Metabolic dysfunction and alcohol exposure drive steatotic liver disease, yet how ethanol intersects with metabolic steatohepatitis remains incompletely defined. Our iterative model examined how ethanol ingestion alters high-fat, -fructose, and -cholesterol (FFC)-driven liver injury, identifying metabolic dysfunction and alcohol-associated liver disease (MetALD)-relevant features. A pilot study in male and female C57BL/6J mice compared chow diet (CD), FFC, CD + ethanol (E), and FFC + E on high-glucose/fructose (sugar) water (SW) at 6, 16, and 24 wk. A redesigned study lowered glucose/fructose

Liver fibrosis arises from a sustained imbalance between oxidative stress, cell death, and matrix deposition. Using the carbon tetrachloride (CCl4) mouse model, we show that the dietary flavonoid, fisetin orchestrates a broad hepatoprotective program that blunts this cascade. Male C57BL/6 mice received CCl4 for six weeks, without/with daily fisetin at 50 or 100 mg/kg. Co-treatment with fisetin preserved body weight and lowered the liver index, and markedly attenuated biochemical injury, reducing serum ALT, AST, ALP, GGT, and TBIL while restoring total protein and albumin levels. In this study, the natural active ingredient glycyrrhizic acid (GA) was used to replace traditional synthetic surfactants to construct GA-functionalized Tanshinone IIA (TSA) nanostructured lipid carriers (TSA@GA NLCs). The prepared TSA@GA NLCs had a particle size of 268.7 ± 2.3 nm and an encapsulation efficiency of 84.90 ± 5.45%. They exhibited a spherical core-shell structure, demonstrated good preliminary short-term stability, and displayed in vitro release profiles consistent with those of an oral sustained-release delivery system. In a carbon tetrachloride (CCl4)-induced mouse model of liv

β-Caryophyllene (BCP), a selective agonist of cannabinoid receptor type 2 (CB2), has garnered attention as a promising nutraceutical agent for modulating organ damage. The current study aimed at evaluating the pharmacological role of BCP in Thioacetamide (TAA)-induced liver fibrosis, with particular attention to the involvement of CB2-mediated signaling. Following the induction of fibrosis by TAA, rats were treated with BCP for 6 weeks. In a separate group, AM630, a selective CB2 receptor antagonist, was co-administered with BCP in order to validate CB2-dependent actions. TAA administration tr

Chronic liver diseases (CLDs), encompassing a spectrum from steatosis and inflammation to fibrosis, cirrhosis, represent a major global health burden, causing approximately 2 million deaths annually [1]. The management of CLDs is significantly hampered by the limitations of conventional approaches, including non-targeted drug delivery, systemic toxicity, and inadequate diagnostic sensitivity for early-stage lesions. Nanotechnology-driven biomaterial platforms have emerged as pioneering solutions to these challenges, enabling precise theranostic strategies tailored to the distinct pathophysiolo

BACKGROUND: Activated hepatic stellate cells (HSCs) are key drivers of liver fibrosis, which progresses irreversibly without effective intervention. The natural alkaloid berberine (BBR) has been reported to possess anti-fibrotic activity. However, the poor solubility and injection-associated systemic toxicity of free BBR confine it to oral administration, resulting in its low bioavailability. Nanocarrier encapsulation addresses these drawbacks, and we therefore aimed to develop an HSC‑preferential delivery system with loaded BBR to improve its anti-fibrosis effects without causing systemic tox

INTRODUCTION: Mast cells (MCs) are myeloid cells of the innate immune system. As a first line of defense, they fulfill effector functions and immune modulatory properties. Upon activation, they release pro-inflammatory mediators such as cytokines and proteases. It has been suggested that MCs may contribute to the development of liver fibrosis. However, investigating hepatic MC biology in mice is challenging due to low MC numbers and a lack of suitable detection techniques relying on MC proteins and their modifications. Here, we evaluated whether the expression strength of MC markers correlates

Liver fibrosis is a significant public health issue that necessitates an ongoing urge for efficient novel treatments. Our newly synthesized fused azine derivative compounds (1a, 1b, 2a, and 2b) were used in this study based on our promising results achieved by our team in previous studies on tacrine derivatives. This study investigated these compounds' hepatoprotective effect on mice with paracetamol (APAP) induced liver fibrosis and clarified their possible mechanisms. The administration of these compounds successfully reduced APAP-induced hepatic fibrosis through decreasing serum liver enzym

OBJECTIVE: Liver fibrosis is characterised by excessive accumulation of extracellular matrix and can ultimately progress to cirrhosis and malignant transformation. Activation of hepatic stellate cells (HSCs), largely driven by transforming growth factor-beta (TGF-β) signalling, is a central event in fibrogenesis. SAE1 is a subunit of the small ubiquitin-like modifier-activating enzyme E1; its role in liver fibrosis and HSC activation remains unclear. This study aimed to investigate the regulatory function and underlying mechanism of SAE1 in hepatic fibrogenesis.
METHODS: A carbon tetrachlorid

