Network Pharmacology-Based Prediction and In Vivo Validation of Fumonisin B1-Induced Liver Injury and Early Fibrosis.

Fumonisin B1 (FB1) is a widespread mycotoxin contaminating food and feed, exhibiting severe hepatotoxicity. This study aimed to systematically investigate the underlying molecular mechanisms of FB1-induced liver injury and early liver fibrosis by combining network pharmacology with an in vivo animal model. First, network pharmacology and molecular docking were utilized to screen the core targets and key signaling pathways of FB1-induced hepatotoxicity. Subsequently, an in vivo C57BL/6 mouse model was established via intragastric administration of FB1 (5 mg/kg/day for 28 days). Serum levels of

Integrative metabolomics and malonyl-proteomics reveal the crosstalk between metabolic reprogramming and lysine malonylation in liver fibrosis.

Lysine malonylation (Kmal), a recently identified post-translational modification (PTM) of proteins, has been linked to energy metabolism and implicated in various pathological disorders, including liver diseases. However, the specific roles of Kmal in hepatic fibrosis remain enigmatic. This study aims to investigate novel post-translational modifications and the underlying mechanisms involved in liver fibrosis. Utilizing untargeted metabolomics on carbon tetrachloride (CCl4)-induced mouse fibrotic liver tissues, we observed significant metabolic dysregulation during the liver fibrotic process

Hepatocyte CD146 and soluble CD146 promote hepatic stellate cell activation via the integrin αvβ1-p38 MAPK signaling axis to drive liver fibrosis.

During chronic liver injury, hepatocytes promote liver fibrosis by interacting with hepatic stellate cells (HSCs). However, the key intracellular factors that regulate intercellular communication in hepatocytes remain poorly understood. Although cluster of differentiation 146 (CD146) is predominantly recognized as an endothelial cell marker, our study found that CD146 is substantially upregulated in hepatocytes under fibrotic stress and may play an independent role in transcellular signaling. This study aimed to elucidate the functions and underlying mechanisms of hepatocyte CD146 in liver fib

The effect of losartan nanoparticles vs losartan on TIMP1 and α-SMA immunohistochemical and gene expression in carbon tetrachloride-induced liver fibrosis.

Liver fibrosis is characterized by scar formation as a response to injury. Although many drugs have demonstrated antifibrotic efficacy in vitro and in animal models, effectively translating these into effective clinical therapies remains a challenge. Nanotechnology offers potential therapeutic approaches for treating liver fibrosis. This study aimed to assess and compare the antifibrotic effects of losartan and losartan nanoparticles (LP-NPs) on CCl4-induced liver fibrosis in rats.Thirty-two adult male Sprague-Dawley rats were divided into four groups: a control group, a CCl4-induced fibrosis

Integrated Network toxicology and Transcriptomics Uncover that the Involvement of NF-κB/NLRP3 Pyroptosis Axis in Mediating BaP-Induced Hepatotoxicity and Fibrosis.

Benzo[a]pyrene (BaP), a ubiquitous environmental pollutant, exerts hepatotoxicity primarily through its metabolite BPDE. This study combined network toxicology and experimental validation to delineate the mechanisms of BaP-induced liver injury and fibrosis. Network analysis identified ICAM-1 as a potential core target, and previous clinical data confirmed its upregulation in liver injury patients. In a rat model, BaP exposure induced dose-dependent hepatic injury and oxidative stress. Mechanistically, BaP activated the NF-κB/TNF-α signaling axis, upregulating ICAM-1 and initiating NLRP3 inflam

Nrf2 contributes to the protective effect of iron overload on thioacetamide-induced chronic liver injury in rats.

Chronic liver disease (CLD) is a global health concern that progresses to liver cirrhosis and cancer. This progression can be partly replicated in rodents through experimental administration of thioacetamide (TAA). Hepatic iron accumulation is a relatively common finding in CLD patients, as excess intracellular iron promotes the progression of CLD by generating reactive oxygen species. We previously reported that dietary iron overload abrogates TAA-induced liver cirrhosis in rats, raising a possibility that hepatic iron accumulation exerts a cytoprotective function. Here we investigated the ro

LC2767 alleviates LPS-induced liver injury and fibrosis by regulating FXR expression, NF-κB activation, and gut microbiota.

