Pharmacokinetic investigation on eight primary active constituents of Weifuchun capsules in healthy and chronic atrophic gastritis model rats.

Weifuchun capsule (WC) is clinically employed in treating chronic atrophic gastritis (CAG), yet differences in its pharmacokinetic profile between normal and chronic atrophic gastritis model rats remain insufficiently characterized. In this study, a reliable and precise ultra-high performance liquid chromatography-tandem mass spectrometry method was developed and validated for the simultaneous quantification of eight major active constituents including p‑coumaric acid, ginsenoside Rg1, kaempferol, luteolin, hesperetin, apigenin, naringenin, and tangeretin in rat plasma. This method was subsequ

Trends in the burden and quality of care for upper gastrointestinal diseases from 1990 to 2021: An analysis of the global burden of disease study 2021.

BACKGROUND: Upper gastrointestinal (GI) diseases, primarily gastroesophageal reflux disease (GERD), peptic ulcer disease (PUD), and gastritis and duodenitis (GD), are highly prevalent, chronic, and recurrent conditions worldwide, imposing a substantial burden on population health and quality of life. However, long-term trends in their quality of care and the disease-specific burden remain poorly characterized. This study aimed to analyze global trends in the disease burden and quality of care for upper-GI diseases, from 1990 to 2021. METHODS: Based on data from the Global Burden of Disease St

Anwei decoction alleviates chronic atrophic gastritis by modulating the gut microbiota-metabolite axis and NLRP3 inflammasome activity.

BACKGROUND: Chronic atrophic gastritis (CAG) is a clinically refractory gastric disease often characterized by high recurrence rates and adverse drug reactions. Anwei decoction (AWD), a traditional Chinese medicine formula, has been shown to significantly improve clinical symptoms in patients with CAG, as demonstrated by a multicenter cohort study (overall effective rate: 82.5%, P < 0.01). However, the unclear molecular mechanisms and therapeutic targets of AWD limit its international acceptance. AIM: To investigate the therapeutic mechanisms of AWD against CAG from an integrated perspective.

Identification of LGR5 and TFF2 as Biomarkers in High-Risk Chronic Atrophic Gastritis: From Multi-Omics Mining to Clinical Validation.

Источник пометил публикацию как отозванную

INTRODUCTION: High-risk chronic atrophic gastritis (CAG; OLGA/OLGIM Ⅲ-Ⅳ) carries significant gastric cancer (GC) risk yet lacks reliable gastric stem cell (GSC)-based biomarkers. We evaluated GSC markers LGR5 (proliferative) and TFF2 (protective) for risk stratification. METHODS: TCGA/GEO bioinformatics analysis preceded immunohistochemical validation in 60 clinical samples. Protein co-expression (Wnt/β-catenin, Ki67, Bax) was assessed. Diagnostic/prognostic power was tested via ROC and Kaplan-Meier analyses. Functional networks were deciphered through GO/KEGG enrichment. RESULTS: High-risk

The oral-gut axis in chronic atrophic gastritis: current perspectives and integrated strategies.

Chronic atrophic gastritis (CAG) is a key precursor to gastric cancer, characterized by progressive mucosal atrophy, inflammation, and microbial dysbiosis. The Correa cascade model highlights Helicobacter pylori as a primary driver, progressing from gastritis to atrophy, intestinal metaplasia (IM), dysplasia, and malignancy. However, 20%-30% of CAG cases lack H. pylori involvement, emphasizing the roles of non-H. pylori microbial dysbiosis, environmental factors, and the oral-gut axis in disease progression. Oral microbes, such as Porphyromonas gingivalis, translocate to the stomach, amplifyin

[Haploinsufficiency of A20 in a family caused by a heterozygous deletion in the TNFAIP3 gene].

The proband was a 3-year-4-month-old boy who had experienced recurrent diarrhea, fever, joint swelling and pain for more than a year accompanied by oral and perianal ulcers. Gastrointestinal endoscopy revealed multiple colorectal ulcers. The proband's 36-year-old father had a three-year history of oral ulcers and a one-year history of recurrent diarrhea; gastroscopy showed chronic non-atrophic gastritis with erosion and duodenitis, while colonoscopy was unremarkable. Whole exome sequencing revealed that the proband and his parents had no pathogenic single nucleotide variants or small insertion

[Eosinophilic Gastrointestinal Diseases: Diving beyond the Tip of the Iceberg].

Eosinophilic Gastrointestinal diseases (EGID) are group of chronic inflammatory diseases of the gastrointestinal tract, characterized by infiltration of eosinophils in its layers. These diseases can affect any organ in the GI tract, and can be classified to subgroups according to the anatomical organ involved: Esophagus - Esoniphilic Esophagitis (EoE), Stomach - Eosinophilic Gastritis (EoG), Small Intestine - Eosinophilic Enteritis (EoN) and Colon - Eosinophilic Colitis (EoC). The pathogenesis is not fully understood, but it is likely to involve a combined interaction between hereditary and en

Depressive symptoms and their gastrointestinal correlates: A nationwide observational study in the Chinese middle-aged and elderly population with nonmalignant gastrointestinal disorders.

The pathophysiological interplay between depressive symptoms and benign upper gastrointestinal (UGI) disorders encompassing peptic ulcer disease, gastritis, duodenitis, and gastroesophageal reflux disease remains incompletely defined. These chronic inflammatory conditions impose a substantial health burden on Chinese middle-aged and elderly adults, yet the temporal relationship linking depressive symptomatology to disease onset remains underexplored. This nationally representative cohort study seeks to address this critical knowledge gap through longitudinal analysis. This national population-

Integrated network pharmacology, metabolomics, and transcriptomics of Lianpuyin Jiawei decoction in Helicobacter pylori induced chronic gastritis.

