Polystyrene nanoplastics potentiate cadmium-induced pyroptosis and apoptosis in primary duck hepatocytes: differential responses to N-acetylcysteine.

INTRODUCTION: Nanoplastics (NPs) can act as carriers for heavy metals, and co-exposure to NPs and co-pollutants may produce greater toxicity than single exposure. However, the combined toxicity of nanoplastics and heavy metals in poultry liver cells remains poorly understood. METHODS: Primary duck embryonic hepatocytes (PDEH) were used as an in vitro model. Cells were exposed to polystyrene nanoplastics (PS-NPs, 80 nm, 10 μg/mL) and/or cadmium chloride (CdCl2, 4 μg/mL) for 12 h. Pyroptosis- and apoptosis-associated endpoints were assessed by brightfield microscopy, PI staining, SEM, flow cyto

Discontinuing Immune Checkpoint Inhibitor-Based Therapy After Complete Response in Unresectable Hepatocellular Carcinoma: Evidence, Bias, and a Research Framework.

Immune checkpoint inhibitor (ICI)-based regimens can induce radiographic complete response (CR) in a small but clinically important subset of patients with unresectable hepatocellular carcinoma (HCC). Whether treatment can then be stopped safely remains unresolved. Evidence comes predominantly from retrospective cohorts and post hoc analyses: selected complete responders have durable disease control after cessation, but neither patient-selection criteria nor an optimal post-CR treatment duration has been validated. This structured critical narrative review distinguishes direct evidence in HCC

When to Operate After Neoadjuvant Therapy for Borderline Resectable Pancreatic Cancer: A Critical Review and Multidimensional Decision Framework.

BACKGROUND: Surgical timing after neoadjuvant therapy for borderline resectable pancreatic cancer remains difficult because post-treatment imaging may not reliably distinguish residual tumor from fibrosis, whereas tumor biology and patient fitness strongly influence surgical benefit. METHODS: We performed a critical narrative review of studies and guidelines addressing surgical timing, radiological response, tumor-marker dynamics, pathological response, and physiological readiness after neoadjuvant therapy for borderline resectable pancreatic cancer. RESULTS: Current evidence supports a mult

Hesperidin-Loaded Chitosan Nanoparticles Restore Hepatic Homeostasis by Targeting Nrf2/HO-1 and NF-κB/p53 Signaling in Malathion-Exposed Rats.

The present study investigated the hepatoprotective effects of hesperidin (HES) encapsulated in chitosan nanoparticles (HES-CNPs) against Malathion (MAL)-induced liver toxicity in rats. Ninety male Wistar rats were randomly divided into six groups: a control group, groups treated with HES (100 mg/kg BW) or HES-CNPs (100 mg/kg BW), a MAL-exposed group (27 mg/kg BW), and two combination groups receiving MAL (27 mg/kg BW) together with either HES or HES-CNPs (100 mg/kg BW) for four consecutive weeks. MAL administration induced profound biochemical and molecular alterations in rats. It significant

Localised Autophagy Inhibition by Nanodiamonds Potentiates Arsenic Therapy With Favourable Safety Profile in Solid Tumours.

Arsenic trioxide (ATO) shows limited efficacy against solid tumours, largely because it induces protective autophagy that attenuates its pro-apoptotic activity. Our previous studies established that nanodiamonds (NDs) function as nanoparticle autophagy inhibitors (NAPIs) when delivered systemically, markedly enhancing ATO efficacy in orthotopic liver tumour models by blocking NUPR1-mediated autolysosomal clearance. However, systemic administration remains inefficient in modulating the local autophagic microenvironment within tumours. Here we developed an interventional strategy based on intrat

Radiobiological comparison of holmium-166 and yttrium-90 for hepatocellular carcinoma treatment in vitro.

Radioembolization for hepatocellular carcinoma uses microspheres radiolabeled with yttrium-90 or holmium-166, yet their radiobiological effectiveness has not been directly compared, leaving radionuclide selection uninformed by biological considerations. We assessed the survival and DNA damage induction following 1.5 or 2.5 Gy of [ 166 Ho]Ho-DOTA or [ 90 Y]Y-DOTA delivered over 24 h on three human hepatocellular carcinoma cell lines. Both radionuclides led to a similar, dose-dependent reduction of survival in all cell lines, with a modest but nonsignificant trend towards increased toxicity for

Per- and polyfluoroalkyl substances exposure and risk of liver cancer: A comprehensive review of epidemiological and mechanistic evidence.

Per- and polyfluoroalkyl substances (PFASs), such as perfluorooctanoic acid and perfluorooctane sulfonic acid, are synthetic compounds that are persistent in the environment and widely used in industrial and consumer products. Their bioaccumulative potential and pervasive human exposure have raised serious concerns about hepatotoxicity and carcinogenicity. This review thoroughly examines current epidemiological and experimental evidence linking PFASs exposure to an increased risk of liver cancer, particularly hepatocellular carcinoma. Epidemiological studies show higher PFASs levels in hepatoc

A dual-function guanidinium scaffold for copper sequestration and redox protection in Wilson disease models.

