Multi-omics mapping of TIMP1-associated stromal-myeloid remodeling in autoimmune gastritis and gastric cancer.

BACKGROUND: Autoimmune gastritis (AIG) is a chronic immune-mediated atrophic disease that can induce cancer-related epithelial remodeling, although its association with gastric adenocarcinoma depends on histological and clinical modifiers. Gastric cancer (GC) progression is shaped by the tumor microenvironment (TME), including cancer-associated fibroblasts (CAFs), myeloid cells, and extracellular matrix (ECM) remodeling. Whether AIG-related mucosal remodeling shares stromal programs with GC-associated stromal-myeloid niches remains unclear. METHODS: We integrated one AIG bulk-transcriptomic c

Gastric bacterial microbiota and gastric carcinogenesis: from dysbiosis to immune remodeling and therapeutic opportunities.

Gastric cancer remains a leading cause of cancer-related mortality worldwide. The classical Helicobacter pylori-centric paradigm of gastric carcinogenesis is being progressively expanded, as it is now recognized that the stomach harbors a complex and metabolically active microbial ecosystem whose compositional shifts fundamentally influence oncogenic progression. This review proposes a unifying dysbiosis-carcinogenesis-immune remodeling-therapeutic intervention (DCIT) framework to integrate the rapidly growing body of evidence linking gastric microbiota dysbiosis to malignancy. We survey the c

[Molecular mechanisms of chronic gastric mucosal inflammation induced by Helicobacter pylori via immune evasion].

Helicobacter pylori (H. pylori) persistently colonizes the gastric mucosa and induces sustained inflammation, making it a major driver of the development and progression of chronic gastritis, peptic ulcer disease, and gastric cancer. Its pathogenicity does not rely on direct tissue damage inflicted by a single virulence factor, but rather on the coordinated actions of multiple factors, including cytotoxin-associated gene A (CagA), vacuolating cytotoxin A (VacA), lipopolysaccharide, and urease that disrupt the gastric epithelial barrier and perturb mucosal immune homeostasis. Concurrently, H. p

Magnolol confers neurotrophic effects against MPTP/p-induced Parkinson's disease in mice via anti-inflammatory, anti-apoptotic mechanisms and PI3K/AKT/GSK3β/MAPK/mTOR signalling regulation.

The current hypothesis investigated the neurotrophic effect of magnolol (ML) against MPTP/p-induced neurotoxicity in Parkinson's disease (PD) mice, focusing on the molecular mechanisms of PI3K/Akt/GSK3β and MAPK signalling pathways. To determine the effective dose, 6 mice/group were employed for the dose-dependent study and brain-protective study. Behavioural deficits (open field test, narrow beam walking), dopamine (DA) depletion, lipid peroxidation, antioxidant levels, histology (H&E, PAS, and MT), inflammatory cytokines, DAT and VMAT2 expressions, SN region expressions of BDNF, GDNF, VEGF,

Associations Between Antiparietal Cell Antibody Values and Atrophy in a South and Southeast Asian General Population.

GOALS: To investigate the association between atrophy severity and antiparietal cell antibody (APCA) levels in South and Southeast Asia. BACKGROUND: APCA is an autoantibody that damages gastric parietal cells; autoimmune gastritis (AIG) is a chronic gastric inflammatory disease related to APCA and severe predominant corpus atrophy. Although a positive APCA result is a key clinical diagnostic tool for AIG, its rates vary widely among ethnic groups, and its exact relationship with AIG and predominant corpus atrophy remains unclear. STUDY: Associations between histopathology-assessed and endosc

Data augmentation method for computer-aided diagnosis using specular reflection.

UNLABELLED: Drug-resistant Plasmodium falciparum has been on the rise, mainly due to point mutations in the pfDHFR or dihydrofolate reductase gene. This has heightened urgency to discover and develop new antimalarial drugs. This research utilized Pharmacophore Modeling, Virtual Screening, Molecular Docking and Molecular Dynamics Simulation techniques to identify potential pfDHFR inhibitors from virtual databases. Ligand-based pharmacophore hypothesis (H1) (r = 0.949, r2 = 0.883) was formulated using a training set of 28 compounds that demonstrated pfDHFR inhibitory activity across four orders

Gastrin-dependent expansion of Cck2r+ corpus progenitors accelerates ulcer healing and inhibits gastric dysplasia.

BACKGROUND: The cholecystokinin-2/gastrin receptor (Cck2r) is expressed in corpus isthmus progenitor, enterochromaffin-like and parietal cells, regulating acid secretion and cell turnover. However, the role of gastrin on Cck2r progenitors during mucosal regeneration remains unexplored. OBJECTIVE: To study the role of gastrin-Cck2r axis and corpus progenitors during gastric injury and regeneration. DESIGN: We generated Cck2r-CreERT2; Gastrin-DTR-p2A-TdTomato; Rosa26-ZsGreen mice to trace corpus Cck2r+ progenitors during homeostasis and injury, under conditions of hypogastrinaemia and hypergas

Formononetin Alleviates MNNG-Triggered Chronic Atrophic Gastritis: Its Potential Mechanisms.

