Dual ligand grafted liposomes for CD44 and CD206 targeted delivery of niclosamide against bisphenol A-induced hepatic fibrosis in albino Wistar rats: In vivo therapeutic efficacy and molecular pathway modulation.

Bisphenol A (BPA) is an endocrine disruptor found in plastics, particularly in food and beverages packaging materials, and can lead to excessive deposition of extracellular matrix. This may result in hepatic fibrosis by activating hepatic stellate cells (HSCs). To address this pathological condition, niclosamide (NIC), an FDA-approved anthelmintic agent, has emerged as a promising antifibrotic compound since it inhibits HSC activation. However, its poor aqueous solubility and availability at the site of action limit its therapeutic application. To overcome this limitation, NIC-loaded hyaluroni

Synthesis and pharmacological evaluation of brominated derivatives of natural product celastrol for treatment of hepatic fibrosis.

Effective therapeutic agents against hepatic fibrosis remain scarce. Natural product Celastrol has demonstrated promising anti-hepatic fibrosis activity. However, further clinical development is impaired by its toxicity. We performed a bromination modification at the C-ring, an unexplored moiety of Celastrol, leading to a series of brominated derivatives. Among them, derivative 5 effectively suppressed various stimulus-induced activation of NLRP3 inflammasome to block the activation of HSCs, thus reducing the collagen deposition. Meanwhile, 5 was identified as a covalent PRDX1 inhibitor with h

Disease-derived liver organoids as a preclinical screening platform identify Sargassum japonica as an anti-fibrotic candidate.

Liver fibrosis remains an unmet medical need with limited therapeutic options and high translational failure. Conventional two-dimensional stellate cell cultures and cytokine-induced organoids poorly recapitulate in vivo pathology. Here, we establish a physiologically relevant liver fibrosis model by generating organoids directly from carbon tetrachloride (CCl₄)-injured fibrotic mouse liver tissue. These disease-derived organoids preserved key pathological hallmarks, including extracellular matrix remodeling and metabolic dysfunction, showing strong transcriptomic resemblance to fibrotic liver

Sexual dimorphism in liver fibrotic metabolic dysfunction: Effects of testosterone and estrogen on CCl4-induced liver injury in ovariectomy and orchiectomy models.

Liver fibrosis is closely linked to metabolic dysfunction and hormonal regulation, with sex differences influencing disease progression. This study combined untargeted liver metabolomic profiling with ovariectomy and orchiectomy mouse models to investigate the roles of estrogen and testosterone in liver injury caused by carbon tetrachloride (CCl₄). Estrogen deficiency aggravated fibrotic injury and liver dysfunction in female mice, whereas testosterone deficiency exerted relatively modest effects in males. Estrogen supplementation significantly attenuated fibrosis in estrogen-deficient females

Lin28A Drives hepatic fibrosis by suppressing HMGA2/HIF-1α-Dependent ferroptosis in hepatic stellate cells.

Effective therapies for liver fibrosis are lacking. The RNA-binding protein Lin28A is upregulated in human cirrhotic and mouse fibrotic livers, where it localizes to activated hepatic stellate cells (HSCs). HSCs-specific overexpression of Lin28A worsened carbon tetrachloride (CCl4)-induced fibrosis in mice. In LX-2 and primary mouse HSCs, Lin28A knockdown suppressed activation and induced hallmarks of ferroptosis, including mitochondrial damage, lipid peroxidation, and glutathione depletion. Mechanistically, Lin28A repressed the maturation of let-7 microRNAs, leading to increased expression of

A robust mouse liver organoid platform enables sustained multicellular maturation and fibrosis modeling from a single tissue sample.

Efficient isolation and culture of liver organoids are critical for studying liver fibrosis, liver regeneration and drug toxicity and screening. However, preserving mature hepatobiliary characteristics and concurrently incorporating fibrosis-producing hepatic stellate cells (HSCs) remains a significant challenge, often hindering the large-scale production of organoids capable of replicating key liver functions. Here, we report a robust 3D organoid culture system that enables simultaneous isolation and long-term propagation of primary hepatocytes, cholangiocytes, and HSCs from a single source o

Exposure to microplastics and liver oncogenesis: A comprehensive review on molecular mechanisms and pathogenic pathways.

