Flavonoids from sour jujube leaves: Ultrasound-assisted extraction, UPLC-QQQ-MS/MS quantification, and ameliorative effect on DSS-induced ulcerative colitis in mice.

To valorize sour jujube (Ziziphus acidojujuba) leaves, this work focused on the extraction, quantification, and bioactivity assessment of flavonoids. First, ultrasound-assisted extraction (UAE) was employed to extract total flavonoids from sour jujube leaves (SJL-TF), with the procedure optimized through single-factor design and response surface methodology (RSM). SJL-TF yield reached 48.47 ± 0.36 mg/g under optimal circumstances, which included 61 % ethanol as extraction medium, an ultrasound power of 300 W, an extraction time of 33 min, and a liquid-solid ratio of 16 mL/g, showing higher ext

The Impact of Concomitant Hypothyroid Disease on the Course of Inflammatory Bowel Disease.

BACKGROUND: Inflammatory bowel disease (IBD) is a chronic, immune-mediated inflammatory disorder of the gastrointestinal tract. In IBD, systemic inflammation and immune dysregulation may also impact extraintestinal organs, such as the thyroid gland. Despite this, little is known about the influence of concomitant hypothyroidism on the clinical course of IBD. METHODS: A retrospective analysis was conducted among adult patients with IBD and at least one thyroid stimulating hormone (TSH) measurement within a large healthcare network. Patient charts were reviewed, and baseline demographics, disea

Enteric GABAergic neuron-derived γ-aminobutyric acid initiates expression of Igfbp7 to sustain ILC3 homeostasis.

Neuronal signals have emerged as critical factors that regulate group 3 innate lymphoid cell (ILC3) response and tissue homeostasis, but the molecular mechanisms underlying this regulation remain largely elusive. Here, we identified that the enteric GABAergic neuron-derived neurotransmitter γ-aminobutyric acid (GABA) inhibited proliferation and IL-17A production in ILC3s in a manner dependent on the GABA receptors Gabbr1 and Gabbr2. Conditional deletion of Gabbr1 or ablation of GABAergic neurons caused increased IL-17A production and aggravated colitis. Mechanistically, GABA suppressed the exp

Anti-inflammatory Effect of Microwave-Assisted Opuntia humifusa Extract (MA-OHE) against DSS-Induced Colitis.

Ulcerative colitis (UC), including chronic colitis, is characterized by severe inflammation and oxidative stress in the colon. This study investigated the therapeutic potential of microwave-assisted Opuntia humifusa extract (MA-OHE), prepared using water (WE) and water-methanol (WME) solvents, in a chronic colitis model in mice induced by dextran sodium sulfate (DSS). Chemical profiling using UV-vis, FTIR, and HPLC analyses revealed that WME contains significantly higher concentrations of phenolic and flavonoid compounds than WE. These bioactive compounds enhance the antioxidant capacity of WM

Case report: Alectinib-associated intestinal ulceration and colitis in a patient with non-small cell lung cancer and effective treatment with Mesalazine.

BACKGROUND: Alectinib is effective in extending the survival of patients with anaplastic lymphoma kinase (ALK)-positive non-small cell lung cancer (NSCLC) and generally has manageable side effects. However, intestinal ulcers and colitis are rare but serious adverse reactions linked to Alectinib, meriting further investigation into their causes. CASE PRESENTATION: We report the case of a 62-year-old woman with NSCLC and brain metastases, who tested positive for ALK. She had been treated with Alectinib for nearly 4 years. The patient experienced diarrhea for 4 days, and a subsequent colonoscopy

Neutrophil-derived serine proteases induce FOXA2-mediated autophagy dysfunction and exacerbate colitis-associated carcinogenesis via protease activated receptor 2.

Autophagy plays a critical role in colitis-associated colorectal cancer (CAC). However, non-autonomous regulation of macroautophagic/autophagic flux during inflammation remains largely unexplored. Here, we show that F2rl1/Par2 deficiency (F2rl1[ΔIEC]) aggravated azoxymethane-dextran sulfate sodium-induced CAC based on tumor number and burden, promoted autophagy dysfunction characterized by SQSTM1/p62 accumulation and autophagosome-lysosome fusion inhibition in IECs, and reduced lysosomal acidification by suppressing FOXA2-induced V-ATPase ATP6V0E1 transcription. FOXA2 or ATP6V0E1 overexpressio

Anti-inflammatory and gut microbiota regulatory effects of ultrasonic degraded polysaccharides from Auricularia auricula-judae in DSS-induced colitis mice.

