BACKGROUND: Ebola virus disease (EVD) is often regarded as severe and highly fatal, but growing evidence suggests that subclinical or minimally symptomatic infections occur and frequently go undetected. During the 2017 outbreak in Likati Health Zone, only 8 cases were confirmed despite many reported exposures. We evaluated the extent of asymptomatic or unrecognized Ebola virus infection and associated factors among contacts of reported cases.
METHODS: In November 2017, we conducted a cross-sectional community-based serosurvey among contacts originally identified through Ministry of Health rec

Ebola virus (EBOV) infection causes severe hemorrhagic fever marked by dysregulated cytokine production, impaired antiviral defenses, and multi-organ failure. Macrophages are primary targets of EBOV, and viral replication profoundly alters macrophage transcriptional programs, driving hyperinflammation. Although long non-coding RNAs (lncRNAs) are increasingly recognized as regulators of immunity and viral pathogenesis, their roles in EBOV infection remain poorly understood. We performed comprehensive transcriptomic profiling of primary human monocyte-derived macrophages infected with the highly

Bundibugyo virus disease (BVD), caused by Bundibugyo virus (BDBV), is associated with substantial morbidity and mortality, with previous outbreaks reporting case fatality rates of 30%-50%. The ongoing BDBV outbreak in the Democratic Republic of the Congo and Uganda highlights the urgent need for virus-specific research tools and medical countermeasures. Unlike Ebola virus disease caused by Zaire ebolavirus, no licensed vaccines or specific therapeutics are currently available for BVD. The lack of research tools has limited studies with BDBV. To facilitate antiviral testing and neutralization s

BACKGROUND/OBJECTIVES: Ebola Virus Disease (EVD) remains a public health threat in sub-Saharan Africa. The 10th Ebola outbreak in the Democratic Republic of the Congo (DRC) in 2018-2020 led the Rwanda Ministry of Health to launch a large-scale Ebola vaccination campaign using the two-dose Ad26.ZEBOV and MVA-BN-Filo regimen. The campaign was implemented by local organizations, the Center for Family Health Research and Rinda Ubuzima, in partnership with the Rwanda Biomedical Center.
METHODS: The campaign targeted those who live near or routinely cross the Rwanda/DRC border and unvaccinated firs

The ongoing 2026 Bundibugyo ebolavirus outbreak in eastern Democratic Republic of the Congo highlights the continued vulnerability of Central Africa to recurrent Ebola emergence. This outbreak emerged less than six months after the previous one ended, appears to represent one of the shortest documented inter-epidemic intervals in the Democratic Republic of the Congo based on currently available outbreak reports. Current surveillance systems remain largely reactive, focusing on the detection of human cases after zoonotic spillover has occurred. While strengthening health systems, diagnostic cap

The 2026 Bundibugyo virus disease (BVD) outbreak has exposed persistent weaknesses in health-security preparedness across the East African Community (EAC). Although regional preparedness has improved, major gaps remain in early detection, cross-border collaboration and surveillance, laboratory capacity, infection prevention and control, and community engagement. These vulnerabilities have been intensified by declining external health-security funding since 2025, incomplete implementation of domestic health-financing commitments, and the fragility of systems that were built with substantial don

The ongoing 2026 outbreak of Ebola virus disease caused by Bundibugyo virus (BDBV) in the Democratic Republic of the Congo and Uganda has renewed attention to one of the least studied human-pathogenic orthoebolaviruses. Since its discovery in western Uganda in 2007, only two recognised outbreaks of BDBV had been reported, limiting opportunities to define the epidemiology, pathogenesis, diagnosis, clinical spectrum, and optimal management of BDBV or to develop species-specific countermeasures. The current outbreak, declared a Public Health Emergency of International Concern by WHO on May 17, 20

BVD is a severe filoviral haemorrhagic fever caused by the Bundibugyo virus (BDBV), for which outbreak-related evidence remains limited compared with that available for Zaire ebolavirus. This perspective summarizes current evidence on transmission dynamics, presymptomatic infectiousness, post-recovery viral persistence and the duration of infectiousness, distinguishing BDBV-specific data from evidence extrapolated from other orthoebolaviruses. Available epidemiological data from the 2007-2008 outbreak in Uganda indicate that BDBV transmission occurs primarily through direct contact with the bl

