PURPOSE: The transcription factor Ets-1 is widely implicated in hepatic carcinogenesis, yet its precise role in balancing fibrogenesis and regeneration remains poorly defined. This study elucidated the clinical relevance, upstream regulatory mechanisms, and dual cytoprotective/antifibrotic functions of Ets-1 during chronic liver injury and repair.
MATERIALS AND METHODS: Ets-1 networks were evaluated using human cirrhotic tissues adjacent to hepatocellular carcinoma, a rat carbon tetrachloride fibrosis/recovery model, and Ets-1 knockout mice. Upstream regulation and epigenetic gating were char

Hepatic fibrosis (HF) is driven by hepatic stellate cell (HSC) activation. Although cyclic adenosine monophosphate (cAMP) is a key antifibrotic signal, whether pharmacological cAMP elevation acts through protein kinase A (PKA) or exchange protein directly activated by cAMP 1 (EPAC1) remains unclear. Alprostadil (PGE1) elevates intracellular cAMP, but its antifibrotic mechanism is undefined. Using CCl₄-induced HF in rats and primary HSCs, we found that Alprostadil markedly reduced liver injury, collagen deposition, and HSC activation. Fibrotic livers showed increased total cAMP but decreased PK

Liver diseases, including viral hepatitis, fibrosis, cirrhosis, and hepatocellular carcinoma (HCC), remain major global health burdens due to their high prevalence and limited therapeutic options. The need for safer and more effective hepatoprotective agents has renewed interest in traditional herbal medicines such as Radix Bupleuri. Its major bioactive constituents, saikosaponins (SSs), exhibit diverse pharmacological activities. This review synthesizes recent advances in the pharmacodynamics, molecular mechanisms, and toxicological characteristics of SSs, emphasizing their dual hepatoprotect

Liver fibrosis is defined as the substitution of hepatocytes with extracellular matrix (ECM) proteins, accompanied by abnormalities in matrix remodelling. We aimed to assess the efficacy of melatonin-pretreated bone marrow mesenchymal stem cell-derived exosomes (MT/Exos) in mitigating liver fibrosis. MT/Exos were isolated from their parent cells and characterised by measuring protein content, particle size, and cluster of differentiation (CD) markers. In vivo studies (MT/Exos (100 µg/rat) via the caudal vein twice weekly for 4 weeks) include assessment of liver biomarkers, oxidative stress mar

BACKGROUND: Hepatic fibrosis is a progressive pathology driven by hepatic stellate cell (HSC) activation and excessive extracellular matrix deposition. While mesenchymal stem cell (MSC) therapies show promise, their clinical translation is hindered by the inherent safety constraints of live cell transplantation. MSC-derived apoptotic vesicles (MSC-ApoVs), a structurally distinct class of extracellular vesicles, emerge as a compelling cell-free alternative. However, their specific therapeutic efficacy and mechanistic targets in hepatic fibrosis remain elusive.
METHODS: MSC-ApoVs were isolated

Liver fibrosis can progress to cirrhosis and hepatocellular carcinoma, yet effective antifibrotic therapies remain limited. Fibroblast activation protein α (FAPα) is a promising biomarker and therapeutic target, but existing probes are constrained by limited tissue penetration and single-modality imaging, and effective FAPα-targeted antifibrotic agents remain lacking. Here we show that Cs-FAP, a hemicyanine-based near-infrared fluorescence/photoacoustic dual-modal probe, enables real-time and noninvasive visualization of FAPα activity in fibrotic liver tissue. Using a Cs-FAP-guided screening p

Repeated exposure to hepatotoxins such as carbon tetrachloride (CCl4), thioacetamide (TAA), or diethylnitrosamine (DEN) has been frequently used to induce fibrosis/cirrhosis in rodents. In this narrative review, we compared two-stage medium-term protocols using different CCl4 or TAA regimens in DEN-initiated rats and mice. In addition, we revisited our previous two-stage DEN/CCl4 or DEN/TAA rat models to compare fibrosis grade, the mean number, and the percentage of liver area occupied by hepatocellular lesions positive for placental glutathione S-transferase (GST-P). Thus, we evaluated two gr

Liver fibrosis may develop into cirrhosis and liver cancer, lacking effective approved treatments. Breviscapine extracted from Erigeron breviscapus possesses low toxicity and favorable pharmacokinetics, yet its anti-fibrotic mechanisms remain unclear. This study verified its protective effects on cell and rat fibrosis models. It suppresses HSC activation and ECM accumulation by inhibiting TGF-β1/Smad3 pathway, reversing EMT. Breviscapine is a promising candidate for hepatic fibrosis therapy.

