Lassa virus (LASV), a member of the Arenaviridae family, causes Lassa fever. There are no vaccines available for prevention of Lassa fever, and the primary therapeutic for treatment (ribavirin) has questionable antiviral activity and no formal approval for use. Here, we evaluated ARN-75039, an orally bioavailable broad-spectrum mammarenavirus entry inhibitor, against LASV infection in outbred Hartley guinea pigs exposed to a guinea pig-adapted LASV. ARN-75039 was administered for 14 days either 3 or 7 days postexposure (dpe) to LASV. In the first two studies, once-daily dosing at 3 dpe provide

Identifying genes under local adaptation is an essential step to understand the mechanisms of adaptive evolution. Pathogen-mediated selection is expected to enhance host fitness by favouring resistance to locally prevalent pathogens. However, such pathogen-driven adaptation has been documented in only a few natural systems. Here, we sequenced the Major Histocompatibility Complex Class I region (MHC-I) of 739 Mastomys natalensis captured in Guinea and Nigeria, where the rodent is reservoir to two distinct Lassa virus (LASV) lineages. As predicted, the MHC-I profiles of the two countries, while

BACKGROUND: Lassa fever is endemic in Guinea, with high seroprevalence in the forest region. However, clinical cases have been only anecdotally reported. In August 2022, a nosocomial outbreak occurred at a private clinic in the capital, Conakry, an area previously considered low risk.
METHODS: Suspected cases were confirmed by real-time reverse-transcription polymerase chain reaction within 24 hours. Viremia was monitored during hospitalization, and whole-genome sequencing was performed in-country within 13 days of outbreak detection. Outbreak investigation involved rodent testing in the home

BACKGROUND: Lassa fever is a zoonotic disease, caused by Lassa virus (LASV), that was first reported in 1965. After 55 years, its research landscape remains largely underexplored. Therefore, a bibliometric study is needed to identify LASV research landscape.
METHODS: Scopus database was searched and the resulting data was analyzed using VOSviewer (version 1.6.15) for Windows, bibliometrix (version 3.0.0) with Biblioshiny, and R (version 4.5.2) to reveal trends in LASV publications, including keywords, authors' productivity over time, and collaboration among research institutions and universit

Lassa virus (LASV), the causative agent of Lassa fever, must be handled under biosafety level 4 (BSL-4) conditions, requiring validated inactivation protocols to ensure laboratory and public safety. Although LASV is an enveloped virus theoretically susceptible to physical and chemical inactivation methods, quantitative data on its inactivation kinetics remain limited. This study systematically evaluated the efficacy of thermal treatment (56 °C, 70 °C, 95 °C), laboratory chemical inactivants (beta-propiolactone, formaldehyde, methanol, TRIzol), and five commercial disinfectants against infectio

Lassa fever (LASV; Mammarenavirus lassaense) is an endemic zoonosis in West Africa. Human infections arise from rodent-to-human transmission, primarily from the synanthropic reservoir Mastomys natalensis (M. natalensis). In Sierra Leone, small-mammal communities vary across land-use gradients, shaping LASV transmission risk. How anthropogenic environments facilitate the rodent-rodent interactions remains poorly understood. Small mammals were sampled over 43,266 trap nights in Sierra Leone's LASV-endemic Eastern Province, detecting 684 rodents and shrews. To assess potential transmission, space

Lassa fever is a zoonotic disease endemic to several West African countries, with seasonal outbreaks and potential to cause future epidemics. Vaccines and other medical countermeasures are in development, requiring reliable assays for evaluation. A WHO International Standard (IS) for anti-Lassa virus (LASV) antibodies can support harmonization of serological assay results, aiding vaccine and therapeutic assessment, surrogates of protection identification, and improving epidemiological understanding. In this study, a pooled plasma sample from Lassa fever survivors was evaluated in a multi-centr

BACKGROUND: Lassa fever remains a major public health threat in West Africa, requiring coordinated scientific, policy, and financing responses. Regional scientific convenings are increasingly used to connect research evidence with policy action, yet their contribution to epidemic preparedness is not well documented.
METHODS: We conducted a qualitative health systems and policy analysis of the 2nd ECOWAS Lassa Fever International Conference (ELFIC 2025) in Abidjan, Côte d'Ivoire. Data sources comprised 302 scientific abstracts, plenary and ministerial session records, and the official Minister

The first workshop dedicated to Lassa virus–specific correlates of protection (CoP) was held in 2024 and was convened by the Coalition for Epidemic Preparedness Innovations (CEPI). Experts from multiple disciplines reviewed existing knowledge and identified gaps in understanding Lassa virus- and vaccine-induced immune responses. Discussions covered key areas including epidemiology, immunogenicity, preclinical and clinical research, data science, and regulatory considerations, with the goal of pinpointing opportunities to discover CoP.

