Genomic characterization of highly pathogenic avian influenza A(H5N1) clade 2.3.4.4b into wild seabirds from coastal Ecuador.

INTRODUCTION: Highly pathogenic avian influenza A(H5N1) clade 2.3.4.4b has spread widely across South America, but its genomic diversity and patterns of introduction into Ecuador remain poorly characterized. METHODS: In this study, we report and characterize four genomes of A(H5N1) viruses detected in four individuals of two Ecuadorian seabird species: two magnificent frigatebirds (Fregata magnificens) and two blue-footed boobies (Sula nebouxii) sampled in 2023. RESULTS: These genomes represent confirmed detections of avian influenza in wild birds in the country. The viruses were assigned to

Cross-binding antibodies capable of neutralising diverse hantaviruses are produced in response to Puumala virus infection.

BACKGROUND: Orthohantaviruses (hantaviruses) are emerging rodent-borne pathogens that can cause severe human disease. They are present on multiple continents and are responsible for thousands of human cases per year. Despite this, no licenced therapeutics are available, vaccines for most strains are lacking, and the immunological response to infection is poorly characterised. This study aimed to analyse the humoral immune response to Puumala virus (PUUV) infection to inform future studies focussing on the production of therapeutic monoclonal antibodies and vaccination strategies. METHODS: Ser

Spiroplasma infection complicated by macrophage activation syndrome and fulminant hepatitis in a kidney transplant recipient.

A 65-year-old kidney transplant recipient was admitted with isolated fever. Initial tests revealed pancytopenia and elevated C-reactive protein levels but failed to detect any pathogen. A bone marrow aspirate was performed because of signs suggestive of hemophagocytic lymphohistiocytosis, but the results were negative. The patient subsequently developed fulminant hepatitis. Liver biopsy showed severe acute cytolytic hepatitis with a neutrophil-rich infiltrate, suppurative hepatocytic necrosis, and hemophagocytosis. Etoposide, N-acetylcysteine, and piperacillin-tazobactam were initiated. Howeve

Chapare Virus: A Re-Emerging Zoonotic Arenavirus With Limited Preparedness.

Chapare virus (CHAPV) is an emerging New World arenavirus that causes Chapare haemorrhagic fever (CHHF), a severe and often fatal illness. First identified in Bolivia in 2003, CHAPV remains geographically limited but has shown potential for nosocomial transmission. It causes symptoms similar to other viral haemorrhagic fevers, including fever, muscle pain, bleeding, and neurological complications. The virus is primarily spread through contact with infected rodents, although limited human-to-human transmission has been documented. CHAPV has a bisegmented RNA genome with ambisense polarity. It s

A STAT1-Knockout Mouse Model for Chapare Virus Infection and Pathogenesis.

Chapare virus (CHAPV) is an Arenaviridae family member and causative agent of Chapare hemorrhagic fever (CHHF). Endemic to Bolivia, CHAPV was found to be the cause of several outbreaks of CHHF in Bolivia in 2003 and 2019 with high case-fatality rates and instances of human-to-human transmission. The pathogenesis of CHAPV infection is poorly understood, and no vaccines or antivirals are available, in part due to a dearth of available animal models. Mice lacking signal transducer and activator of transcription 1 (STAT1-/-) have been shown to succumb to infection by related arenaviruses, includin

A Transgenic Syrian Hamster Model for New World Hemorrhagic Fever Mammarenavirus Infection.

Mammarenaviruses are endemic in rodent populations worldwide, and their zoonotic transmission can lead to a severe, life-threatening hemorrhagic fever (HF) syndrome. In the Americas, five New World mammarenaviruses (NWMs) are known to cause HF. In the absence of FDA-licensed vaccines or antiviral therapies, these viruses pose a significant public health risk and a threat to national security. Research into NWM pathogenesis and the development of effective countermeasures is severely limited by the lack of small-animal models that faithfully reproduce human disease. The pathogenic NWMs use huma

Neural network modeling of Lassa fever spread and disability effects.