Wilson disease (WD) is caused due to mutations in the copper ATPase gene ATP7B, resulting in accumulation of copper and the consequent disruption of cellular redox balance through reactive oxygen species generation. Current therapies mainly depend on copper chelation to lower metal burden which sometimes also strip copper from cuproproteins and disturb key physiological copper-dependent processes. It also does not directly suppress pathological copper reactivity i.e., free radical generation, a major driver of WD progression. To overcome these limitations, we have rationally designed Gua-Cu-3,

HCE6-mediated photodynamic therapy induces hepatocellular carcinoma cell death via inhibition of autophagic flux.

Porphyrin derivatives are potential candidates for photodynamic therapy against cancer. In this study, hydrophilic chlorin e6 trisodium salt (HCE6) was selected for treatment of hepatocellular carcinoma (HCC). SK-hep1 cells and a xenograft mouse model were used to investigate the antitumor efficacy and the autophagic mechanism of cell death. Time and concentration dependent cellular uptake of HCE6 indicated that HCE6-PDT can significantly induce death of SK-hep1 cells without notable toxicity. In addition, the treatment suppressed cell migration and colony formation and arrested cell cycle pro

Experimental models of hepatic injury in Wilson disease: stage-specific mechanisms and translational applications.

BACKGROUND: Wilson disease (WD) is an autosomal recessive disorder of copper metabolism caused by pathogenic variants in ATP7B, with the liver being one of the earliest and most prominently affected organs. ATP7B dysfunction impairs copper incorporation into ceruloplasmin and biliary copper excretion, resulting in progressive hepatic copper accumulation and a cascade of pathological responses, including proteostasis disturbance, mitochondrial dysfunction, oxidative stress, metabolic remodeling, inflammation, and fibrogenesis. Given the dynamic progression of WD-associated liver injury from ear

Safety, Tolerability and Antiviral Activity of GST-HG131 in Chronic Hepatitis B: A Randomized, Double-Blind Phase IIa Study.

BACKGROUND AND AIMS: Chronic hepatitis B (CHB) remains a major global health burden. Current therapies rarely achieve a functional cure due to low rates of HBsAg clearance, the persistence of the viral replication template-covalently closed circular DNA (cccDNA)-and the need for lifelong treatment. GST-HG131 is a first-in-class, orally bioavailable small-molecule inhibitor of PAPD5/7, key host factors involved in HBV mRNA stabilization and HBsAg production. We evaluated the safety, tolerability and antiviral activity of GST-HG131 in patients with CHB receiving nucleos(t)ide analogue (NUC) ther

Somatic burden versus silent toxicity: Distinct longitudinal pathways linking gastric and hepatic diseases to depression.

BACKGROUND: We still don't know much about how diseases of specific digestive organs might lead to depression. We hypothesized that gastric and hepatic diseases contribute to depression through distinct longitudinal pathways due to their biological differences. METHODS: We analyzed five waves (2011-2020) of the China Health and Retirement Longitudinal Study (CHARLS), including 24,065 adults aged ≥ 45 years. Participants were classified into control, stomach disease, liver disease, and comorbid groups. We used linear mixed-effects models to examine longitudinal associations with depressive sym

Alcohol consumption: from biochemistry to behavior, health, and beyond.

Alcohol metabolism is a multi-step biochemical process driven primarily by hepatic oxidation of ethanol to acetaldehyde and then to acetate. Although this pathway protects the body from prolonged ethanol exposure, transient acetaldehyde accumulation generates substantial toxicity, and chronic alcohol intake activates auxiliary pathways that amplify oxidative stress, inflammation, and tissue injury. Both metabolic capacity and physiological response to alcohol vary widely between individuals, reflecting genetic and epigenetic variation in the alcohol- and aldehyde-dehydrogenase enzymes, the cyt

Lacticaseibacillus paracasei CM6 postbiotics alleviate SO/SP-induced hepatic lipotoxicity in HepG2 cells: Insights into lipid-redox homeostasis, glutamate metabolism and PLA2G4A-dependent lysophospholipid remodeling.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is characterized by metabolic imbalances and lipid-induced cellular toxicity. Although postbiotics are emerging as reliable functional food ingredients for metabolic disorder management, molecular mechanisms underlying their hepatoprotective effects remain incompletely elucidated. In this study, we integrated microbial whole-genome sequencing with paired intracellular and extracellular metabolomic profiling to explore the regulatory effects of the Lacticaseibacillus paracasei CM6 postbiotic against sodium oleate/sodium palmitate

Drug Delivery Systems for Hepatocellular Carcinoma: Bridging Pathophysiological Barriers and Advanced Nanomedicine Strategies.

Hepatocellular carcinoma (HCC) is characterized by high heterogeneity, a complex tumor microenvironment, and multiple physiological barriers that limit conventional therapies and cause significant toxicity. Drug delivery systems (DDS), particularly nanoscale delivery systems, offer promising opportunities to overcome these obstacles through passive targeting, active targeting, and stimuli-responsive drug release. Nevertheless, despite substantial progress in preclinical research, the clinical translation of most DDS remains challenging. This review systematically outlines the pathophysiologica
