Ferroptosis is an iron-dependent form of cell death driven by lipid peroxidation1. Here we identify spermine-a polyamine derived from spermidine2-as an endogenous iron chelator that directly suppresses ferroptosis. Integrating metabolomics, stable isotope tracing and biophysical studies of the interaction between spermine and Fe2+ ions, we demonstrate that aldehyde dehydrogenase 18 family member A1 (ALDH18A1) promotes an alternative glutamine-dependent pathway for de novo spermine synthesis. This process limits iron availability and lipid peroxidation in hepatocellular carcinoma. Genetic or ph

Hypertension is a major global public health issue characterized by complex pathogenesis and a high risk of multiple comorbidities. Gut microbes play a crucial role in blood pressure regulation. This review aims to clarify the roles of distinct gut microbiota compositions in regulating blood pressure and to analyze their potential mechanisms of action in hypertension. Gut microbes produce metabolites that regulate the host immune system, metabolic homeostasis, and other physiological processes, including blood pressure. Key microbial metabolites, such as short-chain fatty acids (SCFAs) and tri

Lipopolysaccharide (LPS), a component of Gram-negative bacteria, is a potent activator of the innate immune system and has been implicated in the pathogenesis of eye diseases, including age-related macular degeneration (AMD), primary open-angle glaucoma (POAG), and diabetic retinopathy (DR). LPS enters systemic circulation through bacterial lysis or active secretion, leading to endotoxemia-either acute or chronic. Chronic endotoxemia, often resulting from gut dysbiosis, may link microbial imbalances to eye diseases. Indeed, chronic LPS exposure may contribute to both systemic and local inflamm

BACKGROUND AND AIMS: Recommendations on anticoagulation in patients with short bowel syndrome (SBS) following acute mesenteric ischaemia (AMI) or complicated by catheter-related thrombosis (CRT), are limited. Guidance is specifically lacking on antithrombotic drug selection, dosing, treatment duration and monitoring. Moreover, current recommendations do not account for the potentially impaired drug absorption in patients with SBS. This multidisciplinary position paper aims to develop a practical management guide for antithrombotic therapy in patients with SBS following AMI, as well as CRT.

Cardiometabolic diseases are increasingly recognized as disorders of chronic low-grade systemic inflammation and gut barrier dysfunction that mutually reinforce one another. Each condition amplifies the other through progressive injury to the intestinal epithelium. Compromise of the mucus layer, altered tight junction dynamics, dysbiosis, and impaired epithelial restitution promote intestinal permeability and enable the translocation of lipopolysaccharide and other microbial products into the circulation, thereby inducing metabolic endotoxemia. This gut derived inflammatory signal activates To

Pulmonary hypertension (PH) is a severe disorder characterized by progressive pulmonary vascular remodeling, increased pulmonary arterial pressure, and right heart failure. Beyond local abnormalities within the pulmonary vasculature, growing evidence suggests that PH is shaped by systemic inflammation, metabolic dysregulation, gut microbiota imbalance, and disruption of host circadian organization. In this review, we examine the circadian clock-gut microbiota-gut-lung axis as a time-dependent regulatory network in PH. We focus on core clock gene dysfunction, impaired intestinal barrier integri

Vascular cognitive impairment (VCI) is a major form of dementia with a complex pathogenesis. Emerging evidence indicates a strong interaction between intestinal mucosal barrier integrity and neuroinflammation, which has been proposed as a candidate contributor in VCI progression. This review comprehensively reviews the initiation and amplification of neuroinflammation in chronic cerebral ischemia. It highlights how impairment of the intestinal mucosal barrier, gut microbiota dysbiosis, and translocation of microbial metabolites contribute to central neuroinflammation via circulatory and neural

