Presence of Legacy and Emerging PFAS in Human Liver Specimens Banked in the United States from 2000 to 2024.

Per- and polyfluoroalkyl substances (PFAS) are persistent, bioaccumulative chemicals linked to liver toxicity and metabolic disease. 54 PFAS were measured in 211 adult human livers collected between 2000 and 2024 to reveal temporal trends, relative PFAS abundance, and demographic predictors of hepatic burden. PFAS were detected in 210 individuals, with 15 compounds found in ≥30 livers. Total summed PFAS concentrations decreased by 94% over the 24-year period in weighted linear regression, and by 68% after adjusting for age, sex, and liver health in multivariate models. Since 2019, a ∼950-fold

Microplastics, nanoplastics and liver disease: an emerging health concern?

Human liver tissue has been found to contain microplastics and nanoplastics (MNPs), with evidence that hepatic MNP accumulation has markedly increased over the past 10 years, prompting critical questions regarding their potential causal role in liver disease. In cell-based and murine models, MNP exposure can trigger oxidative stress, fibrogenesis and inflammation, pathological features that resemble those of advanced liver disease, suggesting shared mechanistic pathways. Furthermore, the capacity for MNPs to act as vectors for microbial pathogens, antimicrobial resistance, endocrine-disrupting

HMGCR genetic variation and risk of new-onset type 2 diabetes in statin users.

BACKGROUND: Statins are widely prescribed lipid-lowering agents, but their use is associated with an increased risk of new-onset type 2 diabetes mellitus (NO-T2DM) ranging from 9% to 13%. While genetic variants in 3-hydroxy-3-methylglutaryl-CoA reductase (HMGCR), the pharmacological target of statins, and the solute carrier organic anion transporter family, member 1B1 (SLCO1B1), a key hepatic transporter, have been implicated in statin response and toxicity, their role in statin-associated diabetogenesis remains unclear. OBJECTIVE: To investigate the association of four common HMGCR variants (

In vivo base editing rescues liver pathophysiology and peroxisome dysfunction in a mouse model of Zellweger spectrum disorder.

Zellweger spectrum disorder (ZSD) is caused by biallelic loss-of-function variants in PEX genes required for peroxisome biogenesis, which is critical for normal cellular metabolism and signalling. The PEX1-p.G843D (c.2528G>A) allele, present in approximately 30% of individuals with ZSD, frequently results in chronic liver disease that can progress to cirrhosis, hepatocellular carcinoma and degraded neurological health. Here we report the development and application of an adenine base editing strategy to correct an established homozygous Pex1-p.G844D ZSD mouse model that manifests liver patholo

Serum amyloid A in the gut-liver axis: Implications for alcohol-associated pathology.

Alcohol-associated liver disease (ALD) remains a major global health concern, with alcohol consumption a leading cause of liver-related mortality. Gut-liver axis has been implicated in ALD development; however, the mechanisms of gut-liver crosstalk are not fully understood. Serum amyloid A (SAA) is a family of highly conserved acute-phase proteins, which is essential in immune-mediated inflammatory processes, like ALD. SAA can bind a diverse set of ligands and perform multiple functions in tissue specific ways. It has been shown that SAA has dual roles in the inflammatory process. It can stimu

Autoradiography and preclinical PET studies with radiolabeled asyn-44 and ACI-12589 for imaging α-synuclein.

BackgroundParkinson's disease (PD) and multiple system atrophy (MSA) are considered α-synucleinopathies, characterized by the presence of pathological α-synuclein (α-syn) aggregates. A positron emission tomography (PET) tracer for imaging α-syn aggregates in vivo is highly sought after, as disease progression correlates with the accumulation of aggregated α-syn. We recently reported [18F]asyn-44 as a radiotracer for α-syn, worthy of evaluation in higher species, based on in vitro binding data from human brain tissues and in vivo PET imaging studies in rodents.Objective[3H]ACI-12589 is a promis

Loss of PPARα-UGT1 axis exacerbates triclosan-induced steatohepatitis in humanized UDP-glucuronosyltransferase 1 mice.

Triclosan (TCS) is an antimicrobial toxicant found in a wide range of consumer products and has been detected in human tissues at concentrations ranging from 0.001 to 5 ppm. Hepatotoxic chemicals such as TCS can cause steatotic liver disease, a condition referred to as toxicant-associated steatotic liver disease. Once TCS enters the body, it induces liver UDP-glucuronosyltransferase (UGT) 1A proteins, leading to the formation of TCS glucuronides, which are biologically inactive metabolites. The induction of UGT1A proteins by TCS is regulated by the nuclear receptor peroxisome proliferator-acti

Liposomal nanotherapeutics for cancer treatment: Targeted delivery and immunotherapy.

