Inhibitory role of arecaidine on PPARγ signaling in oral mucosa: Mechanistic insights from transcriptome and experimental analysis.

OBJECTIVE: This study aims to elucidate the effects of arecaidine on oral mucosa through RNA sequencing (RNA-Seq) combined with in vivo and in vitro experimental validation. METHODS: Based on transcriptomic analysis, we preliminarily explored the molecular targets and mechanisms by which arecaidine influences oral mucosa. Subsequent validation was performed using arecaidine-treated human primary oral mucosal fibroblasts. RESULTS: In vivo experiments revealed that the arecaidine-treated group exhibited significantly restricted oral cavity opening compared to the control group, with markedly r

Malignant Transformation Rate of Oral Premalignant Disorders: A Large Database Analysis.

OBJECTIVES: Oral leukoplakia (OL), oral lichen planus (OLP), and oral submucous fibrosis (OSF) are precancerous lesions known to progress to oral squamous cell carcinoma (OSCC). The transformation rates to OSCC are controversial and have been based on small, heterogeneous, single-institution studies. We present the largest study to date looking at the malignant transformation rates of these oral premalignant disorders (OPMD). METHODS: We conducted a retrospective cohort study within the TriNetX database. Patients aged ≥ 18 with a diagnosis of OL, OLP, and OSF between 2008 and 2018 were includ

INHBA+ macrophages and Pro-inflammatory CAFs are associated with distinctive immunosuppressive tumor microenvironment in submucous Fibrosis-Derived oral squamous cell carcinoma.

Transcriptomic and metabolic profiles of tumor cells and stromal cells in oral squamous cell carcinoma (OSCC)-derived from oral submucosal fibrosis (OSF) (ODSCC) have been reported. However, the complex intercellular regulatory network within the tumor immunosuppressive microenvironment (TISME) in ODSCC remains poorly elucidated. Here, we utilized single-cell RNA sequencing (scRNA-seq) and spatial transcriptomics (ST) data from GEO database and multiple immunofluorescence staining (mIF) to reveal distinctive TISME of ODSCC. Results found that compared to OSCC without OSF history (NODSCC), OSCC

HPV-Negative Oral Squamous Cell Carcinoma Arising from Oral Submucous Fibrosis with p16INK4A Positivity and Cellular Senescence: A Case Report.

BACKGROUND: Oral Submucous Fibrosis (OSF) is a chronic, progressive, and potentially malignant disorder primarily associated with areca nut chewing. While Oral Squamous Cell Carcinoma (OSCC) typically develops in the tongue and floor of the mouth, its occurrence in the buccal mucosa in the context of OSF is less common. The molecular mechanisms underlying OSF-associated OSCC remain unclear. p16INK4A is widely recognized as a surrogate marker for HPV-driven carcinogenesis; however, its role as an indicator of cellular senescence is increasingly acknowledged. Given that senescence contributes to

Evaluation of stromal mucin expression in oral submucous fibrosis and its progression to oral squamous cell carcinoma.

INTRODUCTION: Oral potentially malignant conditions (OPMDs) are abnormalities in the oral mucosa linked to a higher chance of developing oral cancer. Oral submucous fibrosis (OSMF) is a chronic condition causing tissue fibrosis, collagen deposition, and scar tissue production. The highest prevalence of OSMF is reported in Southeast Asian countries, with areca nut consumption being a significant factor. MATERIALS AND METHODS: This retrospective study involved 20 histopathologically confirmed cases: 10 of oral submucous fibrosis (OSMF) and 10 of oral squamous cell carcinoma (OSCC) arising in th

The Role of Klotho in Oral and Maxillofacial Diseases: Mechanisms and Research Progress.

Klotho, an anti-aging protein, has been extensively studied in systemic conditions such as chronic kidney disease and cardiovascular disorders. In recent years, its pivotal protective role and clinical significance in various oral and maxillofacial diseases have been increasingly demonstrated. It has been demonstrated that Klotho regulates oxidative stress, apoptosis, inflammation, and fibrosis via multiple molecular signaling pathways, including Nrf2, NF-κB, PI3K/Akt/FoxO1, insulin/IGF-1, FGF/FGFR, and Wnt/β-catenin. Consequently, these regulatory effects have been observed in conditions such

Formulation, in-vitro characterization and clinical evaluation of curcumin in-situ gel for treatment of oral submucosal fibrosis (OSF).

BACKGROUND: Oral submucosal fibrosis (OSF) is a chronic oral mucosal disease with a cancerization tendency, characterized by restricted mouth opening and dysphagia. Currently, the mainstay treatment is still the local corticosteroid injection. Curcumin can reconstruct the redox balance, inhibit epithelial-mesenchymal transition (EMT), and suppress the excessive deposition of extracellular matrix (ECM) collagen, making it a promising treatment for fibrotic diseases. However, curcumin has poor intestinal absorption and low bioavailability, which limits its efficacy in treating oral mucosa diseas

Incidence Progression and Epidemiological Study of Oral Precancerous Lesions: An Original Research.

INTRODUCTION: Oral precancerous lesions, such as leukoplakia and erythroplakia, are significant contributors to the global burden of oral cancer. Understanding their epidemiology, incidence, and progression is essential to early detection and prevention. In India, these lesions are prevalent due to widespread tobacco consumption and other lifestyle factors, leading to an increased risk of oral cancer development. METHODS: A prospective observational study was conducted with 150 patients showing clinical signs of oral precancerous lesions. Three parameters were analyzed: patient demographics,

Human leukocyte antigen and its association with oral potentially malignant disorders - A systematic review and meta-analysis.

BACKGROUND: The Human Leukocyte Antigen (HLA) genes, located within the Major Histocompatibility Complex on chromosome 6p21, are highly polymorphic and play a vital role in immunity and has strong associations with autoimmune disorders. However, its role in cancers including oral cancers and oral potentially malignant disorders (OPMD) is still being explored and there is no strong evidence available for this association. Hence the present systematic review was performed to synthesize the current available evidence of genetic propensity of HLA alleles in protective or susceptibility role in occ

Co-Transfection With Anti-Fibrotic microRNAs for Treating Oral Submucous Fibrosis.

BACKGROUND: Oral submucous fibrosis is characterized by excessive collagen deposition and is highly associated with a patient's betel nut chewing habit. Arecoline initiates the transforming growth factor-beta (TGF-β)/Smads signaling pathway and activates downstream fibrotic genes. Dysregulation of microRNA (miR) expression is involved in the OSF progression, and miR modulation is a promising treatment. As one miR can target multiple mRNAs, and one mRNA has multiple binding sites to different miRs, we thus propose that simultaneous co-transfection of anti-fibrotic miRs may have a better therape

miR-134-3p Deficiency-Induced Methionine Metabolism Promotes Fibrosis via SLC25A33 in OSF.

BACKGROUND: Fibrosis is a disease characterized by excessive collagen deposition by fibroblasts, leading to tissue and organ dysfunction. Fibroblasts are the primary effector cells, and their functional phenotype is regulated by various factors, with metabolic reprogramming being a crucial one. Previous studies have shown that microRNAs induce hypoxia, abnormal energy metabolism, and promote fibrosis. Our research aimed to identify the miRNA associated with oral submucous fibrosis (OSF) pathogenesis and to preliminarily explore its role in the phenotypic transformation of fibroblasts. METHODS
