A novel arctigenin derivative attenuates nonalcoholic fatty liver disease by activating adiponectin receptor 1-mediated AMPK signaling pathway.

Nonalcoholic fatty liver disease (NAFLD), along with metabolic syndrome and obesity, has seen a dramatic rise in frequency around the world. Arctigenin (ATG) has better efficacy in the treatment of non-alcoholic fatty liver in vitro, but its application in vivo models is limited due to its low oral bioavailability and poor water solubility. Compound ARC-18 is a derivative of ATG. As a prodrug, it improves the bioavailability and solubility of ATG, so that ARC-18 can also have better pharmacological activity in oral administration. The main purpose of this article is to explore the therapeutic

Current Trends and Perspectives on Obesity and Metabolic Dysfunction-Associated Steatohepatitis in East Asia.

Obesity is a global epidemic that has rapidly increased in prevalence in East Asian countries. Complications of obesity, such as metabolic dysfunction-associated steatohepatitis (MASH), have increased in parallel with the recent rise in obesity. Rapidly aging populations, increasingly Westernized lifestyles, and a likelihood of developing higher-risk fat types (e.g., visceral adipose tissue) all play a role in the rising influence of obesity and MASH in East Asian populations. Additionally, East Asian patients are at high risk for developing lean MASH at lower body mass indexes than are common

Association of the C-reactive protein-triglyceride-glucose index with metabolic dysfunction-associated steatotic liver disease and long-term all-cause and cardiovascular mortality: evidence from two nationwide prospective cohort studies.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is closely linked to cardiometabolic disorders, and cardiovascular disease is the leading cause of death among affected individuals. Identifying simple biomarkers that capture metabolic-inflammatory burden and predict long-term mortality in MASLD remains a clinical priority. The C-reactive protein-triglyceride-glucose index (CTI) integrates inflammation, dyslipidaemia, and glycaemic status, but its relevance to MASLD and long-term mortality has not been fully elucidated. METHODS: We conducted a multi-stage investigat

Responsive Nanomolecular Probes for Evolving Liver Diseases: Technological Revolutions and Challenges.

Early diagnosis of liver diseases has long faced serious challenges such as insufficient sensitivity, low specificity and difficulty in revealing molecular lesion information by conventional imaging methods. With the rapid development of nanoscience, nanomolecular probes exhibit tuneable structure, modifiable surface and excellent biocompatibility, and their responsive properties have gradually become an ideal medium for connecting molecular pathological changes and image visualisation. This review reveals that functionalised nanoprobes responsive to disease microenvironment-specific signals (

Lacticaseibacillus rhamnosus B6 alleviates metabolic dysfunction-associated fatty liver disease by suppressing intestinal LPS synthesis and regulating lipid metabolism.

INTRODUCTION: Metabolic dysfunction-associated fatty liver disease (MAFLD) has become a global epidemic with an unclear etiology and no effective therapeutic options. Disruption of the gut-liver axis driven by intestinal dysbiosis is closely implicated in MAFLD pathogenesis, making gut microbiota-targeted probiotic interventions promising preventive strategies. METHODS: Lacticaseibacillus rhamnosus B6, a probiotic strain isolated from homemade Bulgarian fermented milk, synthesizes immunomodulatory macromolecules and regulates the intestinal flora. In the present study, we comprehensively inve

Does stigma reduce hazardous health behaviors contributing to steatotic liver disease?

Hazardous health behaviors increasingly contribute to end-stage liver diseases. To address surges in alcohol-associated liver disease and metabolic dysfunction-associated steatotic liver disease, clinicians in hepatology and liver transplantation must increasingly understand factors that influence eating and alcohol behaviors and consider ways to therapeutically influence patients' health choices. Stigma is a prominent influence on patient behavior, but is overused as a sweeping explanatory factor as to why difficulties arise, behavior change does not occur, or when outcomes are suboptimal. Wi

The Role of Butyrate in People with Metabolic Dysfunction-Associated Steatotic Liver Disease and Related Metabolic Comorbidities: A Systematic Review.

BACKGROUND: Metabolic dysfunction–associated steatotic liver disease (MASLD) is the most prevalent chronic liver disease worldwide and is strongly linked to obesity, type 2 diabetes, and cardiovascular disease. Growing evidence highlights the role of the gut–liver axis, particularly microbial metabolites such as the short-chain fatty acid (SCFA) butyrate, in MASLD pathophysiology. However, clinical data on butyrate levels and the abundance of butyrate-producing bacteria in MASLD patients remain inconsistent. OBJECTIVES: To systematically synthesize human evidence evaluating the associations be

Nutrient-driven histone acetylation underlies energy storage and mobilization.

In natural settings, energy storage and mobilization maintain a dynamic balance in response to recurrent overfeeding and fasting. Imbalanced energy storage and mobilization lead to a variety of metabolic dysfunctions. However, whether the metabolic status directly couples with epigenetic modifications and transcriptional outputs remains unclear. Here, we aimed to investigate the epigenetic mechanism underlying this adaptive balance and observed that, in an overfeeding state, increased glucose availability is associated with enhanced histone acetylation coinciding with acetyl-CoA production in

Preliminary insights into gut microbiome shifts as screening proxy for MASLD disease progression.

Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD), a metabolic syndrome with chronic excessive non-alcohol related triglyceride accumulation in liver cells, is characterised by a gradient of hepatic inflammation and fibrosis which lead to hepatocellular carcinoma. Clinical diagnosis is commonly based on non-invasive imaging methods, but definitive and conclusive diagnostic is achieved throughout invasive liver biopsy. Recent research pointed to an association between unbalanced gut microbiome and MASLD pathogenesis. In this prospective pilot study we dissect the gradual disease

Omega-3 fatty acids improve lipid metabolism by regulating miR-34a.

Lipid metabolism disorders and hepatic lipid deposition are major contributors to the development of metabolic diseases such as non-alcoholic fatty liver disease. Omega-3 fatty acids have been shown to exert beneficial effects on lipid homeostasis and liver function, but their molecular regulatory mechanisms remain incompletely understood. Here, we investigated the effects of omega-3 fatty acids on lipid metabolism and hepatic lipid deposition in addition to the modulatory role of miR-34a using an in vivo high-fat diet (HFD) model and an in vitro palmitic acid (PA)-induced HepG2 cell model. We

Network toxicology study and key target validation of chlorpyrifos-induced nonalcoholic fatty liver disease.

Chlorpyrifos (CPF) is a widely used insecticide known for its extended persistence in soil and water. It can induce non-alcoholic fatty liver disease (NAFLD) in mice. However, the specific molecular mechanisms responsible for CPF-induced NAFLD remain partially understood. Utilizing network toxicology analysis, we have identified TP53, HSP90AA1, AKT1, and JUN as pivotal driver genes orchestrating NAFLD progression triggered by CPF. A predictive nomogram constructed on these core genes demonstrates promising translational prospects for NAFLD. Gene-set enrichment analysis (GSEA) underscores the s

The interplay between exosomal miRNAs and cytokine networks in metabolic diseases.

Exosomal microRNAs (miRNAs) are increasingly recognized as central regulators of metabolic pathways, influencing glucose homeostasis, lipid metabolism, and inflammatory responses. Their ability to mediate inter-organ communication positions them as both biomarkers and therapeutic candidates for metabolic diseases such as type 2 diabetes, obesity, and non-alcoholic fatty liver disease (NAFLD). This review synthesizes recent literature on the mechanistic roles of exosomal miRNAs in metabolic regulation, their influence on insulin sensitivity and lipid homeostasis, and their interaction with cyto
