Rice Bran Insoluble Dietary Fiber and Its Bound Polyphenols Alleviate Colitis by Regulating the Autophagy-NLRP3 Inflammasome Axis.

Although dietary fiber alleviates colonic inflammation, most studies have focused on carbohydrate components, neglecting bound polyphenols (BPs). Rice bran insoluble dietary fiber (RIDF), rich in BPs, improves colitis; however, how BPs work and their specific contributions remain elusive. Our study demonstrates that BPs are important contributors of RIDF in alleviating colitis. Relative to rice bran dietary fiber without BPs (RIDF_DF), RIDF markedly ameliorated colitis symptoms, mitigated endoplasmic reticulum stress (ER stress), restored the gut microbiome and metabolites, and blocked the PI3

Visualizing Microvascular Remodeling for Precise Staging of Inflammatory Bowel Disease via Ultralow-Dose Magnetic Resonance Enterography.

Accurate staging of inflammatory bowel disease (IBD) using magnetic resonance enterography (MRE) remains challenging because of limited precision. Although microvascular remodeling is a pivotal driver of IBD progression, its contribution to imaging-based activity assessment remains insufficiently characterized owing to the rapid extravasation and short intravascular residence of current contrast agents. Here, an ultralow-dose MRE strategy is developed using a long-circulating (half-life ≈ 5 h), high-relaxivity (r1 = 40.89 mM-1 s-1 at 3.0 T) ultrasensitive nanoprobe to enable direct visualizati

Engineered Calprotectin-Responsive Probiotics for Inflammatory Bowel Disease Treatment.

Inflammatory bowel disease (IBD) is a complex intestinal disorder for which effective treatment options remain limited. The modulation of gut microbiota balance using probiotics holds promise as a therapeutic approach to restore intestinal microenvironment homeostasis. However, the clinical translation of conventional probiotics is constrained by the suboptimal therapeutic efficacy. Here, we constructed a recombinant Escherichia coli Nissle 1917 strain (EcN-CR-IL2) based on an optimized calprotectin-responsive ykgMO promoter to achieve inflammation-dependent expression of the immunomodulatory

SATB2 as a novel messenger of the cGAS-STING signaling pathway drives CUMS-induced depression via gut-brain axis.

The microbiota-gut-brain axis is involved in the pathogenesis of depression; however, whether the cGAS-STING signaling pathway contributes to onset and progression of depression via gut microbiota dysbiosis remains unknown. This study aims to investigate the role of the cGAS-STING signaling pathway in chronic unpredictable mild stress (CUMS)-induced depressive-like behaviors. Additionally, it aimed to identify novel downstream signaling factors of this pathway. A comprehensive mechanistic investigation was conducted using CUMS rats, HT22 cells, and cGAS+/- mice, with behavioral tests to assess

Active components Alignment of Gegen Qinlian Decoction for inflammatory bowel disease: exploring the possible role of the SphK1/S1P/NF-κB pathway.

ETHNOPHARMACOLOGICAL RELEVANCE: Gegenqinlian Decoction (GQD) has been used as a folk remedy for gastrointestinal diseases in China over thousands of years, and demonstrates significant efficacy against inflammatory bowel disease (IBD). We previously showed that the active components alignment of GQD inhibits nuclear factor kappa B (NF-κB) p65 signal transduction and plays a key role in preventing and treating IBD. With growing insights into the gut microbiota and its metabolites, the molecular mechanisms underlying therapeutic effects of GQD against IBD require further elucidation. This is par

From mechanical stress to mesenteric adipocyte dysfunction in Crohn's disease: an essential role of Piezo1-mediated mitochondrial calcium overload.

BACKGROUND & AIMS: Crohn's disease (CD) features hypertrophied mesenteric adipose tissue (htMAT) where dysfunctional adipocytes promote intestinal inflammation via secreting pro-inflammatory adipokines. Our previous work revealed that pathological extracellular matrix (ECM) remodeling correlates with adipocyte dysfunction in htMAT; but the underlying mechanism remains unknown. This study aimed to determine whether aberrant ECM stiffness drives adipocyte dysfunction and the underlying mechanism. METHODS: Clinical mesenteric specimens were examined. A tunable 3D hydrogel system modelled mechani

Association of Blastocystis subtype 3 with elevated IL-8-mediated inflammation in patients with irritable bowel syndrome.

BACKGROUND: The role of Blastocystis in the pathophysiology of irritable bowel syndrome (IBS) remains controversial, with emerging evidence suggesting subtype-specific differences in pathogenicity. Interleukin-8 (IL-8), a key pro-inflammatory chemokine, has been implicated in mucosal immune activation in both IBS and parasitic infections. OBJECTIVE: This study aimed to investigate the association between Blastocystis subtypes and clinical manifestations of IBS, with a particular focus IL-8 as part of the systemic pro-inflammatory immune response associated with functional GIT disorders and in

Recent Discoveries of Mechanisms Contributing to Necrotizing Enterocolitis.

Necrotizing enterocolitis (NEC) is a devastating neonatal intestinal disease driven by complex interactions between prematurity, microbial dysbiosis, immune dysregulation, and impaired intestinal barrier function. This article highlights recent advances in NEC pathogenesis, focusing on the role of toll-like receptor 4-mediated inflammatory signaling and the contribution of maternal inflammation and chorioamnionitis to disease susceptibility. Transcriptomic and spatial profiling studies further demonstrate that classical NEC and cardiac NEC possess distinct molecular signatures and identify div

A rare instance of Ogilvie syndrome in a pediatric patient with a complex medical history: A case report.