ETHNOPHARMACOLOGICAL RELEVANCE: Helicobacter pylori (H. pylori) infection represents the most significant pathogenetic factor for chronic gastritis (CG) and is also the most critical controllable risk factor for the prevention of gastric cancer. Lianpuyin Jiawei Decoction (LPYJWD), a traditional formula originating from Traditional Chinese medicine, has been used to treat H. pylori-induced gastric disorders. Studies have shown that LPYJWD effectively reduce H. pylori-induced gastric mucosal inflammation and improve gastric mucosal damage. However, the potential pharmacological mechanism of LPY

Histopathologic Features of Chimeric Antigen Receptor (CAR) T-cell Therapy-associated Gastrointestinal Mucosal Injury: A Case Series and Review of Literature.

Chimeric antigen receptor (CAR) T-cell therapy is an effective treatment for refractory hematologic malignancies; however, gastrointestinal adverse events (GI-AEs) are increasingly recognized as its use expands. We identified 6 patients with GI-AEs following CAR-T therapy over a 5-year period at our institution and conducted a comprehensive PubMed literature review to characterize histopathologic patterns and highlight differential diagnostic considerations. In the institutional cohort, the colon was most frequently involved (83%), followed by the small bowel (67%) and stomach (33%). Most pati

Autoimmune Gastritis and Gastric Cancer Risk: Endoscopic and Histopathological Outcomes.

Background and Aims: Autoimmune gastritis (AIG) is a chronic immune-mediated condition characterized by corpus-predominant atrophy, which can lead to vitamin B12 deficiency, achlorhydria, and an increased risk of gastric adenocarcinoma (GC) and neuroendocrine tumours. Diagnosis is often challenging due to a long asymptomatic phase and variable clinical presentation. This study aimed to assess the prevalence of gastric cancer and advanced premalignant lesions and to identify risk factors associated with a worse endoscopic outcome. Methods: This retrospective observational study involving AIG pa

[PDCD4 as tumor suppressor protein in gastric mucosa in early, advanced precancerous lesions and gastric adenocarcinoma].

UNLABELLED: The PDCD4 (programmed cell death protein 4) protein is a key target protein, in particular for the well-studied miR-21. By suppressing its synthesis, miR-21 exerts its influence on key cellular biological processes. Furthermore, miR-21 itself is one of the most relevant predictor markers for assessing the risk of developing gastric cancer. OBJECTIVE: Evaluation of PDCD4 expression as a potential marker for gastric cancer prediction and in the differential diagnosis of gastric mucosal dysplasia. MATERIAL AND METHODS: Comparative immunohistochemical analysis of PDCD4 protein was pe

Within-host diversity and phased variant analysis reveal structures and recombination of Helicobacter pylori subpopulations in stomach.

BACKGROUND: Helicobacter pylori (H. pylori) has a highly plastic genome and can generate substantial within-host diversity during chronic gastric colonization. However, the delineation of its within-host subpopulations, particularly regarding the emergence and spread of antibiotic resistance-conferring mutations, remains poorly understood. RESULTS: In this study, we enrolled 25 chronic gastritis patients from southern China, collecting multiple isolates from distinct gastric regions. Among them, 14 patients exhibited heterogeneity in antibiotic susceptibility across isolates (heteroresistant)

Combating antimicrobial-resistant Helicobacter pylori infection using nanotechnology- an advancement in gastrointestinal disease treatment.

BACKGROUND: Helicobacter pylori (H. pylori) is a worldwide pathogen associated with chronic gastritis, peptic ulcers, and a high risk of developing gastric cancers. The lack of conventional treatments due to increasing antimicrobial resistance has turned this into an urgent crisis call for alternative approaches. Nanoparticles can be engineered to encapsulate antimicrobial peptides that will release their therapeutic payloads and thus target H. pylori more specifically while preserving beneficial microbiota. STUDY AIMS: This systematic review scrutinized nanoparticle-based therapeutics for H.

The importance of Helicobacter pylori eradication: a narrative review.

BACKGROUND: Helicobacter pylori (H. pylori) is a gram-negative bacterium infecting over 40% of the global population, with highest prevalence in low- and middle-income regions. Chronic infection leads to persistent gastritis and can result in peptic ulcer disease, dyspepsia and gastric adenocarcinoma. Despite its high pathogenic potential, population-based screening and eradication programmes remain limited, particularly in Europe. METHODS: This narrative review summarisescurrent evidence on the epidemiology, clinical burden and eradication strategies for H. pylori. A literature search of Pub

Individuals and Family-Screen-to-Treat-Based Helicobacter pylori Eradication Strategy for Gastric Cancer Prevention: A Latin American Perspective.

Gastric cancer (GC) is a public health concern due to estimates indicating it ranks among the most prevalent cancers and leading causes of cancer-related deaths globally. In Latin America, a region significantly impacted by high mortality and incidence of GC, chronic H. pylori infection remains the primary cause of GC, with an overall prevalence of around 42.5% from 2015 to 2022. Asian countries have developed innovative strategies for primary and secondary prevention. Among the most notable primary and secondary screening programs were endoscopy-based screening in Japan and South Korea; a nat
