[Sociodemographic factors associated with yellow fever mortality in Colombia, 2024-2026Fatores sociodemográficos associados à letalidade por febre amarela na Colômbia, 2024-2026].

OBJECTIVE: To analyze the sociodemographic factors associated with case fatality rates for yellow fever in Colombia between January 2024 and January 2026. METHODS: A retrospective observational analytical cohort study was conducted based on records from the national public health surveillance system (Sivigila). Probable and confirmed cases of yellow fever reported between 1 January 2024 and 31 January 2026 were included. Quantitative variables were described using the median and interquartile range, and categorical variables using absolute and relative frequencies. Cumulative survival was est

Unsupervised identification of low-frequency antigen-specific TCRs using distance-based anomaly scoring.

Identifying antigen-specific T cell receptors (TCRs) within the diverse human repertoire remains challenging, particularly for low-frequency clonotypes. Here, we present TCR-RADAR (TCR Rare Antigen-specific Detection by Anomaly Ranking), an unsupervised approach that detects low-frequency antigen-specific TCRs through distance-based anomaly detection in TCR sequence space. Using TCRdist3 to quantify sequence distances, we identify query TCRs that are anomalous relative to reference repertoires within their V-J gene combinations. We validated this approach across three immunological contexts: C

The Mystery of Dürer's Fever.

Albrecht Dürer's illness has often been interpreted through a splenic and malarial framework, largely because of the famous self-portrait in which he points to a painful area on his left side and refers to a "yellow spot." Yet this reading may be too organ-centered. The image may not simply localize pain; it may depict a true cutaneous lesion. Once the malaria hypothesis is weakened, the conjunction of recurrent fever, constitutional decline, and a painful marked lesion becomes newly legible through the modern spectrum of late-onset autoinflammatory disease, especially VEXAS syndrome and relat

Introgression Shapes Genetic Variation Associated With Humid Environments in Aedes aegypti Mosquitoes From Saudi Arabia.

Introgression between genetically distinct populations generates variation that may promote adaptation to diverse environmental conditions. In the mosquito Aedes aegypti, the forest-associated subspecies Aedes aegypti formosus (Aaf) underwent a domestication process approximately 5000 years ago, giving rise to the globally invasive, human-associated arbovirus vector Aedes aegypti aegypti (Aaa). Despite extensive human-mediated transport, the two subspecies remain largely distinct, with admixture documented mainly in coastal Africa and Saudi Arabia. We examined Saudi Arabian Ae. aegypti across

Enhancement of oncolytic flavi- and alphavirus replication in resistant human pancreatic adenocarcinoma cell lines using ruxolitinib.

INTRODUCTION: Overcoming the immunosuppressive microenvironment of pancreatic cancer (PC) using oncolytic viruses appears to be a promising therapeutic approach. However, tumor cells are characterized by significant heterogeneity, including populations of resistant cells with high basal JAK/STAT signaling activity. One potential approach to overcoming this resistance is the use of JAK1/2 kinase inhibitors, such as ruxolitinib, the efficacy of which has already been demonstrated in several studies. OBJECTIVE: To study the effect of ruxolitinib (a JAK1/2 kinase inhibitor) on the replicative act

Discovery and Biological Characterization of 1-[4-(2-Difluoromethoxyphenyl)-5-methylthiazol-2-yl]-3-(naphthalen-1-yl)urea (DFMNTU): A Novel Thiazolyl-Urea Chemotype Targeting Yellow Fever Virus and Host Casein Kinase 2 (CK2).

Yellow fever virus (YFV) is a re- emerging mosquito- borne flavivirus that causes severe hemorrhagic fever with high case- fatality rates. Due to vaccine contraindications, incomplete immunization coverage, and the absence of approved specific antivirals, host- directed drug discovery targeting cellular machinery exploited by YFV is a critical therapeutic priority. Here, 1-[4-(2- difluoromethoxyphenyl)- 5- methylthiazol- 2- yl]- 3-(naphthalen- 1- yl)urea (DFMNTU) was synthesized, characterized by NMR, MS, and HPLC, and evaluated for antiviral efficacy against YFV strain 17 D in Vero cells. Sexually Dimorphic Visual Processing in the Yellow-Fever Mosquito, Aedes aegypti.

Flying insects process motion across the visual scene to regulate their ground speed, correct course deviations, and respond to behaviorally relevant stimuli such as avoiding predators or finding hosts. The yellow-fever mosquito Aedes aegypti provides a useful model for studying these visually guided behaviors, as both sexes use vision to track moving features. Approaching a moving target presents mosquitoes with a demanding visual problem: they must extract target motion from the changing visual scene while simultaneously controlling their own flight and responding to potentially dangerous mo

Insecticide resistance in Aedes aegypti across sub-Saharan Africa: An overview and perspectives for effective arbovirus control.

Arboviruses transmitted by Aedes aegypti and Aedes albopictus, including Zika, yellow fever and especially dengue and chikungunya, represent a growing public health concern in Africa. Insecticide-based strategies remain essential for the prevention and control of arboviral outbreaks. However, insecticide resistance in Ae. aegypti is globally widespread and appears to be rising in sub-Saharan Africa. Here, we reviewed the distribution and phenotypic and molecular evidence of insecticide resistance in Ae. aegypti across sub-Saharan Africa and summarized the underlying resistance mechanisms and k

Confidence Intervals for a Proportion Estimated From Pooled Samples Based on Firth's Corrected Score.

