Hic-5 promotes the progression of nonalcoholic steatohepatitis by regulating hepatocellular fatty acid metabolism through the PTEN/PGE2/EP4 axis.

Nonalcoholic steatohepatitis (NASH) is a metabolic disease characterized by hepatic steatosis and inflammation among other features. Dysregulated lipid metabolism is crucial in the pathogenesis of NASH. However, its regulatory mechanisms remain intricate and poorly elucidated. Hepatic stellate cells (HSCs) have been reported to contribute to hepatocellular lipid metabolism dysregulation and aggravate NASH progression. However, the potential mechanisms remain unclear. Here, we demonstrate that hydrogen peroxide-inducible clone 5 (Hic-5), which is highly expressed in HSCs within the liver, is el

Molecular crosstalk between MASLD and IVDD revealed through integrated biomarker discovery analysis.

BACKGROUND: Non-alcoholic fatty liver disease (NAFLD), recently retermed as metabolic dysfunction-associated steatotic liver disease (MASLD), and intervertebral disc degeneration (IVDD) are major health burdens with rising prevalence. Despite affecting different organ systems, emerging evidence suggests potential molecular crosstalk between these conditions. However, the underlying mechanisms remain poorly understood. METHODS: We employed comprehensive bioinformatics analysis to investigate shared pathogenic mechanisms between MASLD and IVDD through integrated bulk and single-cell RNA sequenc

TFR2 p.A75V mutation aggravates liver iron overload in alcoholic liver disease via ERK pathway.

Liver iron overload, a contributing factor to liver damage, is frequently observed in advanced alcoholic liver disease (ALD). Genetic factors may influence the progression of ALD. This study explored the involvement of hemochromatosis-related genes in liver iron deposition among ALD patients. Clinical data from 97 ALD patients revealed that those with liver iron overload had worse liver function and prognosis. Among 43 patients who underwent exon sequencing of the HFE, HJV, HAMP, TFR2, and SLC40A1 genes, the TFR2 p.A75V mutation emerged as a potential contributor to iron overload. To further i

The Role of Crosstalk Between the Unfolded Protein Response and Autophagy in Diseases Associated with Sympathetic Nervous System Imbalance: Mechanisms and Therapeutic Perspectives.

Sympathetic nervous system (SNS) imbalance is a common pathological basis for cardiovascular diseases, non-alcoholic fatty liver disease, and diabetes. This review focuses on these diseases, analyzing two core mechanisms: excessive sympathetic excitation induced by endoplasmic reticulum stress (ERS) or autophagy dysfunction in key central nuclei (e.g., hypothalamus, rostral ventrolateral medulla); and ERS/autophagy abnormalities in peripheral target organs caused by chronic SNS overactivation. Existing studies confirm that chronic SNS overactivation promotes peripheral metabolic overload via s

Post-Translational Changes in Serum Albumin in Patients with Alcohol-Associated Hepatitis.

Post-translational modifications of human serum albumin (HSA) have been described in patients with liver disease. This prospective cohort study aimed to characterize HSA microheterogeneity in hospitalized patients with alcohol-associated hepatitis (AH) and investigate its clinical relevance. We analyzed HSA isoforms by mass spectrometry in 49 patients with AH (at admission and day 14) and 20 healthy controls. Survival at 30, 90, and 365 days was assessed. Differences in HSA isoform abundance were compared between controls and AH patients, as well as between 90-day survivors and non-survivors.

Liver-Pancreas Fat Deposition: Impact on Cardiometabolic Multimorbidity and Cardiac Dysfunction.

BACKGROUND & AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD) and pancreatic steatosis (PS) are interconnected ectopic fat conditions linked to cardiometabolic dysregulation. Their combined effect on the long-term risk of cardiometabolic multimorbidity (CMM; ≥ 2 of diabetes, hypertension, coronary heart disease, and stroke) and cardiac remodelling remains unclear. METHODS: We examined cross-sectional associations between PS and the severity of MASLD histology in a biopsy-proven MASLD cohort from China. Subsequently, using the UK Biobank, we assessed the long-term risk of

High-protein diets and metabolic dysfunction-associated steatotic liver disease: A double-edged sword in liver health.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is among the most prevalent chronic liver conditions globally and is closely linked with a range of metabolic disorders. Recently, the high-protein diet (HPD) has garnered attention for its potential benefits in weight management and metabolic health; however, its impact on MASLD remains a subject of debate. This article provided a systematic review of epidemiological studies, clinical trials, and foundational research concerning the role of HPD in MASLD. It examined the mechanisms by which HPD influences liver metabolism, inflam

GCN5 drives MASLD progression through LXRα/SREBP1c signaling pathway-mediated de novo lipogenesis.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a global health concern that affects nearly one-quarter of the world's population. General control non-repressed protein 5 (GCN5), a histone acetyltransferase (HAT), has been implicated in the progression of several diseases, but its role in MASLD remains unclear. Here, we provide the experimental evidence that progressive human and male murine MASLD is driven by GCN5, but not by p300/CREB binding protein associated factor (PCAF) activation. Hepatocyte-specific GCN5 overexpression accelerates MASLD progression, whereas its abl

FK506-binding protein-5 in high-fat diet-induced metabolic dysfunction-associated steatotic liver disease.