Ebola virus disease (EVD) remains one of the most severe and complex viral hemorrhagic fevers confronting global public health systems. Since its first recognized emergence in 1976, recurrent outbreaks caused primarily by Zaire ebolavirus have demonstrated the extraordinary epidemic potential of filoviruses, particularly in regions with fragile healthcare infrastructure. The unprecedented West African epidemic of 2013-2016 fundamentally altered scientific understanding of Ebola virus transmission, pathogenesis, clinical progression, and outbreak management. Subsequent outbreaks in the Democrat

BACKGROUND: Uganda experiences recurrent Ebola disease (EBOD) outbreaks, yet national suitability maps often pool virus species or are too coarse for district-level planning. We aimed to generate species-resolved, 1 km relative environmental suitability surfaces for reported Ebola spillover events in Uganda; quantify the contribution of environmental, anthropogenic, and reservoir-related predictors to modeled suitability; and translate continuous outputs into operational prioritization tiers for district surveillance and preparedness.
METHODS: We compiled 71 laboratory-confirmed reported Ebol

Multiplex bead-based immunoassays (MIAs) are promising tools for simultaneously detecting humoral immunity to multiple targets, potentially playing a crucial role in serosurveillance and vaccine response assessments. However, evaluation of assay performance is paramount prior to widespread use. This study presents a performance evaluation of a pan-filovirus MIA through characterization of the analytical range for the EBOV glycoprotein (GP) target and assessments of assay precision and antigen discrimination. The precision of the MIA was evaluated by comparing the detection of anti-filovirus an

Since mid-May 2026, a rapidly evolving Ebola virus disease (EVD) outbreak caused by the Bundibugyo strain (Orthoebolavirus bundibugyoense) has been spreading in eastern Democratic Republic of Congo (Ituri, North and South Kivu provinces) and Uganda (Kampala and Wakiso areas). Uganda has so far reported 14 imported cases and 5 secondary transmissions according to the World Health Organization (WHO). As of 30 July 2026, 3605 confirmed cases, including 1587 deaths, have been reported in the Democratic Republic of the Congo. Unlike previous Zaire outbreaks, no specific licensed vaccine or antivira

BACKGROUND: There are currently no licensed vaccines for disease caused by Bundibugyo virus (BDBV). The recombinant vesicular stomatitis virus-Zaire Ebola virus glycoprotein (rVSVΔG-ZEBOV-GP) vaccine has been widely deployed during Ebola virus disease outbreaks caused by Ebola virus (EBOV). Human studies have shown cross-reactive BDBV antibody responses following licensed Ebola virus disease vaccine administration, but the durability and longitudinal kinetics of these responses in populations vaccinated during outbreaks remain unknown. We aimed to evaluate BDBV glycoprotein seroreactivity foll

Ebola virus (EBOV) causes severe and often fatal Ebola virus disease (EVD) in humans, posing a major threat to global public health. Since EVD was first identified in the Democratic Republic of Congo (DRC) in 1976, the World Health Organization (WHO) has declared it a Public Health Emergency of International Concern (PHEIC) on three occasions. EBOV infection is characterized by rapid progression from non-specific febrile illness to severe gastrointestinal symptoms, followed by hemorrhagic fever and multi-organ dysfunction, with case fatality rates reaching as high as 90%. The liver is a primar

BACKGROUND: Ectopic pregnancy (EP) is an obstetric emergency; early diagnosis is critical for maternal survival. In resource-limited settings, such as Ebola treatment units (ETU), delayed diagnosis or management of EP often leads to rupture and life-threatening hemorrhagic shock.
CASE PRESENTATION: We report the case of a pregnant woman in her 30s who was admitted to the ETU with suspected Ebola virus disease in the Democratic Republic of Congo (DRC). Her clinical presentation included fever and abdominal pain, which began 4 days before her admission. Thirteen months earlier, she had recovere

East and Central Africa experiences heightened threats for Ebola Virus Disease (EVD), with the latest outbreak caused by the Bundibugyo ebolavirus (BDBV) reported in the Democratic Republic of the Congo (DRC) and Uganda on 15th May 2026. The BDBV outbreak comes just over a year following the declaration of Sudan Virus Disease (SVD) outbreak in Uganda on 30th January 2025. Following the 2025 outbreak, a race against the clock mobilization occurred over the subsequent four days to launch a ring vaccination trial for a candidate vaccine against SVD. During this four-day period, the investigation
