Inflammatory responses following CRISPR modification of the nuclear localisation sequence in endogenous interleukin-1 alpha.

Interleukin (IL)-1α is a pro-inflammatory member of the IL-1 cytokine superfamily and is important for inflammatory responses to infection and injury. Unlike pro-IL-1β, pro-IL-1α is mainly localised to the nucleus upon expression. This is mediated by a nuclear localisation sequence (NLS) responsible for its importin-dependent transport into the nucleus. This nuclear localisation and the presence of histone acetyl transferase (HAT)-binding domains within the pro-domain suggest a role of this cytokine in gene transcription regulation. In addition, nuclear trafficking of pro-IL-1α is proposed to

Fluoroalkylated antimicrobial peptides enables cytosolic delivery and eradication of intracellular Staphylococcus aureus.

Intracellular bacteria survive are difficult to eradicate by conventional antibiotics. Antimicrobial peptides (AMPs) have garnered increasing attention due to their ability to exert broad-spectrum antibacterial effects through various mechanisms. However, they are also unable to effectively penetrate cell membranes to enter the cytoplasm. Fluorination has been proven to effectively enhance the ability of biopeptides to penetrate cell membranes. We designed a fluoroalkylated Omiganan (PFC-OMN) by conjugating a perfluorocarbon tag to a cysteine-terminated Omiganan (OMN) via a disulfide bond, ena

Broad synergistic effects of an AMP with multiple antibiotics against drug-resistant bacteria via multifaceted mechanisms.

Antimicrobial peptides (AMPs) have emerged as promising alternatives or adjuncts to antibiotics. However, most studies on AMP-antibiotic combinations focus on specific drugs or pathogens, with limited exploration of broad-spectrum synergistic effects. Herein, we demonstrate that CATHPb1, a snake-derived AMP, exhibits extensive synergistic antimicrobial activity when combined with 12 antibiotics representing six distinct mechanisms of action. Moreover, CATHPb1 shows a low potential for inducing resistance and significantly enhances anti-biofilm and anti-persister activities in combination with

A folate receptor-targeted NIR-II probe improves intraoperative imaging and surgery of metastatic ovarian cancer.

Intraoperative fluorescence imaging holds great promise for achieving complete cytoreduction in ovarian cancer, but its clinical utility is currently constrained by the limitations of existing imaging agents. In particular, the leading clinical probe, OTL38, operates in the near-infrared I (NIR-I; 700 to 1000 nanometers) window, which suffers from limited tissue penetration and suboptimal resolution. Here, a folate receptor α (FRα)-targeted NIR-II (1000 to 3000 nanometers) imaging probe was developed by incorporating a 4-(4-iodophenyl)butanoic acid moiety. This structural modification enables

Multiomic analysis of malignant ascites defines RORC-expressing dendritic cells at sites of metastasis.

Malignant ascites is caused by the peritoneal dissemination of cancer and has a poor prognosis. Here, we performed multiomic analyses on ascites samples from patients with cancer and identified RORC-expressing dendritic cells (RORC DCs) enriched in malignant ascites. RORC DCs expressed PIGR and were transcriptionally similar to murine RORγt+ DCs/Thetis cells implicated in immune tolerance. We devised a high-purity sorting strategy to enable in vitro experiments with patient-derived RORC DCs, which indicated that these cells were phenotypically stable and stimulated T cell proliferation while a

The potential of neutrophil gelatinase-associated lipocalin in management of acute kidney injury and peritoneal dialysis-related peritonitis: a narrative review.

Neutrophil gelatinase-associated lipocalin (NGAL) is a biomarker extensively studied in multiple diseases. While its application in chronic kidney disease (CKD) and kidney transplant patients is relatively limited, NGAL has shown significant promise in the early detection and diagnosis of acute kidney injury (AKI), which may improve more timely management and potentially better clinical outcomes. In addition, NGAL has demonstrated promising utility in identifying peritoneal dialysis-related peritonitis (PDRP) and monitoring the treatment response. This review aims to provide an in-depth overvi

An integrated network pharmacology and proteomics approach reveals the anti-fibrotic effect of Fushen Granule on peritoneal fibrosis.

BACKGROUND: Fushen Granule (FSG), a Chinese medicine formular, was used clinically to improve the efficiency of dialysis in end-stage renal disease patients receiving peritoneal dialysis treatment. However, the mechanisms of its antifibrotic effect on peritoneal fibrosis (PF) have not yet been studied. In this study, we aimed to identify the potential mechanism of FSG in intervening in PF. METHODS: Network pharmacology analysis and molecular docking were used to predict the related ingredients and potential targets of FSG in treating PF. TGF-β1 induced MeT5A cells were used for in vitro verifi

A rare fungal threat: Successful management of pan-drug resistant Microascus cirrosus peritonitis in a PD patient.

Microascus cirrosus is a rare environmental mold not previously genetically confirmed as a cause of peritoneal dialysis (PD)-associated peritonitis. We report the first culture- and multilocus sequencing-verified case in an immunocompetent PD patient who fulfilled ISPD diagnostic criteria, presenting with cloudy effluent and neutrophilic inflammation despite minimal systemic symptoms. Early clues included black intraluminal catheter debris and a positive galactomannan index in peritoneal dialysate. Definitive identification required ITS, LSU, SSU, and β-tubulin sequencing after initial morphol

Peritoneal dialysis-associated peritonitis caused by Brevundimonasaurantiaca.

Brevundimonas aurantiaca is an aerobic, non-fermenting Gram-negative bacillus traditionally regarded as an environmental organism within the genus Brevundimonas. Although this species had long been known microbiologically, human infection caused by B. aurantiaca was not reported until 2022, and peritoneal dialysis-associated peritonitis due to this organism had not been previously described. We report a case of peritoneal dialysis (PD)-associated peritonitis in a 55-year-old man with end-stage kidney disease receiving continuous ambulatory peritoneal dialysis. B. aurantiaca was isolated from t

Development and validation of a nomogram for peritoneal dialysis-associated peritonitis.

Peritoneal dialysis (PD)-associated peritonitis is a leading cause of technique failure. We aimed to develop and internally validate a Cox regression‑based nomogram for predicting peritonitis‑free survival in incident PD patients. This single-center retrospective cohort study included 372 patients who underwent peritoneal dialysis between June 2018 and December 2024, with follow-up through December 2025. LASSO‑Cox regression was used to select predictors from 23 candidate variables. A nomogram was constructed to predict 6‑, 12‑, and 24‑month peritonitis‑free survival. Model performance was ass