Interval estimation of a proportion estimated from testing of pooled samples has been studied by a variety of researchers, who have examined both exact and asymptotic methods. Recent work in point estimation has seen the use of Firth's correction to maximum likelihood estimation to reduce bias effectively. We consider the Firth-corrected score statistic, using either expected or observed information, as the first derivative function of a penalized likelihood and develop score-based confidence intervals wholly within this framework. We evaluate their performance, comparing them by information t

Imported mosquito-borne infectious diseases in travellers and migrants: Global patterns, clinical overlap, and prevention gaps-an umbrella review of 21 systematic reviews.

BACKGROUND: Imported mosquito-borne infectious diseases (MBIDs) pose public health challenges in nonendemic regions, but cross-disease evidence remains fragmented. We conducted an umbrella review of systematic reviews to identify gaps and inform clinical practice and research priorities. METHODS: Five databases (PubMed, Embase, Scopus, Web of Science, and the Cochrane Library) were searched from inception to December 3, 2025, for systematic reviews on nine imported MBIDs, including chikungunya, dengue, Rift Valley fever, yellow fever, Zika, lymphatic filariasis, malaria, Japanese encephalitis

Congenital Arboviruses: A Review.

Congenital arboviruses represent a significant public health concern because of their ability to cross the placental barrier and adversely affect fetal development. In this review, we delineate the epidemiology, pathogenesis, clinical manifestations, and diagnostic challenges associated with key congenital arboviral infections, including Zika, dengue, chikungunya, yellow fever, Oropouche, and West Nile virus. While vertical transmission mechanisms vary among arboviruses, inflammation, placental disruption, and viral replication within fetal tissues are common pathological features. Early diagn

Evolutionary Dynamics-Aware De Novo Design of a Thermostable NS1-Derived Diagnostic Protein for Yellow Fever Virus.

Serological diagnosis of neglected arboviruses remains hindered by extensive intra-genus cross-reactivity, compromising both assay specificity and epidemiological accuracy. In this study, we focused on yellow fever virus (YFV) as a model system to design and validate a generalizable de novo protein design framework integrating evolutionary genomics with structural modeling to rationally engineer diagnostic antigens with reduced cross-reactivity and improved cost-effectiveness. Through comprehensive evolutionary mapping over time of the YFV polyprotein, we identified conserved yet discriminator

Cardiovascular outcomes with tirzepatide versus semaglutide in type 2 diabetes.

Preterm birth is associated with the majority of all death and chronic disability related to pregnancy, birth and the neonatal period. The costs to families and to the health care system are enormous. Current approaches to prevent or arrest preterm labour have been unsuccessful. This failure is largely based on our poor understanding of the regulation of the timing and maintenance of parturition. Oxytocin (OT) is the most potent known uterine stimulant. It is produced in the hypothalamus and secreted into the maternal bloodstream. However, OT also is produced within the uterine decidua in late

Development and implementation of "Mazboot Bunyad": a digital health intervention for the screening and management of undernutrition among children aged 6-59 months, at private provider clinics in urban slums of Karachi, Pakistan.

BACKGROUND: Although factor Xa inhibitors have become a popular choice for chronic oral anticoagulation, effective drug reversal remains difficult due to a lack of specific antidote. Currently, 4-factor prothrombin complex concentrate (4F-PCC) is considered the treatment of choice for factor Xa inhibitor-related major bleeding. However, safety of 4F-PCC and its risk of thrombosis when used for this off-label purpose remain unclear. The purpose of this retrospective study is to determine the rate of thromboembolism when 4F-PCC is used for the emergent reversal of factor Xa inhibitors. METHODS:

Sarcopenia and muscle dysfunction in young adults with type 1 diabetes mellitus: associations with bone mineral density.

OBJECTIVES: Complex congenital heart defects that present earlier in life are sometimes channelled in the single ventricle pathway, because of anatomical or logistic challenges involved in biventricular correction. Given the long-term functional and survival advantage, and with the surgeons' improved understanding of the cardiac anatomy, we have consciously explored the feasibility of a biventricular repair in these patients when they presented later for Fontan completion. We present a single institution's 10-year experience in achieving biventricular septation of prior univentricular repairs,

Active learning for the accelerated discovery of complex concentrated NiCoCr alloys in additive manufacturing.

BACKGROUND: According to the World Health Organization, air pollution is closely associated with climate change and, in particular, with global warming. In addition to melting of ice and snow, rising sea level, and flooding of coastal areas, global warming is leading to a tropicalization of temperate marine ecosystems. Moreover, the effects of air pollution on airway and lung diseases are well documented as reported by the World Allergy Organization. METHODS: Scientific literature was searched for studies investigating the effect of the interaction between air pollution and climate change on

Which Stranger's Disease? Immigration, Immunization, and the Whitening of Cuba in the Age of Atlantic Revolutions.

In 1804, Cuban physician Tomás Romay tried and failed to create the first yellow fever vaccine. The article analyzes his experimental efforts, foregrounding the enslaved and enlisted subjects at the center of this early vaccine trial. Though a scientific failure, this brief experiment, the desires and logics embedded within it, and the measures deployed in its wake - in the form of European whitening campaigns - allow us to consider the political uses of immunity during the Age of Atlantic Revolutions. Historicizing these events within the wider geopolitics of the Caribbean, the article explic

A rare case of colonic ulcer.

A 72-year-old woman presented to our outpatient department due to diarrhea that began 10 days ago without a known trigger. The patient presented with frequent (four to five times a day), loose yellow stool, containing significant mucus. She reported no obvious symptoms such as abdominal distension, pain, chills or fever, and her medical history included diabetes and hypertension. Physical examination of the abdominal was unremarkable. To determine the cause of diarrhea, we ordered a colonoscopy for her. To determine the diagnosis, we performed colonoscopy which detected local mucosal edema and
