Gastrointestinal adverse events associated with Hedgehog pathway inhibitors: a disproportionality analysis.

OBJECTIVE: The Hedgehog pathway inhibitors (HPIs) are effectively used to treat basal cell carcinoma (BCC). Despite the benefits, HPIs are reported to cause various gastrointestinal adverse events (GIAEs). The study was conducted to detect possible safety signals of HPIs concerning GIAE through disproportionality analysis. METHODS: A retrospective case/non-case disproportionality analysis was performed in the USFDA Adverse Event Reporting System database using OpenVigil 2.1, a web-based pharmacovigilance tool. All the GIAEs reported due to the HPIs, like vismodegib, sonidegib, and glasdegib,

Pharmacological Blockade of TRPM8 Ion Channels Fails to Reduce Behavioral and Biological Markers of Acute Oral Pain in Irradiated C57BL/6J Mice.

Radiation-associated pain (RAP) after head and neck cancer treatment often results in significant discomfort, yet the mechanisms underlying this pain remain poorly understood. Transient receptor potential cation channel subfamily M (melastatin) member 8 (TRPM8) channels, known to mediate cold sensation, have been implicated in RAP. Previous studies suggested that inhibition of TRPM8 might offer a therapeutic approach for alleviating radiation-induced pain. We tested the effects of PBMC, a small molecule inhibitor of TRPM8, on radiation-induced glossitis and associated pain behaviors in C57BL/6

Vitamins and oral mucosal diseases: From bench to the bedside.

Vitamins are essential biocatalysts that have wide-ranging functions in supporting cellular metabolism. Vitamin deficiencies can significantly impact oral health, manifesting in oral mucosal and periodontal conditions. Deficiencies in the vitamin B complex are associated with glossitis, angular cheilitis, and recurrent aphthous stomatitis. Vitamin C deficiency leads to scurvy-related periodontal issues, whereas vitamin A insufficiency predisposes to oral epithelial keratinization abnormalities such as dry mouth and susceptibility to Candida. A reduction in vitamin D levels is linked to the ris

Histopathological and ultrastructure alterations in tongue and parotid tissues of rats in consequence to long-term alcohol intoxication and ameliorative role of 10-dehydrogingerdione.

BACKGROUND: Alcoholism adversely affects oral health, leading to issues such as dental caries, gingivitis, and oropharyngeal cancers. One notable oral effect is glossitis, which manifested as tongue inflammation. Chronic alcohol consumption can lead to sialadenosis, disrupting salivary gland function. AIM: This study aimed to demonstrate how alcohol intoxication induces histopathological and ultrastructural degenerative alterations in the tongue and parotid tissues of experimental rats and the therapeutic potential of 10-dehydrogingerdione (10-DHGD). METHODS: A total of 40 albino male rats wer

A review of the pathology of herpesvirus infections in pinnipeds.

Twenty-two herpesviruses, all within the family Orthoherpesviridae and belonging to the Alphaherpesvirinae (n = 3) and Gammaherpesvirinae (n = 19) subfamilies have been reported in 14 different pinniped species (n = 8 phocids, n = 5 otariids and n = 1 odobenid). This review critically assessed the purported pathology reported for herpesvirus infections and found a variety of lesions present dependent on both the herpesvirus and host species. Lesions included hepatic and adrenal necrosis, interstitial pneumonia, thyroid atrophy, lymphodepletion, lymphocytolysis, ulcerative stomatitis, glossitis

Sherlock Holmes and the mystery of the deadly diet.

In honor of the new section on Forensic Dermatology that appears in Clinics in Dermatology, we present a modern-day detective story we created based on Sherlock Holmes, the famous fictional character created by Sir Arthur Conan Doyle (1859-1930). In our tale, a victim suspects that he is being slowly poisoned. An alteration of the victim's tongue (atrophic glossitis) and jaundice provide diagnostic forensic dermatologic clues. The serum toxicology is negative, and the police find no evidence of common poisons in the household. Sherlock Holmes, however, suspects foul play and must solve the cri

Herpetic Geometric Glossitis in an Immunocompetent Patient: An Atypical Presentation of Oral Herpes Simplex Virus (HSV) Infection.

Herpetic geometric glossitis (HGG) is a rare, atypical manifestation of herpes simplex virus type 1 (HSV-1) infection, most commonly reported in immunocompromised individuals. We report a case of a 43-year-old immunocompetent male presenting with edematous heme-crusted lips, fissured tongue, and severe oral pain, leading to significant oral intake limitation and subsequent inpatient admission. Dermatology was consulted for possible Stevens-Johnson syndrome, which was ruled out following evaluation.  Clinical findings were consistent with HGG, an uncommon presentation of HSV infection. The pati

Medullary haemorrhagic interstitial nephritis in multisystem inflammatory syndrome in adults following COVID-19.

Multisystem inflammatory syndrome in adults (MIS-A) is a severe postinfectious complication of COVID-19. Renal involvement is common; however, histopathological findings have not been adequately characterised. We here report a case of a previously healthy man in his 60s who developed persistent fever, diarrhoea, glossitis and digital swelling approximately 3 weeks after COVID-19 infection. Antibiotic therapy was ineffective and renal function deteriorated. He was diagnosed with MIS-A and corticosteroid therapy produced marked clinical improvement. He was discharged 3 weeks after hospital admis

Machine learning models in predictive factors for megaloblastic character of macrocytic anemia.

BACKGROUND: Differentiating megaloblastic from non-megaloblastic macrocytic anemia usually relies on vitamin B12 and/or B9 assessment and bone marrow examination. In resource-limited regions, these diagnostic tools are not readily available. This study aims to develop a machine learning model to distinguish between these two forms of macrocytic anemia using commonly accessible clinical and laboratory parameters. METHODS: A logistic regression model was developed using a dataset containing clinical features and routine laboratory data. Key predictive variables were identified through the Seque

How Nutrient Deficiencies Impact the Oral Mucosa and How to Manage Them: A Narrative Review.

BACKGROUND: Malnutrition and micronutrient deficiencies may impact the oral mucosa even before any other symptoms become clinically obvious. This makes the oral cavity an important site for early detection of nutritional imbalance. Across the literature review, the most documented vitamin is vitamin B12. Other widely reported nutrients are iron, folate, and zinc. Less consistently documented are vitamins A, C, and D. Their correlation with recurrent aphthous stomatitis (RAS), atrophic glossitis, angular cheilitis, burning mouth symptoms (BMS), mucosal erythema, and oral candidiasis (OC) is bro