Alcoholic liver disease (ALD) develops following chronic alcohol consumption and is characterized by steatosis, inflammation, fibrosis, and cirrhosis. Increasing evidence suggests that dysregulated Wnt/β-catenin signaling contributes to the progression of ALD. Therefore, the present study investigated the hepatoprotective effect of cardionogen-1 (CDNG-1), a Wnt/β-catenin signaling modulator, against alcohol-induced chronic liver injury in C57BL/6J mice. Mice were administered ethanol for 14 weeks to induce chronic liver injury, followed by treatment with CDNG-1 (50 and 100 μM/100 g, i.p.) or s

The current study examines the role of testosterone and androgen deprivation in regulating mitochondrial quality control, autophagy, and apoptosis during CCl4-induced chronic liver injury. Male rats were allocated to Sham, CCl4, TP + CCl4 (testosterone-treated), and Cas + CCl4 (castrated) groups for 4, 8, and 12 weeks, validated through in vitro assays. Mitochondrial function (MitoTracker, MFN2), lysosomal integrity (LAMP1), autophagy markers (LC3B, Beclin-1, p62/SQSTM1), apoptosis regulators (Bcl-2, cleaved caspase-3), and mTOR signaling were assessed by qRT-PCR, immunohistochemistry, and imm

Liver fibrosis is a chronic pathological condition caused by repeated or sustained liver injury, for which effective pathological treatments remain limited. Silymarin, a hydrophobic compound with anti-inflammatory, antioxidant, and antifibrotic properties, has shown therapeutic potential. However, its clinical application is restricted by poor aqueous solubility and low gastrointestinal (GI) bioavailability. In this study, we developed silica-containing redox nanoparticles (siRNP) as an oral nanocarrier system to improve the delivery of silymarin to the liver. Silymarin-loaded siRNP (Sil@siRNP

BACKGROUND: The occurrence of hepatic fibrosis (HF) is closely related to the activation of hepatic stellate cells (HSCs), but the epitranscriptional regulatory mechanisms involved have not been fully elucidated. This study aims to explore the role and molecular mechanisms of the m6A reader protein IGF2BP2 in HSC activation and HF.
METHODS: This study used a CCl4-induced mouse HF model and a TGF-β1-stimulated LX-2 cell model. The interactions between IGF2BP2, AEBP1, and GRB2 were analyzed using bioinformatics, RIP-qPCR, MeRIP-qPCR, RNA pull-down, ChIP-qPCR, dual-luciferase reporter assays, an

As toxicology shifts towards nonanimal testing, quantitative models are essential to predict adverse health effects from molecular or cellular perturbations. Quantitative Adverse Outcome Pathways (qAOPs) represent such models, building on mechanistic knowledge and quantifying the key event relationships (KERs) described in AOPs. Despite the recognized need, the number of qAOPs remains limited. Bayesian-based approaches are often chosen for developing qAOP for their flexibility, but most use discretized variables, limiting their predictive power. In addition, these models are mainly built from

AIMS: CCL28 functions as a key chemoattractant for IgA antibody-secreting cells (IgA+ ASCs) and is crucial for maintaining mucosal immune defense. Recent research has revealed IgA+ ASCs with immunosuppressive activity in non-mucosal tissues under inflammatory conditions. Here, we explored the contribution of CCL28 to chronic liver damage caused by carbon tetrachloride (CCl4).
MATERIALS AND METHODS: We administered CCl4 to wild-type and CCL28 knockout (CCL28 KO) mice to induce chronic liver damage. Liver damage, inflammation, fibrosis, and the expression of chemokines and immunosuppressive mol

BACKGROUND: Familial Mediterranean Fever (FMF) is a monogenic autoinflammatory disease commonly treated with lifelong colchicine therapy. Although colchicine is considered safe, its long-term hepatic effects remain debated. Distinguishing liver injury attributable to FMF itself from that induced by colchicine is essential for guiding clinical management.
OBJECTIVES: To determine whether FMF or colchicine exposure are independently associated with liver cirrhosis and fatty liver disease.
METHODS: This retrospective cohort study utilized electronic medical records from a tertiary care centre i

Liver fibrosis is a major global health challenge with limited treatment options. The marine natural product Psammaplysene D (PD) exhibits promising antifibrotic activity but suffers from significant hERG channel inhibition (>99% at 5 μM) and moderate pharmacokinetics. To address this cardiotoxicity, we employed rational optimization guided by computational toxicity prediction (ADMETlab 2.0), leading to the design and synthesis of derivatives and identification of compound C11. Patch-clamp studies confirmed C11 eliminated hERG liability (IC50 > 100 μM). C11 retained antifibrotic efficacy compa