Acute colonic pseudo-obstruction or Ogilvie's syndrome is an acute dilatation of the large bowel without an obstructive lesion. Symptoms include abdominal distension, nausea and vomiting, constipation, and occasional dyspnea. The disorder most commonly involves the cecum and right colon and is most frequently seen in male patients above the age of 60. It is rare for Ogilvie's to occur in the pediatric population though some infrequent instances have been reported. Here, we report a case of a patient with significant abdominal distension and gaseous distension seen on X-ray, which was diagnosed

Whole king oyster mushroom (Pleurotus eryngii) alleviates high-fat diet-induced low-grade colonic inflammation in association with the gut microbiota-barrier-immune axis.

Whole food-based dietary strategies have attracted increasing attention for their role in preventing metabolic disorders. This study investigated the effects of whole Pleurotus eryngii (WPE) on high-fat diet (HFD)-induced low-grade colonic inflammation in mice, with a focus on the gut microbiota-barrier-immune axis. WPE supplementation attenuated HFD-induced body weight gain, improved colonic histopathology, and enhanced mucus production with increased goblet cell abundance and mucin expression, alongside restoration of epithelial barrier-related proteins and genes. WPE attenuated mucosal immu

Quercetin Ameliorates Cognitive Impairment in Obese Mice by Activating the PI3K/AKT/GSK3β Signaling Pathway via the Microbiota-Gut-Brain Axis.

Obesity and its associated insulin resistance are important risk factors for cognitive impairment. In this study, a 10-week C57BL/6 mouse model was established to investigate the effects of dietary quercetin supplementation at 0.05% (w/w) on HFFD-induced obesity-related cognitive impairment. The results showed that quercetin significantly reduced HFFD-induced body weight gain (P < 0.05) and improved serum lipid abnormalities and cognitive performance. Integrated multi-omics analysis further revealed that quercetin reshaped the gut microbiota composition, notably increasing Akkermansia muciniph

High CLA-producing Lactiplantibacillus plantarum MNN1 alleviates irritable bowel syndrome through activation of intestinal PPARα signaling pathway.

Irritable bowel syndrome (IBS) is a common gut-brain axis disorder characterized by visceral hypersensitivity, abdominal pain, and anxiety. Although probiotics have emerged as a promising strategy for IBS management, the specific metabolite-driven mechanisms and their role in gut-brain axis modulation remain largely unclear. In this study, 18 Lactiplantibacillus plantarum strains were screened for conjugated linoleic acid (CLA) production, and strain MNN1 was identified as a high-CLA producer, with a yield of 426 μmol/L, approximately threefold higher than that of the reference strain WCFS1. Gut microbiota-driven imbalance of the gut-brain-immune homeostatic network in constipation: Pathogenic mechanisms and evolving therapeutic prospects.

Chronic constipation (CC) is increasingly recognized as a systems-level disorder that cannot be explained by impaired intestinal motility alone. Here, we integrate evidence from clinical cohorts, animal models, and mechanistic studies to examine how gut microbial dysbiosis and altered microbial metabolites perturb epithelial barrier integrity, immune homeostasis, enteric neural activity, and gut-brain signaling. We propose a "gut-brain-immune homeostatic network" in which short-chain fatty acids (SCFAs), tryptophan metabolites, bile acids, and other microbial signals may sustain self-reinforci

Repurposing Ionophores to Tackle Antimicrobial Resistance with a Focus on PBT2.

Antimicrobial resistance (AMR) remains a growing global health challenge. While resistant bacterial strains continue to emerge and spread, the development of new antibiotics has remained slow. This gap has increased interest in drug repurposing as a promising strategy to combat multidrug-resistant (MDR) pathogens. Ionophores are a structurally diverse group of natural and synthetic compounds that transport ions across biological membranes. Many ionophores were originally developed and widely used for the prevention and treatment of coccidiosis, a common protozoan parasitic intestinal disease i

Gut Microbiota Remodeling Drives Cognitive Improvement: A Microbiota-Dependent Study of Malvidin-3-O-glucoside against Alcohol-Related Cognitive Impairment.

Gut microbiota dysbiosis triggers alcohol-related cognitive impairment (ARCI). Malvidin-3-O-glucoside (Mv3G), a highly bioactive monomeric anthocyanin abundant in pigmented fruits and cereals, can relieve neurodegenerative diseases by regulating gut microbiota (GM). Nevertheless, the gut microbiota-mediated protective effect of Mv3G against ARCI remains unknown. In this study, an alcohol-induced ARCI mouse model combined with fecal microbiota transplantation (FMT) was built to explore Mv3G's protective effects. Results show that Mv3G acts in an intact gut flora-dependent manner. It repairs int

The role of gut microbiota in paediatric short bowel syndrome.

Short bowel syndrome (SBS) is one of the leading causes of intestinal failure in children. Its development is influenced not only by the length and function of the residual intestine but is also closely related to the homeostasis of the gut microbiota. In recent years, the role of the gut microbiota in intestinal adaptation, metabolic regulation, and the development of complications in pediatric SBS has gradually garnered attention. This review summarizes the characteristics of the gut microbiota, mechanisms underlying dysbiosis, associations with complications, and microbiota-targeted therape
