Targeting NF-kappa B/proinflammatory cytokines/ TGF-β/ TIMP-1 pathway by crocin enhances recovery from hepatic fibrosis in rats.

BACKGROUND: Liver fibrosis is a dynamic and potentially reversible process until irreversible structural changes occur. This study evaluated the curative effect of crocin on carbon tetrachloride (CCl₄)-induced hepatic fibrosis in rats and explored the underlying mechanisms. METHODS: Thirty male rats were allocated into three groups: a control group treated subcutaneously (SC) with corn oil for 8 weeks followed by intraperitoneal (IP) saline for 2 weeks; a spontaneous recovery group (CCl₄-SC for 8 weeks followed by saline IP for 2 weeks); and a crocin recovery group (CCl₄-SC for 8 weeks follow

Ferroptosis and hepatic fibrosis induced by cooperative exposure to polylactic acid nanoplastics and copper: Emphasis on gut microbiota dysbiosis.

Co-exposure to polylactic acid nanoplastics (PLA-NPs) and copper (Cu) in the environment poses a health risk, yet their combined toxic effects remain poorly understood. This study investigated the synergistic hepatotoxicity and underlying mechanisms, focusing on the gut-liver axis, in a mouse model of subacute exposure. Results demonstrated that co-exposure caused significant synergistic effects, including exacerbated changes in body weight (BW), increased hepatic index ratio, and severe liver injury marked by elevated Aspartate Aminotransferase/Alanine Aminotransferase/Alkaline Phosphatase (A

Intestinal decontamination with rifaximin attenuates LSEC dysfunction and liver fibrosis in mice.

BACKGROUND: The gut microbiome plays a pivotal role in the development and progression of liver disease. Liver sinusoidal endothelial cells (LSECs), as the first hepatic barrier exposed to blood from the portal circulation, may be influenced by gut-derived microbiota and their byproducts. This study aimed to investigate the interaction between gut microbiota and LSECs and to clarify how this interaction impacts the progression of liver cirrhosis. METHODS: Liver cirrhosis was induced by carbon tetrachloride (CCl4) injection and bile duct ligation (BDL). CCl4 and BDL mice were administered rifa

Next-generation therapies for hepatocellular carcinoma: CAR-T cell and anticancer peptide synergy.

Hepatocellular carcinoma (HCC), the most common primary liver cancer, is characterized by late diagnosis, limited treatment efficacy, and a highly immunosuppressive tumour microenvironment. Chronic liver injury, fibrosis, and inflammation are central to HCC pathogenesis, leading to genetic and epigenetic changes, particularly under oxidative stress from reactive oxygen species (ROS). Aberrant activation of pathways such as Wnt/β-catenin, contributes to uncontrolled proliferation, resistance to apoptosis, and metastasis. This review emphasizes how immunotherapy particularly immune checkpoint in

Advances in pharmacological interventions for hepatic fibrosis: from pathogenic mechanisms to novel therapeutic targets.

Hepatic fibrosis (HF) is a pahological consequence of dysregulated wound healing, characterized by excessive deposition of extracellular matrix (ECM) that disrupts liver architecture and function. It is driven by diverse etiologies, including viral, metabolic, cholestatic and toxic insults. Additionally, the pathogenetic mechanisms progressing from initial injury to established fibrosis involve hepatocyte dysfunction, impaired autophagy, oxidative stress, immune modulation, and ultimately the activation of hepatic stellate cells, leading to a sustained imbalance between ECM synthesis and degrad

Spermidine-Based Carbon Quantum Dots Alleviate Liver Sinusoidal Endothelial Dysfunction by Inducing LSEC-Derived NO to Ameliorate Hepatic Fibrosis and Portal Hypertension.

Portal hypertension (PHT), a life-threatening complication of chronic liver disease, is driven by increased hepatic vascular resistance, with liver sinusoidal endothelial cell (LSEC) dysfunction and impaired nitric oxide (NO) signaling as key contributors. This study synthesized spermidine-based carbon quantum dots (ST-CQDs) and investigated their therapeutic effects on PHT. ST-CQDs (average size 2.12 nm) exhibited good biocompatibility and effectively induced NO production in human immortalized LSECs (hiLSECs) by upregulating endothelial NO synthase (eNOS). In BDL and CCl4-induced PHT rat mod

The Regulatory Role of Iron Transporter SLC39A13 in Liver Fibrosis.