Lassa fever is one of the viral hemorrhagic illnesses produced through the Lassa virus and may be transmitted to individuals via contact with diseased rodents or their feces and urine. The current study presents the solutions to the nonlinear zoonotic spread of Lassa virus fever leading to disabilities model by designing a supervised neural network. The model is divided into the human (susceptible, severely infected, mildly infected, and recovered) and rodent (susceptible, infected) populations. The dynamic structure of one class to the other performs the zoonotic spread of Lassa virus fever m

SIRPA suppresses integrin-dependent virus endocytosis.

New World arenaviruses (NWAs) that cause viral hemorrhagic fever, such as Junín virus, have few therapeutic options. Entry of these viruses into cells is mediated by binding to cell surface receptors, followed by endocytosis and trafficking to a low pH compartment. We showed previously that Signal Regulatory Protein Alpha (SIRPA), a critical cell surface receptor that inhibits macrophage phagocytic activity, decreases internalization by NWAs as well as other pathogenic RNA viruses that traffic to low pH compartments. Here we demonstrate that proteins involved in the SIRPA/integrin signaling ax

Comparison of Vaccine Platforms for Machupo Virus.

BACKGROUND/OBJECTIVES: Pathogenic mammarenaviruses cause severe hemorrhagic and neurologic disease in humans. Machupo virus (MACV), a New World (NW) mammarenavirus, causes Bolivian hemorrhagic fever in humans, and there are no approved vaccines. METHODS: Here, we describe and compare the immunogenicity of three vaccines expressing the MACV glycoprotein complex (GPC) in C57BL/6 mice: a recombinant vesicular stomatitis virus (rVSV) and two different lipid nanoparticle (LNP)-encapsulated nucleoside-modified mRNA (mRNA-LNP) vaccines. The first mRNA-LNP vaccine, designated MACV mRNA, expresses the

Efficacy of an Fc-silent antibody targeting the apical domain of human transferrin receptor 1 in transgenic mouse models of Junín virus infection.

Several New World mammarenaviruses (NWMs), including Junín virus (JUNV), can cause a severe, potentially fatal hemorrhagic fever (HF) syndrome. In the absence of FDA-approved vaccines or antivirals, there is an urgent need to develop broadly active countermeasures against these viral infections. Cellular entry by all pathogenic NWMs is mediated by transferrin receptor 1 (TfR1). Here, we report on the pharmacokinetics (PK) and antiviral activity of a mouse/human chimeric antibody, targeting the apical domain of human TfR1 and lacking effector functions (ch128.1/IgG1 Fc-silent) in two transgenic

Serological evidence of Lassa virus in commensal rodents from Senegal.

BACKGROUND: Lassa fever, a neglected zoonotic hemorrhagic disease caused by Lassa virus (LASV) and endemic in West Africa, remains a significant public health concern associated with rodent exposure. Senegal lies at the western fringe of the LASV area, but only one 1988 serosurvey reported low antibody prevalence. Given recent ecological shifts, including the expansion of invasive Rattus rattus and Mus musculus, we reassessed LASV exposure in rodents from Senegal. METHODS: We retrospectively analyzed 618 archived rodent sera collected in 2012-2013 from domestic and peri-domestic environments

In silico prioritization and cheminformatics identify structurally diverse small-molecule inhibitors of Lassa virus glycoprotein-mediated membrane fusion.

Lassa virus (LASV) is a hemorrhagic fever arenavirus of significant public health concern, infecting millions of people per year in Africa. Here, we developed a computational strategy to identify specific inhibitors of LASV glycoprotein-mediated virus cell entry, leveraging a previous screen of 297,156 small molecules from the MLPCN library, with the results deposited in the PubChem database. Data mining methods were developed to efficiently select small molecules prioritized for both potency and specificity in inhibiting Lassa virus infection. Cheminformatics classification then identified di

Understanding machupo virus: A neglected arenavirus with global health importance.

