Characterization of Mouse-Adapted Marburg and Ravn Viruses in Inbred BALB/c and Outbred CD-1 Mice.

Marburgviruses cause severe hemorrhagic disease in humans, yet characterization of rodent models for pathogenesis and medical countermeasure development remains limited. Here, we investigated clinical course, blood chemistry, viral dissemination, and mucosal viral detection in inbred BALB/c and outbred CD-1 mice infected with mouse-adapted (MA)-Marburg virus (MARV)/Angola or MA-Ravn virus (RAVV) to further mouse model development. Regardless of virus or mouse strain, infection resulted in widespread viral dissemination in animals sampled up to 5 days post-infection. However, in animals followe

Genomic Insights into Marburg Virus Strains from 2023 and 2025 Outbreaks in Kagera, Tanzania.

Marburg virus (MARV) is the primary cause of Marburg virus disease (MVD), a severe hemorrhagic fever with a high case-fatality rate. The first reported MVD outbreak in Tanzania occurred in 2023, followed by a second outbreak in 2025, both within the Kagera region. During those MVD outbreaks, 174 suspected cases were identified; of those, 10 were laboratory confirmed. After complete genome assembly and bioinformatic analyses, we found the MARV strains of the 2023 and 2025 outbreaks to be closely related and clustered with MARV strains that caused outbreaks in Rwanda (2024) and Uganda (2014). Machine Learning-Enabled Quantification of Hepatocellular Necrosis in the Liver After Lethal Marburg and Ebola Virus Exposures.

The liver is a key target of pathogenic Marburg and Ebola viruses (MARV and EBOV, respectively), with injury common in severe filovirid disease, yet high-resolution histopathologic quantification is lacking. We addressed this by deploying deep learning (DL) models for pixel-level quantitative analysis of digitized liver pathology slides in lethal rhesus monkey (RM) models of MARV and 2 EBOV variants, Makona and Kikwit. Our DL model segmented arteries, veins, bile ducts, and hepatic necrosis, achieving interobserver variability in necrosis segmentation comparable to that of 3 pathologists. DL-q

Immunoinformatics-guided design of a universal chimeric multi-epitope subunit vaccine against Marburg virus disease and Ravn virus co-infection.

Marburg Virus Disease (MVD) is a life-threatening hemorrhagic fever, caused by two viruses-Marburg virus (MARV) and Ravn virus (RAVV)-that belong to the family Filoviridae. Due to its high mortality rate and the lack of licensed vaccines, MVD remains a major global health concern. In this study, a comprehensive immunoinformatics workflow was used to design a universal chimeric multi-epitope subunit vaccine targeting conserved antigenic regions of both viruses to reduce MVD-related mortality. Antigenicity analysis identified glycoproteins (GP) of MARV and RAVV as the most immunogenic viral prot

MARVpred: machine learning prediction of inhibitors targeting Marburg virus Gene 4 Small ORF protein.

The Marburg virus (MARV), responsible for severe hemorrhagic fevers with mortality rates as high as 90%, remains a significant public health threat. This study employs machine learning to identify inhibitors targeting the MARV Gene 4 Small ORF protein, crucial for the virus's replication and immune evasion. The Gene 4 Small ORF protein is pivotal in taking over the host's cellular mechanisms, facilitating unchecked viral replication and significant immune system disruption. Effective targeting of this protein holds promise for mitigating the viral lifecycle and entry, potentially curbing the s

Coordination of laboratory and diagnostic services during public health emergencies: a qualitative study.

BACKGROUND: Diagnostics and laboratory testing are critical components of facilities’ systems for emergency response to infectious diseases. Yet, critical gaps exist in the testing and diagnostic capacities of faith-based health providers, particularly those in low and middle-income countries, limiting their response to health emergencies. Accordingly, the Coronavirus Disease 2019 (COVID-19) Strategic Preparedness and Response Plan for the World Health Organization African Region (1 February 2021–31 January 2022) was used to examine the capacity for laboratory and diagnostic services in a Chri

Systematic review on Marburg virus prevalence and persistence in animals.

INTRODUCTION: Marburg Virus Disease (MVD) is a fatal zoonotic disease of humans and nonhuman primates caused by the Marburg virus (MARV) of the Filoviridae family, and presenting as hemorrhagic fever with a high fatality rate. Egyptian fruit bats, Rousettus aegyptiacus, are the principal natural reservoir, with evidence linking them to most human outbreaks. METHODS: This systematic review evaluated the prevalence of MARV in bats, domestic animals, and rodents, as well as the duration of antibodies and potential routes of viral shedding. A comprehensive search of six (6) scientific databases i

Molecular evolutionary insights into the host-virus relationship between Marburg Virus and its bat reservoir.

Bats, often silent carriers of lethal pathogens, serve as natural reservoirs for numerous viruses capable of crossing species barriers with devastating effects. Among them, Rousettus aegyptiacus (R. aegyptiacus) has emerged as a pivotal species in understanding viral spillovers. This fruit bat hosts the Marburg virus (MARV), supporting its replication and occasional transmission to humans, where it causes fatal hemorrhagic fever. However, a striking paradox remains while MARV infection is deadly in humans, R. aegyptiacus remains asymptomatic. Driven by this contrast, our study explores the cod

Detection of Marburg Virus Antibodies 25 Years After Outbreak in Watsa, Democratic Republic of the Congo.

BACKGROUND: To date, 18 Marburg virus disease (MVD) outbreaks have been reported, most recently in Tanzania and Ethiopia. From 1998 to 2000, a large MVD outbreak in Watsa/Durba region, Democratic Republic of the Congo (DRC), resulted in 154 cases and 128 deaths. Given the public health risks posed by MARV, the lack of approved vaccines and therapeutics, and unpredictability of future outbreaks, this study aimed to assess the anti-MARV seroprevalence in Watsa/Durba. METHODS: Samples collected in May 2023 (n = 313) included known MVD survivors (n = 5), close contacts (n = 22), and other residen

Nucleoprotein and glycoprotein based serological assays for detection of Marburg virus infections.

BACKGROUND: Marburg virus disease (MVD) is a rare but highly fatal zoonotic illness. Current serological tools are limited by their reliance on glycoprotein (GP)-based antigens or Marburg virus infected cell lysates, which cannot distinguish natural infection induced immune responses from GP-based vaccines. This diagnostic gap complicates the detection of missed cases particularly in areas of high vaccination coverage. METHODS: We developed and validated two complementary assays: a nucleoprotein C-terminal tail (NPct) Mix-and-Read (MR) assay and a recombinant GP1,2ΔTM ELISA. Serum samples use

Diagnostic and surveillance strategies for Marburg virus persistence in the male reproductive tract: insights from Ebola and Zika viruses.

The recent outbreak of the Marburg virus (MARV) in Rwanda was effectively controlled, but concerns remain regarding the virus's persistence and its long-term effects. MARV can remain in the testes of nonhuman primates, where it harms Sertoli cells and compromises the blood-testis barrier, which may have implications for male infertility. This persistence is aided by immunosuppressive regulatory T cells (Tregs), which allow the virus to evade the immune system. Drawing comparisons with Ebola and Zika viruses, we propose that MARV may contribute to male infertility and recommend further studies