Liver fibrosis, driven by excessive collagen synthesis following hepatic injury, poses a significant health challenge. SLC39A13/ZIP13, a recently characterized intracellular iron transporter, is shown to provide iron to the ER/Golgi to help catalyze procollagen hydroxylation during collagen maturation. Here, we investigate whether ZIP13 plays a role during hepatic fibrogenesis modeled by CCl4 stress or other inducers. ZIP13 expression is induced during liver fibrosis. Germline disruption of Zip13 dramatically reduces fibrosis. Surprisingly, these mice do not benefit from ZIP13 loss after CCl4

Polystyrene nanoplastics exacerbate dibutyl phthalate-induced liver fibrosis through PDGFRα-dependent hepatic stellate cell activation.

Nanoplastic particles (NPs) derived from common polymers such as polyvinyl chloride and polystyrene (PS) act as persistent environmental reservoirs that facilitate the transport and cellular internalization of dibutyl phthalate (DBP), a ubiquitous plasticizer contaminating air, water, soil, and food through plastic leaching. The platelet-derived growth factor receptor α (PDGFRα) pathway is a well-established regulator of hepatic stellate cell (HSC) activation and fibrogenesis; however, its mechanistic involvement in plastic particle-induced hepatotoxicity and its intersection with DBP-mediated

Vesicular nucleotide transporter (VNUT)-dependent ATP secretion by hepatic stellate cells promotes liver fibrosis.

Although liver fibrosis presents a substantial global health challenge, therapeutic options that directly target liver fibrosis remain limited. Hepatic stellate cells (HSCs) are key contributors to fibrosis through extracellular matrix production. This study uncovers a previously unrecognized function of HSCs: ATP secretion. We found that HSCs express the vesicular nucleotide transporter (VNUT) on secretory vesicles and actively release ATP. In mouse HSCs, VNUT is localized to the cytosol and around lipid droplets. In a thioacetamide-induced liver fibrosis model, VNUT inhibition with clodronat

Generation of 3D Liver Microtissues from Induced Pluripotent Stem Cells.

The liver is a multifunctional organ essential for detoxification, protein synthesis, glucose regulation, bile secretion, and drug metabolism. However, persistent damage leads to chronic inflammation, excessive extracellular matrix deposition, and progressive fibrosis culminating in cirrhosis, for which liver transplantation remains the only curative option. Yet, the scarcity of donor organs and risks of immune rejection underscore the urgent need for physiologically relevant in vitro liver models to investigate pathogenesis and facilitate therapeutic discovery. Current two-dimensional culture

Indispensable amino acids vastly protect against CCl4-mediated aggravated liver cirrhosis and immunotoxicity.

Without doubt, liver injury and immunotoxicity are patently interconnected. The liver's role as a leading regulator of the immune system is compromised in advanced liver illness, which may cause immune dysfunction. The liver disorders, when they deteriorate the immunity, making it more vulnerable to systemic damage, while certain chemicals can induce liver injury through direct immune responses. Branch-chain amino acids (BCAAs) are indispensable amino acids immensely recommended as pharmacological nutrients for those with chronic liver diseases to restrict the development of systemic complicat

Iridoids in liver diseases: Mechanistic insights and molecular mechanism.

Over the past decade, the prevalence of hepatic disorders has shown a steady upsurge, raising serious public health concerns. Alcohol consumption, indiscriminate use of medications, a high-calorie diet and exposure to viral infections are all thought to be the primary risk factors for developing hepatic diseases. Due to the lack of effective treatments for hepatic disorders, such as metabolic dysfunction-associated fatty liver disease, alcoholic liver disease, liver fibrosis, cirrhosis and liver cancers, and the numerous side effects of existing therapies, medicinal plants are receiving increa

Inhibitory effect of the methanol extract of Clerodendrum volubile Linn. on carbon tetrachloride-induced early hepatic fibrogenesis: in silico and in vivo study.