Cancer has become a major global health crisis and the second leading cause of death worldwide. With over 270 different types, it is estimated to claim 13 million lives by 2030. The complex pathophysiology of cancer, with its diverse genetic, epigenetic, and biochemical pathways, complicates the diagnostic criteria. Therapeutic approaches such as surgical interventions, radiotherapy, chemotherapy, and immunotherapy have been developed. However, the treatment is still challenging due to higher costs, toxicity, off-target effects, and comorbid conditions. Over the decades, liposomes, based on th

Integrated Multi-Omics Reveals Synergistic Hepatotoxicity of Ethanol and PFOS Co-Exposure.

Alcohol-associated liver disease (ALD) and exposure to per- and polyfluoroalkyl substances (PFAS) share key mechanisms of hepatotoxicity, yet their combined effects remain poorly characterized. We evaluated the impact of concurrent ethanol and perfluorooctanesulfonic acid (PFOS) exposure using a murine Lieber-DeCarli model characterized via multi-omic, spatial lipidomic, and metagenomic analyses. Exposure to PFOS resulted in rapid weight loss, while co-exposure led to decreased survival and pronounced hepatomegaly exceeding the effects of either exposure alone despite reduced cumulative ethano

Tissue metal concentrations and toxicity in wild turkeys following chronic exposure to mining-contaminated soils in southeast Missouri, USA.

The Big River watershed in southeast Missouri (SEMO) is a Superfund site with an extensive history of lead (Pb) and zinc (Zn) mining; cadmium (Cd) and other metals also occur in the ore and waste materials. We examined whether SEMO wild turkeys (Meleagris gallopavo) exposed to mining-contaminated floodplain soils and food resources in the Big River watershed had elevated metal concentrations in their tissues, indicating Pb poisoning and associated toxic effects. Compared to reference birds (n = 15) harvested in northern Missouri, SEMO turkeys (n = 23) had significantly elevated Pb concentratio

A multiobjective AI model for LNP engineering enhances tissue-selective mRNA delivery.

Lipid nanoparticle (LNP) delivery of RNA therapeutics is constrained by poor tissue selectivity and off-target toxicity. Most high-throughput screening approaches have focused on single-target efficacy while overlooking off-target uptake. Here we report multiobjective LNP engineering with artificial intelligence (MOLEA), a system that integrates high-dimensional lipid representations, cell-type-resolved transfection data and multitask optimization to design ionizable lipids with both high potency and biological selectivity. MOLEA learns structure-function relationships across diverse cellular

Transcriptomic comparison on the mechanism of action of four major constituent cannabinoids in hemp extract.

BACKGROUND: A variety of health benefits have been claimed, but not scientifically confirmed, for cannabidiol (CBD) and other nonintoxicating cannabinoids in hemp extracts such as cannabichromene (CBC), cannabigerol (CBG), and cannabinol (CBN). On the other hand, CBD has been reported to cause hepatotoxicity in clinical trials and research studies, while little is known about the safety of other cannabinoids. In the current study, we set out to understand the mechanism(s) of action of these cannabinoids, beneficial or adverse, through a comprehensive functional analysis of a recently published

Di(2-ethylhexyl) phthalate exposure: Association with metabolic associated steatotic liver disease and underlying mechanisms.

Di(2-ethylhexyl) phthalate (DEHP) is a widely prevalent environmental pollutant that poses a threat to metabolic associated steatotic liver disease (MASLD). However, the specific mechanism remains unclear. This study combined the population data from the National Health and Nutrition Examination Survey (NHANES), and jointly employed network toxicology, molecular docking technology and in vitro cell experiments to establish a multi-dimensional research system of "population-network-molecule-cell". To clarify how DEHP interferes with the MASLD process. Our analysis based on NHANES data showed th

Polymer-mRNA complexes for monocyte-trafficked, lymph node-targeted cancer vaccination.

Lymph nodes are the primary sites where adaptive immunity is initiated, yet most messenger RNA cancer vaccines reach them inefficiently and instead accumulate in organs such as the liver, limiting therapeutic potency and increasing systemic toxicity. Here we developed a transferrin receptor-associating polyplex formed by electrostatic complexation of mRNA with low-molecular-weight polyethylenimine that had been chemically modified with cyclic disulfide monomers to enhance nucleic acid binding stability, enable thiol-based transferrin receptor engagement and reduce off-target liver uptake. SN38 PLGA nanoparticles provide sustained antitumor efficacy and ameliorate colitis severity in a murine colitis-associated cancer model.

Irinotecan is a cornerstone therapy for metastatic colorectal cancer (CRC); however, the poor solubility and instability of its active metabolite, SN38, limit its therapeutic efficacy. This study aimed to improve SN38 delivery using poly(lactic-co-glycolic acid) (PLGA) nanoparticles and to evaluate their efficacy and safety in vitro and in a murine colitis-associated CRC model. SN38-PLGA nanoparticles exhibited a mean diameter of 170.7 ± 0.95 nm with a polydispersity index of 0.18 ± 0.04. In vitro, SN38-PLGA exhibited selective cytotoxicity-potent against HCT116 CRC cells but less toxic to HEK
