Contrast-enhanced ultrasound quantitative parameters for predicting benign and malignant greater omentum lesions: an applied study.

OBJECTIVE: To explore the value of contrast-enhanced ultrasound (CEUS) quantitative parameters in differentiating benign and malignant diseases of the greater omentum and to establish a new CEUS Greater Omentum Imaging Reporting and Data System (CEUS-GOIRADS). METHODS: We analyzed 15 CEUS parameters from 124 patients undergoing omental biopsy (2020–2024) to develop a noninvasive predictive model and assess its diagnostic value. Statistically significant parameters were further analyzed using receiver operating characteristic (ROC) curve cutoff values to determine odds ratios (ORs) for each gro

A Natural Sweetener-inducible Genetic Switch Controls Therapeutic Protein Expression in Mammals.

Cell-based therapies are recognized as the next generation living therapeutics in medicine, especially through the design of synthetic gene switches to enhance the safety and controllability of engineered cells. However, current small molecule-regulated synthetic gene switches face clinical limitations such as long-term side effects and metabolic disturbances. Here, we develop a natural sweetener psicose-inducible transgene expression (PURE) system based on the transcriptional repressor PsiR from Agrobacterium tumefaciens. We increase the induction sensitivity of PURE using computational docki

Targeting the DNA damage response prevents regrowth of colorectal peritoneal metastasis-derived organoids following treatment with mitomycin C.

BACKGROUND: A minority of Colorectal Cancer (CRC) patients with peritoneal metastases is eligible for cytoreductive surgery (CRS) followed by hyperthermic intraperitoneal chemotherapy (HIPEC). However, recurrence rates are high (~80%). We tested whether inhibitors of the DNA damage response (DDR) could prevent recurrence in an in vitro HIPEC model. METHODS: Peritoneal metastasis-derived organoids (PMDOs; n = 10) were treated with inhibitors of ATR (berzosertib, ceralasertib, elimusertib), CHK1 (rabusertib), and WEE1 (adavosertib) alone, and in combination with MMC, oxaliplatin, or irinotecan.

Peptidoglycan from Bifidobacterium adolescentis enhances IL-10 production in regulatory B cells to alleviate gut inflammation.

The mechanisms by which gut microbiota modulate host immune responses remain incompletely understood. Here, we screened Lactobacillus and Bifidobacterium strains isolated from healthy individuals to identify symbionts capable of suppressing gut inflammation. Among them, Bifidobacterium adolescentis (Bifi-94) induced IL-10 production in mononuclear cells in vitro. Oral administration of Bifi-94 to mice treated with dextran sulfate sodium attenuated weight loss and reduced colonic inflammation scores. In wild-type C57BL/6 mice, Bifi-94 increased IL-10 levels in colonic tissue homogenates without

Morphological and clinical predictive factors in appendiceal mucinous neoplasms: A retrospective analysis of 49 cases.

Appendiceal mucinous neoplasms (AMNs) represent an infrequent type of appendiceal tumour, characterised by their potential for aggressive biological behaviour and the possibility of developing peritoneal carcinomatosis. There is still uncertainty about which variables can predict the clinical evolution and recurrence risk of this disease. The primary objective of this study is to reevaluate cases of AMN diagnosed at our institution to identify morphological and clinical factors that may serve as predictors of recurrence. A retrospective study of 49 patients diagnosed with AMN at tertiary hospi

Cathelicidin-Ka, the first frog-derived TLR2 and TLR4 agonist, induces macrophage activation and promotes inflammation.

TLR-targeted immunotherapy represents a promising strategy for combating infectious diseases by initiating or enhancing protective antimicrobial immunity. Here, we identified the first frog-derived TLR2 and TLR4 agonist, Cathelicidin-Ka (Cath-Ka), from the skin of Kaloula pulchra. The presence of Cath-Ka significantly enhanced proliferation, cytokine production, polarization, chemotaxis, phagocytosis, and intracellular bacterial killing of macrophages and peritoneal cells by targeting TLR2 and TLR4, rather than other pattern recognition receptors, and subsequently activated the downstream MyD8

miR-let-7a suppresses gastric cancer peritoneal metastasis through IRS2-mediated M1 macrophage polarization and direct inhibition of gastric cancer cells.

Gastric cancer, the fifth most prevalent and lethal malignancy worldwide, represents a significant public health challenge that necessitates urgent intervention. Peritoneal metastasis accounts for the majority of recurrences and fatalities following curative surgery for advanced gastric cancer. This study demonstrated that miR-let-7a inhibits gastric cancer peritoneal metastasis through dual mechanisms. Firstly, it promotes the polarization of macrophages to the M1 phenotype by targeting the 3'UTR of IRS2 mRNA, resulting in reduced IRS2 expression and phosphorylation and consequent suppression

Potent dual-function of marine peptide N6NH2 and its D-enantiomer to combat MDR A. veronii infection in tilapia (GIFT, Oreochromis niloticus).

Tilapia (GIFT, Oreochromis niloticus) has been an economically important freshwater species in China because of its high-quality meat, high yield and high fertility, etc. At present, the high-density breeding industry has suffered great damage from Aeromonas veronii (A. veronii) infections, and the extensive use of antibiotics in aquaculture has resulted in the prevalence of antibiotic-resistant bacteria, resulting in huge economic losses. In this study, we investigated the antibacterial effect of a marine peptide-N6NH2 and its D-enantiomer (DN6NH2) on acute A. veronii infections of O. nilotic

Identification and characterization of broad-spectrum cathelicidin family antimicrobial peptides from the pygmy sperm whale Kogia breviceps.

Cathelicidins are crucial antimicrobial peptides (AMPs) that play a key role in the innate immunity of marine vertebrates. The pygmy sperm whale (Kogia breviceps), which resides in environments populated by diverse microbial communities, faces complex pathogenic threats; however, its cathelicidin family AMPs remain uncharacterized. In this study, we identified three novel cathelicidin peptides-Kb4-1, Kb4-2, and Kb5 from the whale (Kogia breviceps). All three peptides exhibited broad-spectrum and potent antibacterial activity, with Kb5, an amphipathic α-helical peptide, showing the strongest an

Methylglyoxal-modification of NLRP3 interrupts NLRP3-NEK7 interaction diminishing inflammasome activation and neuroinflammation.

BACKGROUND: The NLRP3 inflammasome is a supramolecular complex that mediates pyroptosis and the production of inflammatory cytokines. Aberrant activation of the NLRP3 inflammasome has been implicated in the pathogenesis of various diseases, including cancers, neurodegenerative disorders, metabolic diseases, and autoimmune conditions. Methylglyoxal (MGO), a highly reactive carbonyl compound derived from glycolysis, exerts unclear effects on NLRP3 inflammasome activation. This study aimed to clarify the impact of MGO on NLRP3 inflammasome activity and its underlying mechanism. METHODS: NLRP3 inf

The role of the α7 nicotinic acetylcholine receptor in promoting M2 macrophage polarization at inflammatory sites.

The α7 nicotinic acetylcholine receptor (α7nAChR) on macrophages exerts anti-inflammatory effects by suppressing the JAK/STAT and NF-κB pathways. Although the role of α7nAChR in immunoregulatory mechanisms in "individual" macrophages is established, studies on α7nAChR in an "overall population" of macrophages, including M1/M2 polarity, remain limited. Therefore, we examined the role of α7nAChR in M1/M2 polarity in inflammation. We generated peritonitis mouse models via LPS treatment and sterile intestinal manipulation in wild-type and α7nAChR-deficient mice. M1/M2 macrophage polarization was m