Juvenile intestinal ischemia-reperfusion (JII/R) is a pediatric surgical emergency caused by mesenteric vessel occlusion and has a high mortality rate. Malrotation can cause intestinal ischemia in infants due to midgut volvulus. It affects not only the intestine itself but also other organs, such as cardiac tissue. Therefore, searching for more effective therapeutic solutions is an essential critical need. This directed our thoughts to evaluate the role of ambrisentan (AMB) in a rat model of induced JII/R by clamping the superior mesenteric artery. Forty juvenile male Wistar albino rats (3-4 w

Disturbances of 24-hour or circadian rhythms imposed by everyday irregular work and/or social schedules have been linked to vascular disease, including ischemic stroke. Using an established shift work-like paradigm and preclinical model for ischemic stroke, we have shown that environment-induced circadian dysregulation exacerbates stroke outcomes differentially to a greater extent in male than female rats. Because more severe stroke outcomes and circadian rhythm disturbances have been linked to gut pathophysiology, the present study examined the effects of chronic shifts in the LD cycle on gut

Intestinal disorders such as inflammatory bowel diseases and gastrointestinal fistulae are marked by chronic inflammation, barrier dysfunction, and impaired repair, conditions insufficiently addressed by current treatments. Regenerative strategies able to restore epithelial integrity and support stromal and vascular remodeling are therefore highly needed. The stromal vascular fraction (SVF), a heterogeneous and autologous cell population from adipose tissue, offers angiogenic, immunomodulatory, and regenerative properties, while three-dimensional (3D) bioprinting enables its embedding in hydro

BACKGROUND AND PURPOSE: Ischemic stroke (IS) remains a leading global cause of mortality and disability. Although current reperfusion strategies have advanced, the complex pathological mechanisms underlying IS continue to significantly limit clinical prognosis. Recently, the pivotal role of the microbiota-gut-brain axis (MGBA) in the pathological evolution of IS has garnered widespread attention. This review systematically delineates the multidimensional regulatory roles of the gut microbiota and its metabolites in IS pathogenesis, with a specific focus on immuno-metabolic crosstalk, cellular

Inflammatory bowel diseases (IBD), including Crohn's disease and ulcerative colitis, are chronic immune-mediated diseases that are increasingly recognized as systemic diseases with significant cardiovascular consequences. Growing epidemiological evidence suggests that patients with IBD face an increased risk of atherosclerotic cardiovascular disease (ASCVD) that cannot be fully explained by traditional cardiovascular risk factors. This excess risk is most pronounced in younger patients and during periods of active intestinal inflammation. This review summarizes current knowledge on common path

Metabolic syndrome (MetS) is a complex and heterogeneous condition characterized by the coexistence of obesity, type 2 diabetes mellitus (T2DM), hypertension, chronic low-grade inflammation, and metabolic dysfunction. Increasing evidence suggests that the gut microbiota plays a central role in the development and progression of MetS by influencing host metabolism, intestinal barrier integrity, immune activation, endocrine signaling, and vascular homeostasis. This narrative review summarizes current evidence regarding gut microbiota alterations across major obesity-related metabolic phenotypes,

INTRODUCTION: Behçet's disease e (BD) is a chronic, relapsing, multisystem inflammatory disorder that if not successfully treated can lead to severe, organ or life-threatening complications. Despite treatment with glucocorticoids, immunosuppressants, and tumor necrosis factor (TNF) inhibitors, some patients still have refractory disease that mandates additional therapeutic options. The pathogenesis of BD involves dysregulated innate and adaptive immune responses with multiple cytokines signaling through the Janus kinase (JAK)-signal transducer and activator of transcription (STAT) pathway. Despite advances in systemic therapies, metastatic colorectal cancer (mCRC) remains largely incurable, underscoring persistent gaps in our understanding of metastatic progression and therapeutic resistance. Emerging evidence suggests that gut and tumor-associated microbial communities may contribute to metastatic progression by shaping organ-specific niches, disrupting intestinal and vascular barriers, remodeling immune and stromal microenvironments, and altering host-microbial metabolism. This review synthesizes current evidence on the involvement of gut and intratumoral microbial communities
