Strengthening Arboviral Epidemic Response Through Entomological Surveillance: Insights from Bobo-Dioulasso, Burkina Faso.

Arboviral diseases are emerging public health challenges in Burkina Faso, largely driven by the proliferation of Aedes aegypti mosquitoes in the environment. Effective surveillance of arbovirus circulation is critical to inform interventions. From August 2022 to June 2023, we implemented a comprehensive entomological surveillance platform in five sectors of Bobo-Dioulasso. Surveillance methods included oviposition traps to collect eggs, larval surveys in some concessions per sector conducted bimonthly, and adult mosquito collections using BG-Sentinel traps and Prokopack aspirators. Mosquito sa

Review of mosquitoes in Ireland and of relevant mosquito-borne pathogens.

Throughout history, human societies have been greatly affected by mosquitoes and mosquito-borne pathogens such as malaria and yellow fever. On the other hand there have also been reciprocal effects as mosquito populations have been impacted by major societal changes such as agricultural intensification, urbanisation and international trade and travel, causing ever shifting patterns of disease exposure and epidemics. In contrast to continental Europe, mosquito-borne pathogens have not been a major concern in Ireland for many decades. However, the emergence of new invasive mosquito species and t

The impact of maternal flavivirus infections on fetal neurological outcomes: a scoping review.

This scoping review examines the neurological effects of maternal flavivirus infection, specifically Zika virus (ZIKV), Dengue virus (DENV), and Yellow Fever virus (YFV), on fetal brain development. Inclusion criteria focused on studies involving maternal infection and neurodevelopmental outcomes in offspring up to five years of age. The review found that ZIKV, especially the African Strain, exhibited neuroviolence and pronounced fetal brain abnormalities. Despite the presence of transplacental maternal transfer, these antibodies did not fully prevent congenital malformations, including microc

The L1014F Kdr mutation is associated with a higher prevalence and load of the Plasmodium-blocking symbiont Microsporidia MB In Anopheles Gambiae s.l. In Benin.

INTRODUCTION: The Plasmodium transmission-blocking endosymbiont Microsporidia MB was previously identified in Anopheles gambiae s.l., but its association with the carriage of the genotypes of the L1014F kdr mutation, as well as the ecological factors driving its geographical distribution remain understudied. METHODS: Adult mosquitoes were field-collected using human landing catches (HLCs) across 60 villages in the Covè, Ouinhi, and Zangnanado communes of southern Benin. After morphological identification, a sub-sample of An. gambiae s.l. were molecularly speciated, and genotypied for the L1014

Development and validation of a sample-to-result real time multiplex RT-PCR test for human African trypanosomiasis and arboviral febrile illnesses in the Democratic Republic of the Congo.

BACKGROUND: Human African trypanosomiasis (HAT) is a fatal vector-borne disease caused by Trypanosoma brucei. Although HAT incidence has declined, meeting WHO's elimination targets remain difficult, particularly due to limited diagnostic sensitivity for low-parasite load-infections. Arboviruses such as dengue (DENV 1-4), chikungunya (CHIKV), and yellow fever (YFV) virus, present with nonspecific febrile symptoms similar to HAT and are underdiagnosed in Sub-Saharan Africa. Due to this overlap in symptoms and a suspected geographical overlap of vectors and pathogens in the Democratic Republic of

Yellow fever re-emergence in Tolima, Colombia 2024-25: an eco-epidemiological study.

BACKGROUND: Yellow fever virus (YFV) is transmitted by mosquitoes among humans and non-human primates (NHPs) in South America. The 2024-25 yellow fever (YF) outbreak was notable for its spread into new areas, including the department of Tolima in Colombia's Andean region. We investigated the eco-epidemiology of human and NHP YF cases to understand the patterns and drivers of the Tolima outbreak. METHODS: We collected spatiotemporal, sociodemographic, and mortality data on human YF cases in Tolima, as well as the locations of deceased NHPs with YF. We then conducted exploratory, descriptive, a

Global epidemiology and evolutionary dynamics of arboviruses: A systematic review of surveillance, control strategies, and emerging threats.

Arboviral diseases, transmitted primarily by Aedes mosquitoes, represent a growing global health challenge. The spread of dengue, Zika, chikungunya, and yellow fever has been associated with factors such as climate change, urbanization, and increased global mobility. We conducted a systematic review of the literature published between January 2000 and December 2024, screening 487 studies, of which 11 met predefined inclusion criteria and were included in the final synthesis (PROSPERO registration: CRD42021231605). Molecular detection of insecticide resistance markers in Aedes aegypti (Diptera: Culicidae) populations from Haiti.

