AA in Obesity and Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) Nonalcoholic Fatty Liver Disease (NAFLD).

Obesity and MASLD (NAFLD) are chronic, low-grade systemic inflammatory diseases resulting from complex interactions among diet, adipose tissue, gut microbiota, liver, muscle, pancreas, and the brain. Central to this integrative framework are AA and other polyunsaturated fatty acids (PUFAs), which regulate inflammation, insulin sensitivity, neuroendocrine signaling, and energy homeostasis.Excess consumption of energy-dense foods rich in saturated and trans fats leads to adipocyte hypertrophy, altered membrane fluidity, increased antigenicity, and enhanced recruitment of macrophages and T cells

Integrative Transcriptomic Network Modeling Coupled with Patient-Derived Validation for Circulating lncRNA Biomarker Discovery in NAFLD: A Comprehensive Workflow.

.: Nonalcoholic fatty liver disease (NAFLD) is a significant global health concern, impacting roughly 25% of people and leading to chronic liver conditions. It involves excess fat accumulation in the liver without significant alcohol intake and can develop into nonalcoholic steatohepatitis (NASH), fibrosis, or cirrhosis. While liver biopsy remains the gold standard for diagnosis, its invasive nature and associated risks restrict its routine use. Noninvasive biomarkers, such as serum ALT, AST, and various composite scores, are available; however, their clinical usefulness is often limited by va

Hepatic GPR75 exacerbates MASH through GNAI2-dependent signaling.

BACKGROUND AND AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD), including its more severe form, metabolic dysfunction-associated steatohepatitis (MASH), is increasingly recognized as a critical global health challenge. This study investigates the role of hepatic GPR75 in MASH progression. APPROACH AND RESULTS: Although GPR75 is not abundantly expressed in the liver in healthy individuals, its protein levels significantly increase during MASH. Depletion of Gpr75 in either the whole liver or specifically in hepatocytes protected mice from diet-induced hepatic steatosis, w

Probiotic bacterial ghosts enhance oral astaxanthin delivery to alleviate non-alcoholic fatty liver.

Astaxanthin (AXT) is a promising natural bioactive compound for the nutritional intervention of non-alcoholic fatty liver (NAFL), however, its poor stability and limited oral bioavailability restrict its practical application. In this study, a novel probiotic-derived oral delivery system, designated AXT@LPGs, was developed by loading AXT into Lactiplantibacillus plantarum bacterial ghosts (LPGs) to enhance its efficacy against NAFL. In vivo experiments demonstrated that LPGs significantly improved the intestinal accumulation and retention of AXT. Pharmacokinetic evaluation further revealed tha

Apigenin Enhances Hepatic Lipophagy through Regulating Lipid Droplet Size via the AKT-FHL2/PPARγ-CIDEC Pathway.

Large lipid droplets (LDs) resist lipophagy-mediated degradation due to steric hindrance, contributing to hepatic lipid accumulation in nonalcoholic fatty liver disease (NAFLD). Apigenin (API), a flavonoid abundant in fruits and vegetables, improves NAFLD, but its mechanism remains unclear. In this study, we found that API notably reduced LD size, thereby facilitating subsequent lipophagy. Transcriptomic analysis and validation revealed that API downregulated cell death-inducing DFF45-like effector C (CIDEC), a key promoter of LD fusion. Mechanistically, API suppressed CIDEC transcription by i

P3-1 Alleviates NAFLD by Regulating the Liver-Gut Axis and PPARα/RXRα Pathway in Mice.

Plant-derived peptides show hepatoprotective biological functions. An octapeptide (Gly-Phe-Cys-Asp-Arg-Met-Pro-Leu), termed P3-1, was isolated from macadamia nuts for the first time. This study evaluated the ameliorative effect of P3-1 on nonalcoholic fatty liver disease (NAFLD) and clarified the molecular basis of this action. The results showed that P3-1 attenuated hepatic pathological damage, improved lipid metabolism and liver function indices, and enhanced antioxidant capacity in NAFLD mice. In addition, P3-1 enhanced intestinal barrier function, thereby improving hepatic metabolic functi

Engineering a Galactosylated Zein-Orientin Nanoplatform for NAFLD Alleviation via Lipid Metabolic Modulation and Gut Microbiota Remodeling.