The zoonotic Lassa virus (LASV), killing thousands of people annually, is maintained majorly by the multimammate mice Mastomys natalensis and M. erythroleucus; but other rodent reservoirs exist. Knowledge regarding LASV ecology remains limited in certain areas where Lassa fever continues to be incident and even recently emergent. Here, we assess community composition and LASV infection-status of small mammals in key localities across the Guinea savanna belt that spans the countries of Benin and Nigeria. Active LASV infection was determined by conventional (gel based)- and quantitative-PCR scre

Lassa virus (LASV), the causative agent of Lassa fever (LF), poses a significant public health concern in endemic regions due to its high morbidity and mortality. Rapid and accurate diagnosis is critical for effective clinical management and containment during LF outbreaks. However, the genetic diversity of LASV, encompassing at least seven distinct lineages, poses a major challenge for the development of broadly reactive diagnostic tools. Therefore, there is an urgent need to develop detection methods that are effective across diverse lineages. To address this challenge, we generated a panel

OBJECTIVE: Lassa fever is a viral haemorrhagic disease endemic in West Africa. While several vaccine candidates are in development, evidence to guide policy- and decision-making on vaccines remains limited. We convened a Policy Research Working Group (PRWG) to develop a prioritised research agenda to reduce delays related to policy- and decision-making.
METHODS: Using the Child Health and Nutrition Research Initiative (CHNRI) methodology, we conducted a two-phase prioritisation exercise. First, a rapid assessment of literature and expert interviews was conducted to identify evidence gaps, whi

Lassa fever (LF) is a viral hemorrhagic fever endemic to West Africa, with high case fatality in hospitalized patients and limited treatment options, including ribavirin. Preclinical evidence suggests high-dose favipiravir is a promising antiviral treatment alternative. We conducted a randomized controlled open-label phase 2 clinical trial at two reference hospitals in Nigeria to evaluate favipiravir in the treatment of LF. Primary endpoints were the description of classic pharmacokinetic parameters (maximum plasma concentration, time to reach maximum plasma concentration, area under the curve

Large host-virus association databases are increasingly used to explore broad questions in disease ecology, particularly around host range, pathogen diversity, and the potential for spillover. While these databases have been instrumental in large-scale synthesis of host-pathogen biogeography and zoonotic risk, their potential role in addressing fine-scale questions about pathogen prevalence, maintenance, and transmission dynamics remains underexplored. In this study, we build on previous efforts to assess how different types of data, including both entries in databases and the original studies

This paper presents a fractional-order model of Lassa fever transmission by applying the Atangana Baleanu Caputo (ABC) derivative which include the memory and hereditary effects in the disease dynamics. Human and rodent populations in the model, with non-pharmaceutical interventions, such as public enlightenment and quarantine as time-dependent control variables. Qualitative properties positivity, invariant region, equilibrium states, and the basic reproduction number R0 are strictly examined. The disease-free and endemic equilibria are determined as stable with the help of the Lyapunov method

INTRODUCTION: Lassa virus (LASV) causes significant morbidity in West Africa, yet vaccine trial planning is hampered by a lack of high-resolution, community-level seroprevalence data. We aimed to characterize the fine-scale spatial and demographic heterogeneity of LASV exposure in rural Nigeria to inform site selection strategies.
METHODS: We conducted a cross-sectional serosurvey across nine villages in three Nigerian states (Benue, Ebonyi, and Cross River). We recruited 1,874 individuals and tested for LASV IgG using the highly specific Panadea LASV IgG ELISA. We employed Bayesian hierarchi

BACKGROUND: Lassa fever causes an estimated 100,000-300,000 infections annually in West Africa, yet vaccine development and country preparedness efforts have remained fragmented. In response, the Lassa Fever Vaccine Coalition (LFVC) was established in 2024 by the Coalition for Epidemic Preparedness Innovations (CEPI) and the West African Health Organization (WAHO) to align research, policy, and readiness across endemic countries.
METHODS: This practice-based Perspective draws on the authors' experiences as coalition secretariat and founding partners, supplemented by internal program documents

The recurrence of Lassa fever in Nigeria is sustained by human, hospital, rodent, and environmental routes. However, many existing models do not simultaneously capture asymptomatic infection, nosocomial spread, imperfect isolation, and epidemiological memory. This study develops a Caputo fractional-order model that integrates these mechanisms within a single system. The model is shown to admit a unique, non-negative, bounded solution in a positively invariant feasible region. The human-environment reproduction threshold is derived using the next-generation matrix method, while the rodent subsy

Lassa fever is an endemic viral hemorrhagic disease in West Africa with significant morbidity and mortality, yet it remains inadequately addressed in global health priorities. First identified in Nigeria in 1969, the disease continues to affect an estimated 100 000-300 000 individuals annually, with high case fatality rates among hospitalized patients and vulnerable populations. Despite its substantial burden and recurrent seasonal outbreaks, particularly in Nigeria, major gaps persist in both prevention and treatment strategies. Notably, there is no licensed vaccine available more than five d