LC2767, a mix of Lactiplantibacillus plantarum LC27 and Bifidobacterium longum LC67, alleviates high-fat diet- or alcohol-induced liver injury and steatosis in mice. In the present study, we examined the action mechanism of LC2767 on lipopolysaccharide (LPS)-, LPS/D-galactosamine (LGc)-, or LPS/thioacetamide (LTc)-induced liver damage and fibrosis in mice. Short-termly or long-termly intraperitoneal injection of LPS, LGc, or LTc caused liver damage and fibrosis in mice: they increased alanine transaminase (ALT), aspartate transaminase (AST), and γ-glutamyl transpeptidase (γ-GTP) in the blood a

Plasma Adsorption Perfusion (BR-350) Versus Open Albumin Dialysis (OPAL) for Hyperbilirubinemia in Hepatic Failure in Cirrhotic Patients.

BACKGROUND: In the last decades, various extracorporeal liver support systems were developed for hepatic failure with hyperbilirubinemia with the aim to clear the blood from protein-bound toxic molecules. Open albumin dialysis (OPAL) is a complex and expensive system that requires addition of human albumin. Plasma adsorption perfusion (PAP) using the anion exchange resin adsorber, BR-350, is an alternative liver support method that does not need additional blood products and is more cost-effective and less time-consuming. METHODS: At the University Hospital Essen, PAP using BR-350 was perform

Features of the Therapy of Liver Cirrhosis in Aged Male and Female Rats.

Sex differences in the development of toxic liver cirrhosis in 12-month-old Wistar rats and the specific features of liver cirrhosis therapy with bovhyaluronidase azoximer were studied. The severity of pathological changes assessed by ultrasound, histological, biochemical, and hematological methods in male rats with liver cirrhosis exceeded that in female rats. The studied drug demonstrated anti-inflammatory and antifibrotic activity in aged rats with toxic liver cirrhosis, and its effect did not depend on the sex. Sex-related mechanisms may underlie the positive changes on liver cirrhosis und

PFPeA exposure drives hepatoxicity and liver fibrosis via oxidative stress/Wnt5a-induced hepatocyte senescence.

Perfluoropentanoic acid (PFPeA), one of short-chain PFASs, has raised concern due to existence in the environment. The hepatotoxicity of PFPeA remains to be further elucidated. Cellular senescence, a distinct cellular state that occurs under stress conditions, plays a vital role in the progression of various diseases. However, the contribution of cellular senescence to PFPeA induced liver toxicity remains unclear. Our study investigated the specific role of cellular senescence in PFPeA induced liver injury and its underlying mechanisms in C57BL/6 J mice and THLE-2 cells. Our results demonstrat

Next-generation therapies for hepatocellular carcinoma: CAR-T cell and anticancer peptide synergy.

Hepatocellular carcinoma (HCC), the most common primary liver cancer, is characterized by late diagnosis, limited treatment efficacy, and a highly immunosuppressive tumour microenvironment. Chronic liver injury, fibrosis, and inflammation are central to HCC pathogenesis, leading to genetic and epigenetic changes, particularly under oxidative stress from reactive oxygen species (ROS). Aberrant activation of pathways such as Wnt/β-catenin, contributes to uncontrolled proliferation, resistance to apoptosis, and metastasis. This review emphasizes how immunotherapy particularly immune checkpoint in

Advances in pharmacological interventions for hepatic fibrosis: from pathogenic mechanisms to novel therapeutic targets.

Hepatic fibrosis (HF) is a pahological consequence of dysregulated wound healing, characterized by excessive deposition of extracellular matrix (ECM) that disrupts liver architecture and function. It is driven by diverse etiologies, including viral, metabolic, cholestatic and toxic insults. Additionally, the pathogenetic mechanisms progressing from initial injury to established fibrosis involve hepatocyte dysfunction, impaired autophagy, oxidative stress, immune modulation, and ultimately the activation of hepatic stellate cells, leading to a sustained imbalance between ECM synthesis and degrad