Nonalcoholic fatty liver disease (NAFLD) is characterized by excessive hepatic lipid accumulation, and the existing pharmacological alternatives are constrained by considerable side effects and probable toxicity. Orientin (Ori), a natural flavonoid with hepatoprotective potential, is restricted by poor solubility and low bioavailability. Herein, a galactosylated zein nanoplatform (Ori@Gal-Zein) was engineered via the Maillard reaction, resulting in uniform spherical nanoparticles (163.3 ± 2.6 nm) with high loading capacity of Ori (7.89 ± 0.14%) and gastrointestinal stability. Hepatic-Renal Cascade Clearable Aminopeptidase N Probe for Rapid Detection of Early-Stage Liver Disease.

Early liver disease is a prolonged process with vague symptoms and evolving pathological causes, leading to a delayed diagnosis and poor therapeutic outcomes. The development of rapid and effective approaches for liver disease detection in both preclinical research and clinical diagnosis is in high demand yet remains challenging. Herein, we report an activatable fluorogenic probe (CyP-A) that is cleared via a hepatic-renal cascade for monitoring of hepatocyte injury and tissue fibrosis. After systemic administration, CyP-A preferentially accumulates in the liver, where it undergoes APN-mediate

The ability of FGF21 to ameliorate signs of non-alcoholic fatty liver disease in mice depends on sex and diet.

Consumption of high-calorie food leads to the development of obesity and non-alcoholic fatty liver disease (NAFLD). The pathogenesis of NAFLD depends on the composition of the diet. Administration of fibroblast growth factor 21 (FGF21) to obese mice improves metabolism and liver condition; however, the dependence of this effect on sex and diet composition has not been studied. Objective: To investigate the influence of sex on liver condition in mice with diet-induced obesity caused by different diets and on the ability of FGF21 to beneficially affect liver function. Obesity was induced in male

Diabetes duration modifies the association between AIP and MAFLD in type 2 diabetes.

BACKGROUND: Metabolic dysfunction-associated fatty liver disease (MAFLD) is highly prevalent in patients with type 2 diabetes mellitus (T2DM). Although several insulin resistance (IR)-related indices have been proposed as surrogate markers, their comparative performance and the influence of diabetes duration remain unclear. We compared the associations of eight IR-related indices with MAFLD and evaluated the modifying effect of diabetes duration. METHODS: This retrospective cross-sectional study included 5,996 patients with T2DM, of whom 3,087 had MAFLD. Logistic regression was used to evalua

Beyond Weight Loss: Rethinking Dietary and Lifestyle Interventions for Liver Disease Modification in MASLD.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is commonly managed with dietary and lifestyle intervention, and weight loss is often used as a key marker of treatment success. However, changes in body weight alone do not fully describe the effects of lifestyle intervention on liver disease. This narrative review asks when lifestyle-induced changes can support a claim of liver disease modification. Current guidance generally associates sustained weight loss of at least 5% with reduced liver fat, 7-10% with reduced liver inflammation, and at least 10% with improvement in fibros

Metabolic Dysfunction-Associated Fatty Liver Disease-Cardiovascular-Kidney-Metabolic (MAFLD-CKM): A Modified Framework to Predict Long-Term Mortality.

BACKGROUND Cardiovascular-kidney-metabolic (CKM) syndrome reframes cardio-renal-metabolic risk but omits hepatic dysfunction, particularly metabolic dysfunction-associated fatty liver disease (MAFLD). This study assesses whether integrating MAFLD into the CKM framework improves mortality risk discrimination compared with the original CKM framework. MATERIAL AND METHODS We used data of 10 335 adults aged 20 to 74 years from the third National Health and Nutrition Examination Survey (NHANES III) with mortality follow-up through 2019. CKM was classified based on the American Heart Association fra

Exosomal microRNAs as biomarkers of multimorbidity risk in lean and obese women with polycystic ovary syndrome.

Polycystic ovarian syndrome (PCOS) is a multifactorial endocrine disorder that is characterised by reproductive, metabolic and cardiometabolic abnormalities. Besides typical hormonal abnormalities, inter-organ communication has a crucial role in the development of PCOS-associated multimorbidity. PCOS causes early changes in the insulin signalling pathway, inflammatory pathways, androgen synthesis genes, and ovarian theca cell activity even before the appearance of regular symptoms such as irregular periods, polycystic ovaries, weight gain, and hyperandrogenism. Exosomes are tiny extracellular