Chronic liver disease frequently leads to debilitating bone fractures, yet the mechanistic link between liver injury and skeletal fragility remains elusive. This study aimed to investigate a liver-bone axis where dysregulated iron flux, mediated by hepatocyte ferroportin (FPN), drives osteoporosis during the progression of cirrhosis. We utilized a mouse model of progressive liver disease and generated hepatocyte-specific FPN deletion mice to examine systemic iron homeostasis. Bone quality and microarchitecture were evaluated via micro-computed tomography and histological analysis. To assess fr

BACKGROUND: Primary biliary cholangitis (PBC) is a chronic, immune-mediated liver disease characterised by cholestasis, progressive fibrosis and symptoms of pruritus and fatigue. Ursodeoxycholic acid (UDCA) is first-line therapy; however, many patients respond inadequately or are intolerant. Peroxisome proliferator-activated receptor (PPAR) agonists have emerged as second-line options.
AIMS: This review examines PPAR isoform (PPAR-α, PPAR-δ and PPAR-γ) mediated pathways relevant to PBC, and structural, biochemical and clinical efficacy and safety features of PPAR agonists for PBC.
METHODS: P

Interleukin-33 (IL-33) belongs to the interleukin-1 (IL-1) family of cytokines. Under physiological conditions, IL-33 is abundant in the nucleus of epithelial cells from barrier tissues, epithelial cells of blood vessels, and fibroblast-like cells. Upon pathological stimulation, such as cell damage or tissue injury, IL-33 is released from these cells and binds to the ST2 receptor, a member of the toll-like receptor (TLR)/IL-1 receptor superfamily. The IL-33/ST2 axis is involved in regulating multiple inflammatory diseases, tissue damage, fibrosis, and the development of cancers. Recent studies

Cholestatic liver diseases are a heterogeneous group of hepatobiliary disorders caused by impaired bile formation, secretion, or excretion, leading to hepatocyte injury, biliary inflammation, fibrosis, and eventually cirrhosis. Traditional studies have largely focused on isolated mechanisms, including bile acid toxicity, immune dysregulation, and genetic susceptibility. However, recent advances in metagenomics, metabolomics, and immunology have highlighted the critical role of the gut and biliary microbiota in disease pathogenesis. This review proposes the core concept of a "tripartite interpl

Vitamin D deficiency is associated with poor outcomes and increased mortality in cirrhosis. Calcitriol, the active form of vitamin D3, has antioxidant and hepatoprotective properties; however, its effects on diethylnitrosamine (DEN)-induced liver fibrosis remain unclear. This study investigated the effects of calcitriol on oxidative stress, inflammation, and fibrogenesis in DEN-induced liver fibrosis rat model. Male Sprague-Dawley rats (n = 6/group) were assigned to four groups: control (CON), DEN, DEN + low-dose calcitriol (DEN + LVD3: 5 µg/kg BW), and DEN + high-dose calcitriol (DEN + HVD3:

BACKGROUND: The available evidence suggests an association between depression and single liver disease (e.g., chronic liver disease [CLD], metabolic dysfunction-associated steatotic liver disease [MASLD]), but its association with both multiple single specific liver diseases and major adverse spleen outcomes (MASO) has not been systematically evaluated. The aim of this study was to explore the association between depression and all ICD-10 three-digit coded liver diseases as well as MASO risk in the UK Biobank.
METHODS: The data were obtained from the UK Biobank. Depression was assessed at bas

The spleen plays a significant role in hepatic fibrosis progression. B cells are the main splenic lymphocytes, but their specific contributions and underlying mechanisms remain unclear. Hepatic fibrosis was induced in mice by intraperitoneal administration of carbon tetrachloride (CCl4). Splenectomy or sham operation was performed during the fibrotic process. Hepatic fibrosis severity and the proportion of splenic B cells were assessed. Splenic B cells isolated from fibrotic and control mice underwent mRNA sequencing. Protein expression of phosphorylated-p38 MAPK (p-p38 MAPK), total p38 MAPK,

Atomoxetine (Strattera(R)) is a selective norepinephrine (noradrenaline) reuptake inhibitor that is not classified as a stimulant, and is indicated for use in patients with attention-deficit hyperactivity disorder (ADHD). Atomoxetine is effective and generally well tolerated. It is significantly more effective than placebo and standard current therapy and does not differ significantly from or is noninferior to immediate-release methylphenidate; however, it is significantly less effective than the extended-release methylphenidate formulation OROS(R) methylphenidate (hereafter referred to as osm
