Integrating WGCNA and machine learning to identify and validate key biomarkers in MASLD.

BACKGROUND: Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) represents the most prevalent chronic liver disease worldwide. The absence of approved pharmacotherapies is largely attributed to their profound molecular heterogeneity. The identification of novel hub genes in MASLD is therefore critical for unraveling the complex molecular mechanisms driving disease pathogenesis and progression. METHODS: We employed an integrative systems-biology approach utilizing Weighted Gene Co-expression Network Analysis (WGCNA) followed by multi-algorithm machine learning (LASSO, Random Fores

Association of metabolic dysfunction-associated steatotic liver disease with coronary plaque vulnerability and prognosis in patients with acute coronary syndrome.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is increasingly recognized as a risk factor for cardiovascular events. However, MASLD's relationship with optical coherence tomography (OCT)-defined coronary plaque vulnerability in patients with acute coronary syndrome (ACS) remains unclear. Moreover, the mediating role of plaque vulnerability in the association between MASLD and major adverse cardiovascular and cerebrovascular events (MACCE) has rarely been investigated. Therefore, this study used OCT to examine the association between MASLD and coronary plaque vuln

Serum Biomarkers for the Non-Invasive Assessment of Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD): An Update.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD), formerly known as nonalcoholic fatty liver disease, represents the leading cause of chronic liver disease globally. The increasing prevalence of the metabolic syndrome has driven a parallel rise in MASLD-related morbidity and mortality. Accurate assessment of liver fibrosis remains crucial, as liver fibrosis stage is the strongest predictor of adverse clinical outcomes. Liver biopsy, considered the gold standard for assessing liver fibrosis, is invasive and has limitations, prompting the need for reliable noninvasive

Curcumin from Turmeric Alleviates Lipid Metabolism: Network Pharmacology and Cellular Validation.

Curcumin, the primary active compound in turmeric, a widely used medicinal and edible herb in traditional Chinese medicine, exhibits beneficial effects against lipid metabolic dysfunction (LMD). Nevertheless, its comprehensive regulatory network and precise mechanisms remain unclear. In this study, network pharmacology, molecular docking, and in vitro cellular assays were integrated to elucidate the targets and mechanisms by which curcumin acts against LMD. Seven hub targets were identified: AKT1, ALB, INS, IL6, TP53, TNF, and PPARG. Kyoto Encyclopedia of Genes and Genomes analysis revealed si

Long-term ambient benzene exposure increases incident NAFLD risk through accelerated biological aging.

The contribution of ambient air pollutants to non-alcoholic fatty liver disease (NAFLD) remains undefined. This study included 401,472 adults from the UK Biobank (UKB), with residential benzene estimated at 1-km resolution from the DEFRA model (2006-2020). After multivariable adjustment in the Cox models, each interquartile-range (0.151 μg/m3) increment in benzene was associated with a 26% higher risk of incident NAFLD (hazard ratio [HR] = 1.26; 95% confidence interval [CI], 1.21-1.30), following a nonlinear exposure-response relationship. Accelerated biological aging mediated 6.5% of this ass

Global epidemiological patterns and disease burden of metabolic dysfunction-associated steatotic liver disease, metabolic dysfunction and alcohol-associated liver disease, and alcohol-associated liver disease.

The 2023 multisociety Delphi consensus redefined the nomenclature for steatotic liver disease (SLD) by replacing nonalcoholic fatty liver disease with metabolic dysfunction-associated steatotic liver disease (MASLD) and introducing metabolic dysfunction and alcohol-associated liver disease (MetALD) and alcohol-associated liver disease (ALD). This revised framework is expected to enhance epidemiological surveillance, phenotyping of SLD, and public health interpretation. This review summarizes contemporary literature on the global epidemiology, disease burden, and comparative outcomes of MASLD,

Clinical outcomes of lean metabolic dysfunction-associated steatotic liver disease by phenotypic subtypes.

AIMS: Lean metabolic dysfunction-associated steatotic liver disease (MASLD) is often treated as a single entity despite substantial heterogeneity. We assessed whether body mass index (BMI) strata, cardio-metabolic risk factor (CMRF) phenotype, and CMRF burden stratify the risk of clinical outcomes. METHODS: Using the TriNetX Global Network, we identified adults with MASLD from 2005 to 2024. Lean MASLD was defined by BMI less than 25 kg/m2 and stratified into underweight and normal-weight groups. Underweight and normal-weight MASLD were compared after 1:1 propensity score matching. Within norm

Online chiral derivatization coupled with CE-MS for baseline separation of 16 proteinogenic amino acid enantiomers.

Amino acid (AA) enantiomers influence protein function through their distinct spatial structures, making their precise separation essential for elucidating physiological and pathological pathways. In this work, a novel method for the efficient separation of AA enantiomers based on on-line chiral derivatization with (S)-N-(4-nitrophenoxycarbonyl) phenylalanine methoxyethyl ester ((S)-NIFE) coupled with CE-MS has been developed. The method utilizes the ultrafast reaction kinetics of (S)-NIFE with AA enantiomers to facilitate their efficient separation. The online strategy eliminates time-consumi