Auricularia auricula-judae is a widely cultivated mushroom species known for its edible and medicinal properties. Polysaccharides have been the focus of research because of their potential bioactivities; nonetheless, the structural complexity and molecular weight have hindered a complete understanding of their bioactivities. In this study, AP-1 polysaccharide was isolated from A. auricula-judae and subjected to ultrasonic degradation at different time points to improve their anti-inflammatory effects. The results showed that when AP-1 was degraded for 9 min (AP-2) and 20 min (AP-3), the NO inh

Engineering Escherichia coli Nissle 1917 Carrying PD1 Agonists Resolves Intestinal Inflammation via Local Immune Modulation.

Immunological imbalance is a key factor in the progression of intestinal inflammation, yet effective treatments remain elusive. Using a radiation-induced intestinal injury model, we investigated the causes of inflammation at the single-cell level and identified abnormal T-cell activation as a major contributor. To address this, we targeted the PD1 signaling pathway to suppress T-cell activation and evaluated the anti-inflammatory and intestinal repair effects of the PD1 agonist displaying probiotic Escherichia coli Nissle 1917 (EcNMP1-M) in two mouse models. Encapsulated in Eudragit L100-55 fo

Direct Modulation of Host Cells and Gut Microbiota by Orally Delivered Antioxidant Nanocages for Colitis Treatment.

Ulcerative colitis (UC) involves a complex interplay of mucosal inflammation, intestinal barrier dysfunction, and gut dysbiosis, all exacerbated by elevated reactive oxygen species (ROS). Existing therapies often fail to concurrently address these interrelated pathologies. Here we present an orally administrable nanocage system that targets both host cells and gut microbiota and can simultaneously address immune dysregulation, epithelial damage, and microbial imbalances for UC treatment when loaded with antioxidant enzymes. This system features a poly(tertiary amine oxide) (PTAO)-based zwitter

Tricobalt tetraselenide nanoparticles improve intestinal barrier function by reshaping the gut microbiota and fortifying epithelial tight junctions.

The disrupted intestinal mechanical barrier contributes to the pathogenesis of inflammatory bowel disease (IBD) and irradiation-induced intestinal injury (IRIII), both of which lack sufficient medical countermeasures currently. Reshaping the gut microbiota to increase the short-chain fatty acid (SCFA) levels is instrumental for fortifying the mechanical barrier and mitigating these diseases. Inspired by the fact that supplementation of essential trace elements selenium (Se) and cobalt (Co) can enrich SCFA-producing bacteria and promote microbial fermentation, respectively, we prepared tricobal

Sphingosine-1-phosphate alleviates colitis by regulating macrophage polarization and PI3k-Akt signaling.

INTRODUCTION: Inflammatory bowel disease (IBD) is a complex disease that is characterized by tight junction loss and dysregulation of immune homeostasis. The repair of intestinal integrity and immune function in IBD remains a clinical challenge. Sphingosine-1-phosphate (S1P) has been reported to alleviate radiation-induced salivary gland damage by maintaining epithelial integrity. However, its potential to restore function during IBD has not yet been investigated. METHODS: Dextran sulfate sodium (DSS) was added to the drinking water of C57BL/6 mice for 5 days to induce colitis. Subsequently,

Multi-omics derivation of a core gene signature for predicting therapeutic response and characterizing immune dysregulation in inflammatory bowel disease.

BACKGROUND: Inflammatory bowel disease (IBD) presents unpredictable therapeutic responses and complex immune dysregulation. Current precision medicine approaches lack robust molecular tools integrating transcriptomic signatures with immune dynamics for personalized treatment guidance. METHODS: We performed multi-omics analyses of GEO datasets using machine learning algorithms (LASSO/Random Forest) to derive a four-gene signature. Validation employed ten algorithms and nomogram construction. Immune infiltration (CIBERSORT/ssGSEA), single-cell RNA sequencing, and DSS-colitis models characterize