INTRODUCTION: Chronic atrophic gastritis (CAG) is the initial phase in the carcinogenesis of gastric cancer (GC). Therefore, effective treatment for CAG is important in reducing the risk of GC progression. As an isoflavone compound, formononetin (FMN) has been identified as a potential therapeutic agent for acute gastric ulcers and GC. However, no study has reported the protective effect of FMN against CAG and its underlying mechanism. This study aimed to explore the therapeutic effects of FMN on CAG and its underlying mechanisms in vitro. METHODS: Network pharmacology was applied to predict

Autoimmune gastritis potentiates polypoid nodule scar formation after endoscopic resection: implications for risk-stratified surveillance.

BACKGROUND: Endoscopic resection has become a cornerstone treatment for gastric mucosal lesions, yet polypoid nodule scar (PNS) formation emerges as a significant post-procedural sequela. Given the chronic inflammatory environment and hypergastrinemia characteristic of autoimmune gastritis (AIG), this study investigates the association between AIG and PNS development following endoscopic resection of the gastric antrum, aiming to optimize postoperative surveillance strategies for high-risk populations prone to PNS. METHODS: This retrospective cohort study analyzed 109 patients undergoing R0 r

Russell Body Gastritis: Histochemical and Immunohistochemical Findings in a Series of Three Patients.

Russell body gastritis is a discrete morphologic pattern of chronic gastritis, characterized by numerous plasma cells filled with intracytoplasmic Russell body inclusions. It can be mistaken for neoplastic lesions and for extracellular hyaline structures. We report three adult men with Russell body gastritis. The patients were diagnosed and treated for various hematolymphoid disorders. During follow-up, they presented with abdominal pain, gastric erythema and thickening. One patient had Helicobacter pylori infection. The gastric biopsy showed an expanded lamina propria filled with numerous pla

A vicious cycle inducer in gastric pathogenesis: implication of Helicobacter pylori.

Helicobacter pylori is a major human pathogen responsible for chronic gastritis, peptic ulcers, and gastric cancer. Its persistence is facilitated by a complex arsenal of virulence factors, including the CagA oncoprotein, the VacA toxin, adhesins, and the ability to form biofilms. While the roles of individual factors are well-studied, the integrated mechanisms by which they collectively drive carcinogenesis remain a critical knowledge gap. This review integrates evidence showing that CagA delivery and type IV secretion-dependent signals create a transient, infection-driven BRCAness (BRCA1/2 p

Platelets/lymphocytes ratio and neutrophils/lymphocytes ratio in children with H. pylori associated gastritis: a canary in the mine?

BACKGROUND: Low-grade systemic inflammation and localized stomach inflammation are both brought on by Helicobacter pylori (H. pylori), which is a prevalent cause of chronic gastritis. The aim of this study was to assess the association between H. pylori positivity, severity, and both platelets/lymphocytes ratio (PLR) and neutrophils/lymphocytes (NLR) ratios in children. METHODS: One hundred consecutive children who underwent gastroscopy for upper gastrointestinal tract symptoms were analysed retrospectively. They were classified according to identification of the H. pylori antigen in stool be

Comparative Efficacy and Safety of Nitazoxanide-based Triple Therapy Versus Standard Triple Therapy in Treating Helicobacter Pylori Infections: An Updated Systematic Review and Meta-analysis of Randomized Controlled Trials.

Helicobacter pylori infections are the major cause of gastrointestinal disease, mainly chronic gastritis, peptic ulcer, and gastric carcinomas, affecting half of the population globally. Due to the emergence of antibiotic resistance, the efficacy of current standard therapies, particularly clarithromycin and metronidazole, has been reduced. Nitazoxanide (NTZ), a broad-spectrum antimicrobial drug, has shown promising efficacy against H. pylori infections. This study aims to assess the comparative efficacy and safety of NTZ-based regimens versus standard triple therapy in H. pylori infections. A

Integrative Unani pharmacotherapy in chronic gastroesophageal reflux disease: A case report.

BACKGROUND: Gastroesophageal reflux disease (GERD) is a chronic gastrointestinal disorder that often requires prolonged proton pump inhibitor (PPI) therapy. It frequently coexists with Helicobacter pylori-associated gastritis. Limitations of long-term PPI use, symptom recurrence, and patient adherence have increased interest in complementary medical systems. Unani system of medicine (USM) conceptualizes reflux-like disorders as disturbances of digestion and gastric temperament, emphasizing mucosal protection and physiological restoration. Molecular Insights into Helicobacter pylori-Induced Gastritis and Gastric Cancer.

Helicobacter pylori (H. pylori) is recognized as one of the most widespread and persistent bacterial infections globally, with a remarkable ability to colonize the human stomach. This pathogen is a major contributor to the development of gastric diseases, including gastric lymphoma and adenocarcinoma. The H. pylori infection triggers a complex pathogenic cascade within the gastric environment, characterized by prolonged inflammation and heightened oxidative stress, which fosters a milieu of immune dysregulation, where both innate and adaptive immune cells become activated inappropriately, ther
