Renalase deficiency suppresses hepatic triglyceride accumulation in the progression to MASLD/MASH by GAN diet in male mice.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a growing public health concern characterized by hepatic triglyceride (TG) accumulation, inflammation, and fibrosis. Renalase is known for its role in blood pressure regulation and catecholamine metabolism, but recent evidence suggests broader cytokine-like functions. Moreover, its involvement in MASLD remains unclear. In this study, we examined the effects of renalase deficiency on hepatic lipid metabolism in a Gubra Amylin NASH (GAN) diet-induced MASLD model using renalase knockout (KO) mice. Our results show that renalase K

Lysosome-localized IRTKS condensates promote mTORC1 activity leading to MASLD and HCC.

Metabolic dysregulation has been recognized as a crucial driver of tumorigenesis, particularly in metabolic dysfunction-associated steatotic liver disease (MASLD)-related hepatocellular carcinoma (HCC). However, the underlying mechanisms remain poorly understood. Here, we identify lysosome-localized insulin receptor tyrosine kinase substrate (IRTKS) as a key activator of the metabolic master regulator mTORC1 through phospho-antibody array screening. IRTKS forms membrane-associated condensates that selectively interact with the GTPase RRAGD, a key upstream regulator of mTORC1, thereby enhancing

Design, Synthesis, and Biological Evaluation of Chromone Derivatives as STAT1 Inhibitors for Treatment of Nonalcoholic Steatohepatitis.

Nonalcoholic steatohepatitis (NASH) is a chronic liver disease with a high global incidence rate, intricately linked to metabolic disorders. It is characterized by steatosis, inflammation, and hepatocyte ballooning. STAT1 is a transcription factor involved in metabolic regulation and progression of inflammatory responses. In this study, we have identified STAT1 as a novel therapeutic target for NASH. Using our established STAT1 inhibitor screening platform, we identified the compound ZDZ-553, which is characterized by a chromone skeleton, demonstrates potent inhibitory activity against STAT1,

Metabolic dysfunction-associated steatotic liver disease (MASLD): definition, diagnosis, natural history, and treatment.

Non-alcoholic fatty liver disease (NAFLD) is one of the most prevalent causes of chronic liver disease, and strategies to prevent NAFLD-related hepatocellular carcinoma and liver failure are of increasing importance. Recently, the classification of steatotic liver disease (SLD) has been redefined based on alcohol intake: metabolic dysfunction-associated SLD (MASLD, mild alcohol intake), MASLD and increased alcohol intake (MetALD, moderate alcohol intake), and alcohol-related liver disease (ALD, heavy alcohol intake). Because the concordance rate between NAFLD and MASLD is 96-99%, MASLD can be

DNMT1 facilitates the progression of metabolic dysfunction-associated steatotic liver disease by impeding transcription mediated by HNF4α and PPARα.

BACKGROUND/AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most prevalent chronic liver disease worldwide. Aberrant DNA methylation, which is primarily maintained by DNA methyltransferase 1 (DNMT1), has been linked to metabolic dysregulation; however, its contribution to MASLD pathogenesis remains poorly defined. This study aimed to elucidate the role of DNMT1-mediated methylation in transcriptional regulation during MASLD progression and to determine whether DNMT1 inhibition can reverse disease-associated epigenetic and transcriptional alterations. METHODS: We c
