Therapeutic potential of β-sitosterol in methotrexate-induced liver injury: association with STING and ERK-1 pathways.

BACKGROUND: Methotrexate (MTX) is a commonly used drug to treat various cancers and autoimmune disorders, but its clinical utility is often limited by hepatotoxicity. β-sitosterol is a bioactive phytosterol compound that is naturally present in various plant foods and exhibits multiple antioxidant, anticancer and immunomodulatory activities. It has not been fully explored for its potential protective effects against liver injury. This study investigated whether β-sitosterol is associated with reduced hepatic injury in a short-term MTX model. METHODS: The rats were arbitrarily assigned to thre

Targeting fatty acid metabolism in hepatic macrophages mitigates acetaminophen-induced liver injury and promotes regeneration in mice.

BACKGROUND: An overdose of acetaminophen (APAP) can cause severe liver injury and may even lead to death. However, the mechanisms that underlie APAP-induced liver injury (AILI) are not fully understood. The extensive liver necrosis resulting from APAP overdose aggravates the sterile inflammatory response by activating resident macrophages and recruiting various immune cells. Numerous studies have highlighted the crucial role of lipid metabolism in macrophage function. This study investigated fatty acid metabolism in hepatic macrophages and its impact on the progression of AILI. METHODS: We ad

Protective Effect and Mechanism of Rosiglitazone in α-amanitin-induced Hepatotoxicity Via Activation of PPAR-γ/Nrf2 Signaling Pathway.

α-amanitin (α-AMA), the primary lethal toxin of amanita, primarily targets the liver with a high toxicity and a low lethal dose. As the precise mechanism of intoxication is unclear, and specific antidotes are lacking, α-AMA-induced liver injury has a high mortality rate. As a selective PPAR-γ agonist, Rosiglitazone (RSG) alleviates liver injury by upregulating the PPAR-γ/Nrf2 signaling pathway, thereby enhancing antioxidant effects, mitigating inflammation, and reducing apoptosis. This study investigated the protective role and mechanism of action of RSG in α-AMA-induced acute liver injury in

Oxaliplatin-Induced Liver Toxicity: Hepatic Transglutaminase 7 Upregulation Associates with Oxidative Stress, Inflammation, and Apoptosis.

Transglutaminases (TGs) are calcium-dependent enzymes that cross-link proteins, contributing to apoptosis, extracellular matrix (ECM) stabilization, and inflammation. While TG2 has been extensively studied in hepatic injury, the role of TG7 in oxaliplatin-induced liver responses remains unclear. Oxaliplatin, a third-generation platinum chemotherapeutic, effectively treats solid tumors but can induce hepatic stress through oxidative and pro-inflammatory signaling. Adult rats received intraperitoneal oxaliplatin (10 mg/kg weekly) for 6 weeks. qRT-PCR, immunohistochemistry (IHC), immunofluorescen

Liver Necrosis in a Patient Receiving Isavuconazole: Making the Case for Therapeutic Drug Monitoring.

An 81-year-old man prescribed isavuconazole for pulmonary fungal infection developed high serum transaminase levels and biopsy-confirmed hepatic necrosis associated with prolonged high serum isavuconazole levels. Genotyping revealed poor CYP3A5 activity/normal CYP3A4 activity. Older age and slow isavuconazole elimination likely led to isavuconazole-associated hepatic toxicity. This case underscores the importance of isavuconazole therapeutic drug monitoring.