Microplastics have become a serious environmental pollutant that enter different ecosystems and eventually the human body through direct exposure to the environment. This infiltration is a major health risk, especially for the liver, which is a primary metabolic organ highly sensitive to the harmful effects of microplastics. There are many ways that these particles can cause liver damage, such as disrupting metabolism, inducing oxidative stress, causing mitochondrial dysfunction, causing inflammation, changing gene expression, and activating abnormal signaling pathways. Even in the absence of

Route-Dependent Proteomic Landscape in Mouse Models of Carbon Tetrachloride-Induced Hepatic Fibrosis.

Hepatic fibrosis, a pathological consequence of chronic liver injury, is characterized by excessive extracellular matrix (ECM) deposition, increasing the risk of hepatocellular carcinoma. The carbon tetrachloride (CCl4)-induced mouse model is well-established for studying the pathogenesis and treatment of hepatic fibrosis, yet the effect of administration routes on fibrotic development and characteristics remains unclear. This study employed comparative proteomics to evaluate fibrosis induced via three CCl4 delivery methods: intraperitoneal (IP), subcutaneous (SC), and intragastric (IG) admini

Glymphatic Dysfunction in Hepatic Encephalopathy: A Novel Pathogenetic Pathway and Potential Target for New Therapeutic Opportunities.

Hepatic encephalopathy (HE) is a common complication of decompensated liver cirrhosis and acute-on-chronic liver failure. It encompasses a range of symptoms, from minimal cognitive impairments to severe personality changes and even coma. Liver dysfunction, coupled with the development of portosystemic shunting (PSS), leads to the accumulation of toxic substances in the brain, adversely affecting its function. Common precipitating factors for HE include variceal bleeding, infections, and electrolyte imbalances. However, other mechanisms likely contribute to the onset of HE, as some patients exp

3-carbamoyl proxyl nitroxide attenuates CCl4-induced liver fibrosis in mice through antioxidant-inflammatory regulation of TLR4/NF-κB signaling pathway.

Liver fibrosis is a dynamic pathological consequence of chronic liver injury, in which persistent oxidative stress and inflammation drive progressive extracellular matrix deposition. Cyclic nitroxide radicals exhibit diverse biological activities, but their effects on liver fibrosis remain unclear. This study systematically evaluates the therapeutic potential of 3-carbamoyl proxyl nitroxide (3-CP) against carbon tetrachloride (CCl₄)-induced liver fibrosis. In vitro, 3-CP inhibited hepatic stellate cell (HSC) activation, migration, and proliferation, and reduced α-smooth muscle actin (α-SMA) an

Potential Therapeutic Strategies for Steatosis, Oxidative Stress, Inflammation, and Fibrosis in Liver Disease.

Liver disease encompasses a wide range of conditions, each requiring tailored therapeutic approaches. This review describes and critically discusses treatments with robust evidence for improving liver health. Ursodeoxycholic acid (UDCA) is a drug approved by the Food and Drug Administration of the USA to treat primary biliary cholangitis (PBC). In addition, UDCA has been demonstrated to protect against metabolic dysfunction-associated steatohepatitis, fibrosis, and drug-induced liver injury (DILI). The mechanism of action of UDCA has been attributed not only to decreasing the effects of toxic

Emerging Alternaria Toxins Drive LX-2 Cells Transdifferentiation into Myofibroblasts for Liver Fibrosis and CotA Detoxification.

Alternariol (AOH), alternariol monomethyl ether (AME), and tenuazonic acid (TeA) are three major emerging Alternaria toxins in food. In this lncRNA-mRNA omics-based study, we first decrypt the blueprint: how emerging Alternaria toxins induce hepatic stellate cell LX-2 transdifferentiation for liver fibrosis. AOH and AME caused the fibrotic marker α-smooth muscle actin, extracellular matrix collagen expression, and cell contraction, while TeA had no significant effect. Mechanistically, AOH, AME, and AAT (their combination) activated the NF-κB pathway and induced ferroptosis and AMPK/AKT/m-TOR-r

Combined exposure effects and potential mechanisms of volatile organic compounds on liver fibrosis in the US population: an integrative epidemiological and computational toxicology analysis.