Liver disease remains a significant public health problem, claiming several lives globally every year. All forms of liver disease, irrespective of the cause, lead to liver fibrosis if they persist. Liver fibrosis ends up in liver cirrhosis and creates a microenvironment that enhances the development of hepatocellular carcinoma. Clerodendrum volubile is a food and medicinal plant consumed to manage several diseases, including liver diseases. Liver fibrosis was induced in Wistar rats with carbon tetrachloride (CCl4 Olive oil 1:1) for nine weeks and treated with the extract from weeks 7-9. The ac

TG221: An Experimental Model for Liver Cancer Prevention and Treatment Approaches.

Hepatocellular carcinoma (HCC) is a leading cause of cancer mortality. It usually arises in cirrhotic liver, where chronic inflammation and fibrosis create a tumor-permissive microenvironment. Dysregulation of microRNAs (miRNAs), particularly upregulation of the oncomiR miR-221 and loss of the tumor suppressor miR-199a-3p represent key drivers of liver carcinogenesis. The TG221 transgenic mouse, designed to overexpress miR-221 in hepatocytes, provides a relevant in vivo platform for mechanistic studies and for testing preventive and therapeutic approaches. The TG221 model recapitulates miR-221

Simvastatin Targets PKM2 to Alter Metabolic Reprogramming in Hepatic Stellate Cells and Mitigate Liver Fibrosis.

Hepatic stellate cell (HSC) activation plays a crucial role in liver fibrosis progression, with glycolysis and glutaminolysis serving as key components of metabolic reprogramming to sustain HSC activation. Simvastatin (SV) has been shown to ameliorate liver fibrosis; however, its underlying mechanisms remain unclear. This study aimed to explore the protective effects of SV on liver fibrosis with a focus on metabolic regulation. A carbon tetrachloride (CCl4)-induced liver fibrosis model was established in mice, and SV was administered via gavage. LX-2 cells were used for in vitro mechanistic st

Integrating phytochemical analysis and experimental validation of Ziziphus jujuba seed powder and oil to ameliorate CCl4-induced liver fibrosis in sprague dawley rats.

Liver fibrosis, a consequence of chronic liver injury, is characterized by excessive accumulation of the extracellular matrix, oxidative stress, and activation of hepatic stellate cells. This study evaluated the effectiveness of Ziziphus jujuba (Z. jujuba) seed powder and seed oil, both separately and in combination, in alleviating CCl4-induced liver fibrosis, and linked the in vivo effects to their phytochemical profiles. The seed powder extracts, using acetone and ethanol, showed significant antioxidant activity, with DPPH values reaching up to 90.13%, and exhibited a high phenolic content o

Wilson's disease: does a neuropsychiatric phenotype without liver involvement truly exist?

INTRODUCTION: Wilson’s disease (WD) is an inherited disorder of copper metabolism resulting in pathological copper accumulation in multiple organs, predominantly the liver, kidneys, and brain, with subsequent organ damage due to copper toxicity. Although the underlying metabolic defect originates in impaired hepatic copper transport with early hepatocellular injury, the currently accepted clinical phenotypic classification of WD distinguishes two major forms based on presenting symptoms: (1) hepatic and (2) neuropsychiatric, including cases without clinically overt liver disease. The aim of th

Exploring the Impact of Polychlorinated Biphenyls (PCBs) on the Development of MASLD: A Comprehensive Review.

Metabolic dysfunction-associated steatotic liver disease (MASLD), formerly known as non-alcoholic fatty liver disease (NAFLD), is becoming the most common liver disease, affecting between 30 and 40% of the global population. MASLD is a multifaceted disease spectrum that is closely associated with obesity, insulin resistance, type 2 diabetes mellitus and, more broadly, metabolic syndrome. All these conditions increase the risk of liver-related mortality, which explains the intense research efforts in recent years to better elucidate its pathogenesis. The crucial impact of environmental pollutan

Integrative study reveals NR1D1 mediates Hedyotis diffusa's antifibrosis via hypoxia inducible factor-1/ammonia axis.

BACKGROUND: Reversal of hepatic fibrosis (HF) represents a potential cure for chronic liver pathologies; however, clinically approved agents targeting this process remain scarce. Emerging evidence from traditional Chinese medicine (TCM) suggests that Hedyotis diffusa (HD), a botanical agent related to TCM principles of liver pathogenesis, may exert therapeutic effects against fibrotic liver damage. Despite its historical use, the molecular mechanisms underlying its antifibrotic properties and regulatory pathways require systematic elucidation. AIM: To elucidate the efficacy and potential mech