Источник пометил публикацию как отозванную

A high-fat diet (HFD) alters the gut microbiota (GM), impairs metabolic efficiency, and increases gut permeability and inflammation. Obesity and insulin resistance are associated with GM dysbiosis. The GM is strongly associated with metabolic disorders and fatty liver disease. The co-chaperone protein FK506-binding protein-5 (FKBP5) regulates several vital cellular processes. Although FKBP5 has been implicated in stress-related disorders, it has not been directly linked to HFD-induced metabolic fatty liver disease. This study aimed to elucidate how FK506 binding protein 5 impairment affects th

Antioxidant-rich and fibre-enhanced diets reduce liver fibrosis risk in metabolic dysfunction-associated steatotic liver disease: latent class analysis of a cohort study.

Dietary patterns are increasingly recognised as crucial factors influencing the progression of metabolic-associated steatotic liver disease (MASLD). This study aimed to explore the association between dietary patterns and MASLD risk among adults, using latent class analysis (LCA), to our knowledge, for the first time in this context. This cross-sectional study included 8549 adults from the general population. Dietary intake data were collected using a validated FFQ and analysed with Nutritionist IV software. A 3-step LCA was applied to determine distinct dietary patterns among participants usi

Lactobacillus rhamnosus-Derived Extracellular Vesicles Mitigate Nonalcoholic Fatty Liver Disease Progression Through Activation of the Apelin Signaling Pathway.

Non-alcoholic fatty liver disease (NAFLD), a chronic metabolic disorder characterized by ectopic lipid deposition in hepatocytes, currently lacks evidence-based pharmacotherapies, rendering probiotic-based strategies-particularly those utilizing Lactobacillus rhamnosus (LR)-a promising therapeutic avenue. Bacterial extracellular vesicles from Lactobacillus rhamnosus modulate NAFLD progression, but their molecular mechanisms remain unelucidated. Lactobacillus rhamnosus (LR-BEVs) and Escherichia coli (E-BEVs) were isolated via sequential ultracentrifugation, validated for morphology/size by TEM

MLKL in liver parenchymal cells promotes liver cancer in murine metabolic dysfunction-associated steatotic liver disease.

The rising prevalence of hepatocellular carcinoma (HCC) in the last decade is mostly attributable to the growing epidemic of metabolic dysfunction-associated steatotic liver diseases (MASLD). However, the transition from steatosis to steatohepatitis (MASH) and ultimately to HCC is not fully understood. As an executioner protein of necroptosis, the mixed-lineage kinase domain-like protein (MLKL) has been proposed to contribute to MASH and HCC development. To investigate its role in disease progression, mice whose liver parenchymal cells (LPCs) no longer expressed MLKL (MlklLPC-KO) were compared

Pathogenesis and intervention strategies for metabolic dysfunction-associated fatty liver disease from the perspective of the gut-microbiota-liver axis.

Trillions of microorganisms in the human gut are important regulators of health, and the gut and liver have a symbiotic relationship with them. The study found that there is bidirectional communication of substances and signals between the gut and liver, and the gut microbiota is an important medium for mediating bidirectional communication in the gut-liver axis. During metabolic dysfunction-associated fatty liver disease (MAFLD) development, the gut microbiota and its metabolites change to different degrees and affect MAFLD pathogenesis through the gut-liver axis. However, the bidirectional c

CTRP12 in cardiovascular and metabolic diseases: current status and future perspective.

Cardiovascular and metabolic diseases severely threat human health with their management being a global challenge. C1q tumor necrosis factor-related protein 12 (CTRP12), a newly identified adipokine and paralog of adiponectin, is expressed predominantly by adipose tissue. As a secretory protein, full-length CTRP12 can be cleaved by furin to produce a globular isoform, which is the major form in the blood stream. CTRP12 has anti-inflammatory and glucose-lowering properties and also participates in the regulation of lipid metabolism, oxidative stress and cell apoptosis. Dysregulation of CTRP12 h

Efficacy and Safety of Statins in MASLD and Other Chronic Liver Diseases.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most common liver disease worldwide, with an estimated global prevalence of 38% in adults. MASLD confers a significant increase in morbidity and mortality due to its association with cardiovascular disease and progressive liver disease, including cirrhosis and hepatocellular carcinoma. Current treatment paradigms for MASLD are centred around lifestyle modification and weight loss, with a need for pharmacotherapeutic options. Given the strong relationship between MASLD and cardiovascular disease, there is an interest in eva
