Phyllanthus niruri potentiates apoptosis in Huh-7 cells and mitigates diethylnitrosamine-induced hepatocarcinogenesis via Hippo-YAP pathway modulation.

Hepatocellular carcinoma is one of the deadliest cancers occurring worldwide. Substantial evidence suggests that the Hippo-YAP pathway as a promising therapeutic target. Phyllanthus niruri is known for pharmacological activities, including hepatoprotective, anti-viral, and anti-inflammatory effects. However, there is a lacuna of molecular studies on how the bioactives in Phyllanthus niruri interacts with hippo signaling to enhance its potential for liver cancer management. This study aimed to delineate the effects of Phyllanthus niruri extract on cell proliferation and apoptosis, and its mecha

Atorvastatin regulates hepatic transcriptome PXR dependently but distinct from pregnenolone 16α-carbonitrile and does not induce PXR-mediated liver steatosis.

Activation of the pregnane X receptor (PXR) by various drugs promotes liver steatosis. Yet, atorvastatin, a widely prescribed statin and a PXR ligand, is considered hepatically safe, even in patients with metabolic dysfunction-associated steatotic liver disease. To reveal mechanistic differences between atorvastatin and steatosis-promoting PXR ligands we investigated whether atorvastatin elicits distinct transcriptional and epigenomic responses, and whether these underlie its favorable hepatic safety. Mice were treated orally for four days with atorvastatin, pregnenolone 16α-carbonitrile (PCN;

Perfluorooctane sulfonates drives colitis via a gut microbiota-bile acid-endoplasmic reticulum stress axis in mice: Mechanistic validation and targeted interventions.

Perfluorooctane sulfonate (PFOS), a widespread environmental pollutant, is implicated in systemic toxicity, yet its role in colitis remains unclear. This study aimed to investigate whether PFOS exacerbates colitis via the gut microbiota-bile acid-endoplasmic reticulum stress (ERS) axis and to explore potential interventions. Following 15-week oral PFOS exposure (0.1 or 0.3 mg/kg/d), mice developed dose-dependent colitis, featuring weight loss, colon shortening, barrier dysfunction, and elevated inflammation. High-dose PFOS disturbed bile acid homeostasis, depleting conjugated species like taur

Efficacy and safety of first-line EGFR-TKI combined with chemotherapy versus EGFR-TKI monotherapy in patients with EGFR-mutant non-small cell lung cancer and liver metastases: A propensity score-matched analysis.

INTRODUCTION: Liver metastasis (LM) confers a poor prognosis in patients with epidermal growth factor receptor (EGFR)-mutant non-small cell lung cancer (NSCLC). Although EGFR tyrosine kinase inhibitors (TKIs) are the standard first-line treatment, the optimal approach for this high-risk subgroup remains uncertain. This study compared the efficacy and safety of EGFR-TKI monotherapy versus EGFR-TKI combined with chemotherapy in EGFR-mutant NSCLC patients with LM. PATIENTS AND METHODS: We retrospectively analyzed 98 patients with stage IV EGFR-mutant NSCLC and LM treated between 2020 and 2025. F

Entinostat suppresses hepatocellular carcinoma metastasis by upregulating AZGP1 through histone acetylation.

Hepatocellular carcinoma (HCC) has a high mortality rate, primarily driven by metastasis. The role of the histone deacetylase inhibitor (HDACi) entinostat in this process remains controversial, limiting its clinical application. This study aims to define entinostat's function and mechanism in HCC metastasis. We employed in vitro models, transcriptomic sequencing, chromatin immunoprecipitation-qPCR, and an orthotopic mouse model to assess the effects of entinostat on epithelial-mesenchymal transition (EMT), invasion, and tumor growth. Our findings demonstrate that entinostat potently prevented

Targeting metabolic vulnerabilities: REV-ERB agonist SR9009 potentiates sorafenib efficacy in liver cancer.

Hepatocellular carcinoma (HCC) is one of the most common cancers and the third leading cause of cancer-related death worldwide. The prognosis is poor, with a median survival of 12-15 months in patients with advanced-stage disease. Early diagnosis and the development of new, more effective therapeutic strategies are needed to address the challenges posed by this malignancy. Although immune checkpoint inhibitors have replaced multikinase inhibitors as first-line therapy, sorafenib continues to represent a valuable option for patients with contraindications to newer treatments. Based on genome-wi

Engineered CAR-NKT Extracellular Vesicles Suppress Tumor Progression and Enhance Antitumor Immunity.

Chimeric antigen receptor-engineered natural killer T (CAR-NKT) cell therapy represents a promising and innovative strategy in cancer immunotherapy, but is limited by acute toxicity and adverse effects, restricting broader clinical application despite durable responses. In this study, a novel nanobody targeting TM4SF1 is developed, which replaced the conventional single-chain variable fragment (scFv) in the design of CARTM4SF1-NKT cells. Moreover, CARTM4SF1-extracellular vesicles (EVs) therapy as an optimized alternative to direct CAR-NKT cell administration is introduced. Compared with conven

Seco-neokadsuranic acid A antagonizes SH3BP1 to suppress hepatocellular carcinoma progression through PPAR pathway activation.

BACKGROUND: The high mortality of hepatocellular carcinoma (HCC), driven by metastasis and drug resistance, underscores the need for novel therapeutics. The natural triterpenoid seco-neokadsuranic acid A (SA) shows promising antitumor activity; however, its precise molecular targets and mechanisms of action in HCC remain unclear, impeding its clinical development. PURPOSE: To interrogate the direct target engagement of SA and establish its mechanistic basis in suppressing HCC metastasis and remodeling antitumor immunity. METHODS: An integrated multi-level investigative strategy was utilized.

High copper levels induce oxidative stress and inflammatory processes in a cell culture model of Wilson's disease.

Wilson’s disease (WD) is a genetic disorder resulting from mutations in the ATP7B gene that lead to copper overload in hepatocytes. To gain a deeper understanding of the cellular mechanisms underlying WD, a reanalysis of the GSE1073236 dataset was conducted and validated by comparative analyses in a cell culture model of HepG2 and HepG2 ATP7B knockout (ATP7B-KO) cells, as well as in primary hepatocytes from WD patients and controls. Increased expression levels of genes associated with autophagy, oxidative stress and inflammation were observed in copper-treated HepG2 ATP7B-KO cells and in prima

Nuclear receptors-mediated lipid metabolism dysregulation: A potential mechanism linking PFOA/PFOS to hepatocellular carcinoma.

Hepatocellular carcinoma (HCC), a prevalent liver malignancy, is closely associated with dysregulated lipid metabolism. Endocrine disrupting chemicals (EDCs) can bind to nuclear receptors (NRs) and potentially induce carcinogenesis, but their specific influence on HCC progression remains unclear. To investigate this relationship, we combined bioinformatic analyses with experimental validation in the present study. Differential expression analysis of public HCC datasets (GSE14323, GSE17548, and GSE25097) identified candidate genes, which were further refined via weighted gene co-expression netw
