The Crimean-Congo haemorrhagic fever virus hijacks the liver lipid metabolic pathway for virion production.

The Crimean-Congo Haemorrhagic Fever Virus (CCHFV), a tri-segmented negative-strand virus that belongs to the Orthonairovirus genus, is highly pathogenic in humans but not in other host species. Following infection in humans, CCHFV disseminates widely but replicates most strongly in hepatocytes, indicating a preferential liver tropism. As hepatocytes are the primary site for production and secretion of lipids, here we sought to characterize the interplay between CCHFV and lipoprotein metabolism in hepatocytes. First, we found that CCHFV particles display a heterogeneous profile of density, sug

Crimean-Congo Haemorrhagic Fever Virus Exposure in Horses and Donkeys in Border Livestock Markets of Nigeria.

INTRODUCTION: Crimean-Congo haemorrhagic fever virus (CCHFV) is a tick-borne zoonotic pathogen of major public health importance in Africa. Although CCHFV exposure has been reported in several livestock species in Nigeria, data on equids remain limited despite their extensive involvement in livestock movement and trade. This study assessed CCHFV seroprevalence in horses and donkeys in major international livestock markets in northern Nigeria and examined factors associated with exposure. METHODS: From June 2023 to July 2024, a longitudinal sero-surveillance study was conducted in three high-v

Genomic characterisation of Crimean-Congo haemorrhagic fever virus (CCHFV) in Tajikistan identifies a novel reassortant virus.

Crimean-Congo haemorrhagic fever virus (CCHFV) is an important human tick-borne pathogen, able to cause severe haemorrhagic fever. CCHFV is endemic in Tajikistan, which records between 5-38 cases of CCHF a year from southern regions. Molecular surveillance of CCHFV is crucial to implement effective prevention and control strategies, understand viral evolution, study transmission dynamics, and develop effective diagnostics, therapeutics, and vaccines. While the presence of Asia-1 and Asia-2 genotypes has been previously reported, only two historical samples from Tajikistan have been fully seque

Sero-epidemiology of Crimean-Congo haemorrhagic fever in mixed crop-livestock farming households in Burkina Faso: a one health study.

BACKGROUND: Crimean-Congo haemorrhagic fever (CCHF) is a severe, widespread, tick-borne viral zoonotic infection. It is caused by an orthonairovirus that is transmitted by ticks. Sero-epidemiological studies in humans and livestock are valuable indicators of viral circulation and infection risk. This study aimed to investigate the seroprevalence and factors associated with CCHF virus exposure in humans and livestock in mixed crop-livestock farming households in rural Burkina Faso. METHODS: A cross-sectional animal-human linked study was conducted in 149 rural households across 16 randomly sel

A hyperimmune ovine antibody-based therapeutic candidate against Crimean-Congo haemorrhagic fever virus targeted to the Gn and Gc glycoproteins: high antibody levels with neutralization activity but lack of protective efficacy in a mouse model.

BACKGROUND: Crimean-Congo haemorrhagic fever virus (CCHFV) is a tick-borne pathogen that has a wide geographical range and has the potential to cause severe disease in those infected. There are no currently approved vaccines or therapeutics, so the development and assessment of new approaches are urgently required. METHODS: A polyclonal antibody-based therapeutic was developed based on immunization of sheep with the major glycoprotein antigens from CCHFV. Along with purified immunoglobulin G (IgG) from hyperimmune sera, fragmentation of the F(ab')2 region was also performed. The pharmacokinet

Crimean-congo haemorrhagic fever virus circulates within broad ecological networks of ticks and vertebrates.

We produced spatial datasets of the known distribution of Orthonairovirus haemorrhagiae (formerly Crimean-Congo haemorrhagic fever virus, CCHFV) by compiling human cases, virus isolations, and serological data from humans and animals, spanning from Europe to southern Africa, with the aim of virus range modelling. Models based solely on climatic variables produced unrealistic and overly extensive predictions, overestimating suitability in northern Europe and confirming that climate alone poorly explains CCHFV range. Approaches using only tick species distributions underperformed in many parts o

Short Report: Update on CCHFV Seroprevalence in Mauritanian Camels and Cattle Sampled Between 2020 and 2021.

OBJECTIVES: Crimean-Congo hemorrhagic fever virus (CCHFV) is a zoonotic tick-borne virus causing severe and potentially fatal disease in humans, with case fatality rates of up to 30%. In contrast, domestic ruminants and camelids remain asymptomatic. Since the first reported case in Mauritania in 1983, CCHFV antibodies have been repeatedly detected in humans and animals. This study aimed to assess the local circulation of CCHFV in camels and cattle in southern Mauritania and in Nouakchott. METHODS: A total of 317 blood samples (174 cattle, 143 camels) were collected from southern regions borde

Structural and functional characterisation of the Crimean-Congo haemorrhagic fever virus RNA dependent RNA polymerase.

Crimean-Congo Haemorrhagic Fever Virus (CCHFV) is found across Africa, Asia, and the Middle East where it can cause Haemorrhagic outbreaks with high case fatality rates. Central to the viral life cycle is the viral L-protein, a crucial and multifunctional protein which both transcribes and replicates the viral genome. Here, we present the cryoEM structures of an RNA free and a 5' promoter bound complex, describing the core catalytic RNA-dependent RNA polymerase (RdRp). We observe an RdRp that is substantially larger than related L-proteins and contains domain insertions unique to the nairoviru

"Giant tick" attacks: dynamics of Hyalomma lusitanicum and detection of Rickettsia sibirica mongolitimonae in southern France.

Since 2019, media reports have raised concerns about the emergence of "giant ticks" in Europe, particularly Hyalomma marginatum, due to its potential to transmit the Crimean-Congo haemorrhagic fever virus (CCHFV). In 2022, following several reports of unusually large ticks displaying aggressive behaviour toward humans in the Étang de Bolmon area (Bouches-du-Rhône, southern France), a popular area for walking and nature exploration, an investigation was conducted to characterise the local tick fauna, their seasonal activity, and associated microorganisms. Between 2022 and 2025, a total of 2,940

Dispersal of Hyalomma rufipes by migratory birds in northeastern Spain: Implications for Crimean-Congo haemorrhagic fever surveillance.

Migratory birds play a crucial role in the long-distance dispersal of ticks and tick-borne pathogens, contributing to the introduction of tick species and pathogens into new areas. Among those ticks, Hyalomma rufipes is a key concern due to its role as a vector of Crimean-Congo haemorrhagic fever virus (CCHFV) and its potential to establish in southern Europe. We assessed tick infestation, CCHFV presence, and both the probability of H. rufipes infestation and its genetic diversity in migratory birds captured at four key stopovers in northeastern Spain Mediterranean (NESM) ecological region dur

Structures and inhibition of the Crimean-Congo haemorrhagic fever virus polymerase.

Crimean-Congo haemorrhagic fever virus (CCHFV) is a tick-borne virus and causes severe, often fatal, human infections. Lacking licensed vaccines or drugs, CCHFV is a World Health Organization priority pathogen requiring urgent development of medical countermeasures1,2. The CCHFV Large (L) protein functions as the viral RNA-dependent RNA polymerase CCHFV-L, representing a promising antiviral target, and is among the largest viral polymerases in the order Bunyavirales. Here we define the cofactors required for CCHFV-L RNA synthesis in vitro, enabling capture and determination of elongating CCHFV
