Evaluation of respiratory toxicity and peritoneal mesothelioma induced by heat-treated and milled amosite following intratracheal and intraperitoneal injection in rats.

Amoste is highly carcinogenic, and its incomplete removal from the environment continues to pose significant health risks. High temperatures are known to alter the crystal structure of asbestos. In this study, we evaluated the carcinogenicity and respiratory toxicity of heat-treated amosite (AM-1000; heated at 1,000°C) and its milled derivative (AM-1000G; heat-treated at 1,000°C and milled), using untreated amosite as a positive control. Physicochemical analyses confirmed that AM-1000G predominantly consisted of non-asbestiform structures with an aspect ratio of less than 3. In a single intrat

PDK4-driven metabolic reprogramming enhances mesothelial cell invasion in colorectal cancer peritoneal metastasis.

Mesothelial cells play an important role in colorectal cancer peritoneal metastasis (CRC-PM), where they support tumor growth and invasion. In this study, we investigate the molecular mechanisms by which mesothelial cells contribute to CRC metastasis. Using single-cell RNA sequencing (scRNA-seq) on tissue samples from 12 CRC patients with peritoneal metastasis, we identify PDK4 as a key gene in mesothelial cells during metastasis. The expression of PDK4 is significantly greater in mesothelial cells undergoing mesothelial-to-mesenchymal transition (MMT) compared to normal peritoneal cells, sugg

Effect of hyperthermic intraperitoneal chemotherapy on patients with advanced colorectal cancer: a systematic review and meta-analysis.

BACKGROUND: Advanced colorectal cancer (CRC) predisposes to peritoneal metastases (PM), leading to a decreased survival rate. Advanced CRC includes CRC with PM (CRC-PM) and locally advanced high-risk CRC without PM. The effectiveness of hyperthermic intraperitoneal chemotherapy (HIPEC) in prolonging survival and in treating or preventing PM after surgery for advanced CRC is still uncertain. METHODS: A search of PubMed, Cochrane, Embase, and Web of science databases for relevant studies prior to April 2024 was performed. Data were analyzed using Stata/MP 17.0 software. The primary outcomes incl

Association of clinicopathological characteristics and baseline peripheral blood lymphocyte subsets with efficacy of first-line immunotherapy in advanced gastric cancer.

BACKGROUND: Immunotherapy has revolutionized treatment for advanced gastric/gastroesophageal junction adenocarcinoma (G/GEJC). However, identifying convenient biomarkers for predicting therapeutic efficacy remains challenging. This study investigated the association between clinicopathological characteristics and baseline peripheral blood lymphocyte subsets with efficacy of first-line chemotherapy combined with immune checkpoint inhibitors (ICIs) in proficient mismatch repair (pMMR) human epidermal growth factor receptor 2 (HER2)-negative advanced G/GEJC. METHODS: This retrospective study enr

Consensus Molecular Subtypes Inform HIPEC Agent Selection in Colorectal Cancer Peritoneal Metastases.

BACKGROUND: Although hyperthermic intraperitoneal chemotherapy (HIPEC) added to cytoreductive surgery shows promise for colorectal cancer with peritoneal metastases (CRC-PM), effectiveness remains variable. Given the predominance of consensus molecular subtype 4 (CMS4) in CRC-PM and resistance to standard agents, we hypothesized CMS4 exhibits distinct drug sensitivities. MATERIALS AND METHODS: Drug sensitivity data from the DepMap PRISM Repurposing Dataset for 34 CRC cell lines were classified into the 4 CMS subtypes. Five intraperitoneal agents were analyzed: mitomycin-C, oxaliplatin, irinot

Prognostic stratification in gastric cancer with peritoneal metastasis: tumor biology and diagnostic modality as a surrogate for disease burden.

BACKGROUND: Gastric adenocarcinoma with peritoneal metastasis (PM) has a poor prognosis, yet clinical outcomes vary significantly. This study aimed to identify independent prognostic determinants of PM-specific survival (PM-OS), focusing on tumor biology and disease burden surrogate. METHODS: We retrospectively analyzed 166 patients with gastric adenocarcinoma and PM treated at a single center between 2015 and 2024. Prognostic factors were evaluated using multivariable Cox proportional hazards models. To mitigate immortal time bias and confounding by indication, receipt of systemic therapy was

Pathological Complete Response Achieved with Colorectal Cancer-Based Chemotherapy for Locally Recurrent Cecal Neuroendocrine Carcinoma after Surgery: A Case Report.

INTRODUCTION: Primary neuroendocrine carcinoma (NEC) of the colon is extremely rare, accounting for only 0.2% of all colorectal malignancies, and is associated with a poor prognosis. Early diagnosis is often challenging, as endoscopic biopsies are frequently misinterpreted as adenocarcinoma. Although platinum-based regimens such as etoposide plus cisplatin or irinotecan are commonly used, no standard chemotherapy protocol has been established. We report a case of locally recurrent cecal NEC that responded remarkably to a colorectal cancer-based chemotherapy regimen, achieving a pathological co

Patient-derived orthotopic xenograft models recapitulate the peritoneal dissemination of pancreatic cancer and delineate its transcriptional and regulatory programs.

