinel lymph node biopsy (SLNB) offers a minimally invasive staging alternative to observation or elective neck dissection, but its diagnostic accuracy, lymphatic mapping patterns, and survival impact in lip SCC are not well-established. METHODS: We conducted a retrospective study of 101 patients with cT1-2N0 lip SCC treated at a tertiary cancer center (2005–2022), comparing SLNB-guided management (n = 46) to observation (n = 55). Diagnosti

Digital Twins for Predictive Modelling of Thrombosis and Stroke Risk: Current Approaches and Future Directions.

Thrombosis drives substantial global mortality across atrial fibrillation, venous thromboembolism, and atherosclerosis. However, clinical scores treat risk as a static variable and omit evolving comorbidities, functional biomarkers, anatomy, and treatment exposure, leading to misclassification and preventable events. This statement advances a unified scientific agenda for patient-specific digital twins that dynamically integrate multimodal longitudinal data with mechanistic insight to predict thrombogenesis risks. We position these digital twins as hybrid models anchored in physics and data-dr

Involvement of reduced ENPP1 function in the pathogenesis of ossification of the posterior longitudinal ligament.

Ossification of the posterior longitudinal ligament (OPLL) is an ectopic ossification disorder with incompletely understood pathophysiology. In a previously reported 50-patient OPLL cohort, ectonucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1) exonic heterozygous variants were identified in 7 patients (14%), and plasma inorganic pyrophosphate (PPi), a biochemical marker reflecting ENPP1 function, was lower in patients with OPLL than in cervical osteoarthritis controls regardless of ENPP1 variant status. However, the clinical significance of ENPP1 variants and reduced PPi in OPLL, as well

Interventions for treatment of herpes labialis (cold sores on the lips).

RATIONALE: Herpes labialis (also known as cold sores or fever blisters) is a highly prevalent recurrent infection caused by the herpes simplex virus. Recurrences arise from varied triggers. Standard treatments usually involve nucleoside antivirals. Several studies have investigated alternative treatments such as zinc, honey, and photodynamic therapy for their potential to shorten lesion duration and relieve symptoms, but most reviews exclude these therapies. We wished to address this evidence gap by evaluating both conventional and alternative approaches, synthesising evidence on efficacy and

[Chinese expert consensus on the clinical application of lung biopsy in interstitial lung disease].

Interstitial lung disease (ILD) comprises a highly heterogeneous group of pulmonary disorders, whose diagnosis often requires the integration of clinical, radiologic, and pathologic evidence. Lung biopsy is a key method for obtaining a histopathologic diagnosis, but unified standards are lacking for determining indications, selecting biopsy techniques, managing patients perioperatively, and integrating pathologic and clinical information. To address these gaps, the Respiratory Physicians Branch of the Chinese Medical Doctor Association and the ILD Group of the Chinese Thoracic Society spearhea

Ginsenoside Rh2- functionalized liposomes enhanced BRD4-PROTAC delivery and antitumor efficacy via improved tumor targeting and ECM remodeling.

PROTAC technology leverages the ubiquitin-proteasome system to selectively degrade target proteins, presenting a novel strategy for anticancer therapy. ARV825, a BRD4-targeting PROTAC, exerts potent antitumor effects by degrading BRD4, thereby suppressing Bcl-2 and PD-L1 expression, inducing apoptosis, and enhancing T cell-mediated immunity. However, its clinical translation is hindered by poor solubility, low membrane permeability, and off-target effects. While conventional liposomes (lip) improved ARV825 delivery, their efficacy remained limited by insufficient tumor targeting and collagen-r

Inhalable stealth liposomes improve peptide delivery for pulmonary fibrosis treatment.

Pulmonary fibrosis is a life-threatening lung disease with limited therapeutic options and suboptimal clinical outcomes. Peptide-based agents such as CSP7, a heptapeptide derived from caveolin-1, have shown promising antifibrotic activity with low systemic toxicity. However, effective inhaled administration of CSP7 is impeded by physiological barriers in fibrotic lungs, including alveolar edema fluid and clearance by inflammatory macrophages. Here, we report a high-loading stealth liposomal system designed to enhance pulmonary delivery of CSP7 by inhalation. CSP7 was covalently conjugated to a