BACKGROUND: Volatile organic compounds (VOCs) are widespread environmental pollutants that can cause oxidative stress and hepatocellular damage. However, epidemiological evidence linking VOC exposure to liver fibrosis and the underlying molecular mechanisms remains limited. METHODS: Data were obtained from the 2017-2020 National Health and Nutrition Examination Survey (NHANES). Liver fibrosis was defined using the NAFLD fibrosis score (NFS > 0.676). Weighted logistic regression, restricted cubic spline (RCS), and weighted quantile sum (WQS) models were applied to evaluate individual and combi

Pomolic acid alleviates CCl4‑induced liver fibrosis in mice by suppressing β-arrestin 2-mediated pro-fibrotic macrophage polarization.

Liver fibrosis is a wound healing process in response to various chronic liver injuries characterized by the replacement of normal liver by collagen fibers formed from extracellular matrix (ECM), resulting in scarring of the organ. Liver macrophages, a highly heterogeneous and plastic immune cell population, are pivotal in human health and disease, widely involved in the advancement and reversal of liver fibrosis. Interleukin-4 (IL-4) and/or interleukin-13 (IL-13)-induced pro-fibrotic macrophage polarization play a crucial role in promoting liver fibrosis through the secretion of transforming

Hemochromatosis of unknown origin in a young adult dog.

Hemochromatosis is an uncommon condition in dogs. This report describes hemochromatosis of unknown origin in a dog breed not previously identified as predisposed to the condition. A 3-year-old spayed female German shepherd crossbred dog was presented to a local Saskatchewan veterinary clinic with weight loss, muscle wasting, chronic vomiting, diarrhea, and ascites of ~5 to 6 mo duration. The dog was treated symptomatically and was prescribed medication for potential liver failure. Due to poor response to treatment, the owners elected humane euthanasia. The dog was submitted for necropsy at Pra

Pharmacological Mechanisms of Phytochemicals and Pharmaceutical Agents in Protecting Against Methotrexate-Induced Liver Injury.

Methotrexate (MTX) is a widely used chemotherapeutic and immunosuppressive agent for treating various malignancies, autoimmune diseases (AIDs), and inflammatory disorders. Despite its therapeutic efficacy, long term or high-dose MTX treatment is associated with a significant risk of hepatotoxicity, leading to MTX-induced liver injury (MTX-LI). The pathogenesis of MTX-LI involves multiple mechanisms, including oxidative stress, mitochondrial dysfunction, inflammation, and altered metabolic processes, which collectively contribute to hepatocellular damage and fibrosis. Clinically, MTX-LI manifes

Asialoglycoprotein receptor-bound PLGA nanoparticles loaded with Riociguat targetedly ameliorate portal hypertension in liver cirrhosis.

Portal hypertension typically arises with liver cirrhosis and often leads to life-threatening side effects that complicate treatment. Although administrations of vasodilation drugs and/or transjugular intrahepatic portosystemic shunt have been widely used to treat portal hypertension in the clinic, they are usually less satisfactory because those methods may dramatically influence the entire cardiovascular system rather than the liver alone, indicating that an effective and safe approach for reducing cirrhotic portal hypertension is still strongly demanded nowadays. To meet the requirements, a

Kaempferol alleviates high-fat-induced hepatocyte injury by inhibiting ferroptosis.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a key manifestation of metabolic syndrome in the liver. Its core pathological features are excessive lipid accumulation in hepatocytes and the subsequent lipotoxic damage. This not only drives the progression of the disease to hepatitis, liver fibrosis, and cirrhosis but also is significantly associated with an increased risk of hepatocellular carcinoma. NAFLD has become an increasingly serious global public health issue, and there is an urgent need to explore safe and effective prevention and treatment strategies.

The mechanistic nexus between microplastics and chronic liver disease: A review of current evidence.

The progression of chronic liver disease results in severe complications. However, current treatment options remain limited. Research indicates that environmental pollutants, such as microplastics (MPs), pose a threat to the progression of chronic liver disease and represent a substantial challenge to human health. Current studies widely suggest that MPs tend to accumulate in the liver upon entering the body, potentially leading to the onset or acceleration of chronic liver disease. This article seeks to provide a comprehensive overview of the effects of MPs on chronic liver disease through va