Aedes aegypti (Linnaeus, 1762) mosquitoes are the primary vectors of several arboviruses of major public health importance, including dengue virus, Zika virus, chikungunya virus, and yellow fever virus. In Haiti, Ae. aegypti mosquitoes are highly abundant and linked to the transmission and spread of these pathogens. As the use of pesticides for control of mosquitoes increases, there is heightened selection pressure for insecticide-resistant (IR) mosquitoes, mitigating the efficacy of pesticides and leading to an increased risk of continual pathogen transmission. To this end, active surveillanc

Pyrethroids and organophosphate resistance in Aedes aegypti (Diptera: Culicidae) and their underlying mechanisms.

BACKGROUND: For decades, insecticides have been central to controlling the yellow fever mosquito, Aedes aegypti (L.), but extensive use has driven resistance development. This study investigates resistance of Ae. aegypti to pyrethroids (permethrin, deltamethrin) and organophosphates (malathion, pirimiphos-methyl) and their underlying mechanisms across Malaysia, Thailand, Indonesia, and the USA. METHODS: Adult female Ae. aegypti (3-5 days old, non-blood-fed) were subjected to World Health Organization (WHO) tube bioassays using 0.4% permethrin, 0.03% deltamethrin, 5% malathion, and 60 mg/m2 pi

Harnessing insect-derived oils for enhanced efficacy of plant-based repellents against disease-transmitting mosquitoes.

Mosquito-borne diseases such as malaria, dengue, chikungunya, Zika, and yellow fever pose a growing global health threat, worsened by resistance to synthetic pesticides. This study evaluated insect oils from Schistocerca gregaria, Ruspolia differens, and Macrotermes spp. as adjuvants in plant-based mosquito repellents. The formulations were tested for complete protection time (CPT) against Anopheles gambiae, Aedes aegypti, and Culex quinquefasciatus, and volatile organic compounds (VOCs) emitted from treated volunteers were analyzed. Incorporating insect oils doubled CPT compared to essential

Antagonism of RNA silencing in the yellow fever mosquito, Aedes aegypti, by the nsP2 protein of the prototype alphavirus.

Alphaviruses establish persistent infections in mosquito vectors despite robust antiviral RNA interference (RNAi) pathways, suggesting that they employ mechanisms to counteract host immunity. We demonstrate that the nsP2 protein of Sindbis virus (SINV), the prototype alphavirus, functions as a viral suppressor of RNA silencing in Aedes aegypti mosquitoes. Using a SINV mutant (2V) that prevents cleavage at the nsP2-nsP3 junction, we show that proper proteolytic processing to release mature nsP2 is essential for efficient viral replication in mosquitoes with intact RNAi pathways. Replication def

A Multicenter Retrospective Study of Cystic and Alveolar Echinococcosis Patients in the United States (1997-2024).

Echinococcoses are neglected zoonotic diseases caused by larval cestodes of the genus Echinococcus. Regions of the United States rarely experience locally acquired infections, and most contemporary cases are presumed to be imported, predominantly from hyperendemic regions such as Central and East Asia, the Middle East, and North Africa. However, significant knowledge gaps remain regarding the current disease burden within the US healthcare system. The epidemiology, baseline clinical features, and outcomes are summarized for patients diagnosed with cystic echinococcosis (CE) or alveolar echinoc

New Insights Into an Old Vaccine: Potency and Breadth of Neutralizing Antibodies Elicited by Yellow Fever Vaccine 17D Are Boosted by Heterologous Orthoflavivirus Infection.

BACKGROUND: Yellow fever vaccine 17D has controlled yellow fever for almost 90 years and is considered highly successful. However, with recent outbreaks in South America and Africa, yellow fever continues to represent a significant threat to public and global health. Despite the long-standing success, few studies have characterized the capacity of 17D-immune sera to neutralize wild type (WT) yellow fever viruses (YFVs). METHODS: Using 17D and 13 WT YFVs, we conducted focus neutralization tests against sera from a unique cohort of 36 nonendemic vaccinees with diverse Orthoflavivirus histories.

A Replication-Competent Flavivirus Genome with a Stable GFP Insertion at the NS1-NS2A Junction.

The flavivirus NS1 protein is a component of the viral replication complex and plays diverse, yet poorly understood, roles in the viral life cycle. To enable real-time visualization of the developing replication organelle and biochemical analysis of tagged NS1 and its interacting partners, we engineered a replication-competent yellow fever virus (YFV) replicon encoding a C-terminal fusion of NS1 with green fluorescent protein (NS1-GFP). The initial variant was non-viable in the absence of trans-complementation with wild-type NS1; however, viability was partially restored through the introducti

Human genetics of susceptibility to live-attenuated viral vaccines.

Alongside the development of antibiotics, vaccination is the medical innovation with arguably the greatest impact on human health. Testament to its success is the eradication of infectious diseases, such as smallpox, that devastated human populations for almost 4000 years. Live-attenuated vaccines (LAV), which retain the capacity for infection and replication, were the first to be developed and remain highly efficacious, underpinning successful human vaccination campaigns (e.g. polio virus, measles virus, yellow fever virus). The cost of this success is the capacity of LAVs to cause disease in