BACKGROUND & AIMS: The mechanisms of peritoneal dissemination in pancreatic ductal adenocarcinoma (PDAC) remain unclear partly owing to the lack of patient-derived models that recapitulate this process. This study aimed to establish an orthotopic model of PDAC peritoneal dissemination and to uncover the transcriptional and regulatory programs underlying this process. METHODS: Organoids were established from primary pancreatic tumors and malignant effusions of patients with PDAC and orthotopically transplanted into the pancreas of immunodeficient mice to generate patient-derived orthotopic xeno

Incidental peritoneal metastases in the setting of emergency surgery: Review of evidence and algorithm for oncologic decision-making.

BACKGROUND: Incidental peritoneal surface metastases (PSM) identified during emergency surgery represent a challenging clinical scenario for surgeons. While peritoneal metastases are increasingly recognized as a regional disease amenable to cytoreductive surgery and intraperitoneal chemotherapy, guidelines for incidental findings in the emergency setting remain limited. METHODS: We performed a narrative, evidence-based review of PubMed, Scopus, and recent international consensus statements (2019-2025) addressing the management of PSM and their presentation in emergency surgical contexts. Galectin-1 and peritoneal fibrosis: a novel predictor for peritoneal metastasis in gastric cancer.

Current indicators lack the precision required to accurately predict the PM(peritoneal metastasis) of gastric cancer (GC). The accurate prediction of GC with PM and the implementation of targeted therapeutic strategies hold substantial clinical significance. This study aimed to evaluate the prognostic value of galectin-1 expression and peritoneal fibrosis in predicting PM in GC patients. Immunohistochemical and Masson's trichrome staining were conducted on GC tissues and peritoneal tissues from 125 patients who underwent radical gastrectomy. The correlations among galectin-1 expression in GC t

A novel multimodal combining radiomics and tumor-stroma ratio (TSR) improves diagnosis of gastric cancer peritoneal metastasis.

Peritoneal metastasis (PM) is the most common form of metastasis in gastric cancer (GC), frequently leading to severe complications and a significantly poor prognosis. Prompt and early diagnosis of PM in GC is crucial. However, diagnostic laparoscopy and CT scans, while being the primary methods for identifying PM in GC, have notable limitations, such as being invasive and having low sensitivity. Therefore, developing a diagnostic model for PM in GC based on routine examination results holds substantial importance. In this retrospective study, we enrolled 813 patients from two medical centers

Prophylactic and Immunomodulatory potential of gooseberry (Phyllanthus acidus) and oregano (Coleus amboinicus) extracts against tilapia lake virus (TiLV) in tilapia.

Tilapia Lake virus (TiLV) is an emerging pathogen causing up to 90% mortality in tilapia (Oreochromis niloticus) farming. Enhancing tilapia health by modulating the non-specific immune system with herbal immunostimulants offers a promising strategy to mitigate TiLV outbreaks. This study evaluated the prophylactic efficacy of Gooseberry (Phyllanthus acidus) and Oregano (Coleus amboinicus) extracts against TiLV. Crude ethanolic extracts were prepared by Soxhlet extraction and screened for preliminary phytochemicals. The crude extracts were incorporated into the commercial pellet feed at varying

Perioperative safety and tolerability of pressurized intraperitoneal aerosolized chemotherapy (PIPAC) with concurrent systemic therapy in the same admission as a bidirectional chemotherapy: A retrospective comparative study.

OBJECTIVES: PIPAC has emerged as an important method to deliver intraperitoneal chemotherapy repeatedly and safely to achieve high locoregional concentrations to treat patients with peritoneal surface malignancies. Combination of systemic chemotherapy in the same admission of PIPAC, as a bidirectional chemotherapy seems appealing, as it may enhance the efficacy synergistically while improving the logistics and costs of treatment. In this study we aimed to establish the safety and tolerability of this bidirectional approach. METHODS: This is a retrospective study of a prospectively maintained

Stromal cell-derived itaconate promotes endometriosis via macrophage NRF2 and lysosomal pH modulation.

Endometriosis (EM) is driven by immune dysregulation and macrophage dysfunction, yet the underlying mechanisms remain unclear. Here, metabolomic profiling revealed excessive itaconate accumulation in EM lesions, primarily due to elevated cis-aconitate decarboxylase 1 (ACOD1) expression in ectopic stromal cells (ESCs). ESC-derived itaconate was internalized by peritoneal macrophages, where it suppressed pro-inflammatory activity and phagocytosis, thereby facilitating ESC survival and dissemination. Mechanistically, itaconate exerted dual regulatory effects on macrophages: it activated NRF2 sign