Machupo virus (MACV), classified within the Mammarenavirus genus of the Arenaviridae family, serves as the causative agent of Bolivian hemorrhagic fever (BHF), a life-threatening zoonosis primarily confined to rural regions of Bolivia's Beni Department. First identified in 1959 during epidemics in San Joaquín, this bisegmented, ambisense, single-stranded RNA virus exhibits 20%-35% case-fatality rates, with highest lethality among children under 5 and adults over 55. The vesper mouse (Calomys callosus) acts as the principal reservoir, sustaining lifelong asymptomatic infections and shedding vir

Immunogenicity of recombinant vesicular stomatitis virus-vectored vaccines expressing the Machupo virus glycoprotein in different genome positions.

Recombinant vesicular stomatitis virus (rVSV) is a powerful vaccine platform that elicits durable cellular and humoral immune responses. rVSV vaccines have successfully been used for various hemorrhagic fever-causing viruses. Here, we create and evaluate rVSV vaccines for Machupo virus (MACV), a highly pathogenic New World (NW) mammarenavirus that causes Bolivian hemorrhagic fever. VSV genome rearrangements impact attenuation of the viral vector and the immune response to the vaccine. Therefore, in this study, we directly compare the effect of altering the MACV glycoprotein complex (GPC) posit

Lassa fever and Argentine hemorrhagic fever treatment in guinea pigs using broad-spectrum cap-dependent endonuclease inhibitors.

The class Bunyaviricetes encompasses several highly pathogenic viruses that cause lethal hemorrhagic fevers. Due to their limited prevention and treatment options and high pathogenicity, these viruses require handling in biosafety level-4 facilities. Within the bunyaviruses, arenaviruses are particularly notable for their pathogenicity and ability to cause severe hemorrhagic disease in humans. The cap-dependent endonuclease (CEN) is a unique and crucial enzyme involved in the replication cycle of these viruses. As humans do not possess a similar enzyme, CEN represents an ideal target for antiv

Junín virus and Argentine haemorrhagic fever: Virology, pathogenesis, diagnosis, and prevention strategies.

The Junin virus (JUNV) is mainly perpetuated in nature by rodent reservoirs, especially Calomys musculinus, and is transmitted to humans through aerosolization of contaminated rodent excreta. Clinical presentation of Argentine haemorrhagic fever (AHF) is characterized by an incubation period of about 7-14 days and several clinical manifestations such as fever, malaise, hematological abnormalities, hemorrhagic symptoms and neurological complications. The current review summarizes the current knowledge on the taxonomy, structure, genome structure, transmission dynamics, pathogenesis, clinical sy

Monoclonal antibodies raised against the Junín virus glycoprotein show broad reactivity and provide protection in vivo.

UNLABELLED: Argentine hemorrhagic fever (AHF) is a disease caused by the Junín virus (JUNV) that can be fatal in 15%-30% of untreated cases. Although JUNV is the only arenavirus for which efficient therapy and vaccination have been established, its administration is limited. In this study, hybridoma technology was used to generate 30 mouse monoclonal antibodies (mAbs) against the glycoprotein complex (GPC) of JUNV. Of these 30 mAbs, 26 showed strong cross-binding to GPCs from various arenaviruses. In addition, nine mAbs demonstrated potent neutralizing activity in vitro with a 50% inhibitory c

Screening of the Pandemic Response Box Library Identified CRM1/XPO1 as an Anti-Mammarenavirus Druggable Target.

Mammarenaviruses (MaAv) cause persistent infection in their natural rodent hosts across the world and, via zoonotic events, can cause severe disease in humans. Thus, the MaAv Lassa virus (LASV) in Western Africa and the Junin virus (JUNV) in the Argentinean Pampas cause hemorrhagic fever diseases with significant case fatality rates in their endemic regions. In addition, the globally distributed MaAv lymphocytic choriomeningitis virus (LCMV) is an underrecognized human pathogen of clinical significance capable of causing devastating infections in neonates and immunocompromised individuals.
