Current Advances in Antioxidant Nanozymes for Liver Diseases.

The liver with unique physiological functions is susceptible to oxidative stress (OS) damage from reactive oxygen species (ROS) accumulation, further triggering various liver diseases. Compared with conventional pharmacological and surgical therapies, natural antioxidant enzymes represent promise for liver disease therapy due to high ROS scavenging and low immunogenicity. However, their clinical translation is limited by instability and complex preparation. Antioxidant nanozymes not only mimic natural enzymes, but possess advantages including simple structure, low cost and high stability. They

Antibiotic Stewardship in Cirrhosis: Current Recommendations and Clinical Challenges.

Bacterial infections are a major cause of mortality in cirrhosis, with antibiotic prophylaxis and empirical treatment embedded in international guidelines. However, the prevalence of multidrug-resistant organisms and the epidemiology of infections have shifted considerably in recent decades. This review critically appraises the evidence underlying guideline recommendations on antibiotic prophylaxis for spontaneous bacterial peritonitis and acute variceal bleeding in cirrhosis, and antimicrobial management in acute liver failure. The evidence-underlying current recommendations is limited and pr

Hepatoprotective Effects of N-Acetylcysteine and Guarana in a Wistar Rat Model of Acute Acetaminophen Toxicity.

Acetaminophen overdose causes acute liver injury through oxidative stress, inflammation, and hepatocellular damage. This study evaluated the post-exposure effects of N-acetylcysteine (NAC), guarana, and their combination in a rat model of acute acetaminophen toxicity. Thirty male Wistar rats were randomly allocated to the Control, APAP, APAP + NAC, APAP + Guarana, and APAP + NAC + Guarana groups. Acetaminophen (3 g/kg) was administered orally on day 8. Treatments were initiated 4 h later and repeated on days 9 and 10; NAC was administered intraperitoneally at 1.5 g/kg, whereas guarana was admi

From bystander to key player: The mechanisms and therapeutic potential of IL-24 in liver diseases.

Interleukin-24 (IL-24) was initially identified during investigations on melanoma differentiation, and it is a pivotal member of the IL-10 family. As a secreted cytokine, IL-24 is recognized as a tumor suppressor cytokine that induces apoptosis and inhibits angiogenesis. The liver is a unique immunotolerant organ that maintains metabolic homeostasis while continuously responding to insults, such as viral infections, toxic substances, and metabolic stress. In recent years, accumulating evidence has elucidated the role of IL-24 in the initiation and progression of hepatocellular carcinoma (HCC),

Cirrhotic cardiomyopathy: A comprehensive review of pathophysiology, diagnosis, and management.

Cirrhotic cardiomyopathy (CCM) is a prevalent yet frequently underrecognized cardiac complication of liver cirrhosis, characterized by impaired myocardial contractile reserve, diastolic dysfunction, and electrophysiological abnormalities. Despite its clinical significance, particularly in the setting of liver transplantation, CCM remains largely overlooked. We performed a narrative review of the literature on CCM published up to December 2025, covering pathophysiology, diagnostic criteria, imaging modalities, biomarkers, and management strategies. Depending on the diagnostic criteria used, the

Corosolic Acid in Hepatocellular Carcinoma: Molecular Properties, Evidence Boundaries, and Development Priorities.

Hepatocellular carcinoma (HCC) is the most common primary liver cancer and remains a major cause of cancer-related death worldwide. Corosolic acid (CA), an ursane-type pentacyclic triterpenoid with antidiabetic, anti-inflammatory, and antitumour activities, has attracted interest as a potential natural compound for HCC treatment. This review critically evaluates CA as a molecular lead for HCC by integrating its physicochemical characteristics with direct preclinical evidence, cross-model hypotheses, and development constraints. Current studies indicate that CA suppresses HCC cell proliferation

The Role of the Gut-Liver Axis in the Development of Liver Diseases Induced by Selected Environmental Pollutants: A Narrative Review.

Environmental pollution has emerged as a critical global health threat, with xenobiotic exposure increasingly linked to the pathogenesis of chronic liver diseases. The gut-liver axis has been proposed as a potential mediator in the body's response to these insults. This review evaluates how selected environmental pollutants-heavy metals, particulate matter (PM), and endocrine-disrupting chemicals (EDCs), particularly Bisphenol A (BPA)-disrupt this homeostatic balance. Evidence indicates that environmental toxins induce gut dysbiosis and impair intestinal barrier integrity. This "leaky gut" phe

Ligustrum × vicaryi Fruit Polysaccharide Attenuates LPS/D-GalN-Induced Acute Liver Injury in Mice.

The dried fruits of Ligustrum lucidum Ait. are well-established in East Asian medicine for hepatoprotection. Given this ethnopharmacological precedent and their close taxonomic kinship, we hypothesized that the fruit polysaccharide of its horticultural congener, Ligustrum × vicaryi L. (LVFP), may confer analogous hepatic benefits. We assessed LVFP in a murine model of acute liver injury (ALI) induced by lipopolysaccharide and D-galactosamine (LPS/D-GalN) to test this hypothesis. LVFP pretreatment significantly mitigated hepatic pathological damage, normalized serum aminotransferases (AST, ALT)

Cynaroside Attenuates Ethinylestradiol-Induced Cholestatic Liver Injury by Modulating the FXR/BSEP/CYP7A1/SHP Axis.

Cholestatic liver diseases are characterized by disrupted bile acid homeostasis, oxidative stress, and inflammation, ultimately leading to progressive hepatic injury. Although ursodeoxycholic acid (UDCA) remains the standard pharmacological therapy, its therapeutic efficacy is limited in several cholestatic conditions. This study investigated the hepatoprotective effects of cynaroside (CYN), a naturally occurring flavonoid glycoside, against ethinylestradiol (EE)-induced cholestatic liver injury and explored its underlying molecular mechanisms. Female Wistar rats were randomly allocated into f

Sulforaphane Alleviates α-Amanitin-Induced Liver Injury by Modulating Pink1-Parkin Pathway.

BACKGROUND: α-Amanitin (α-AMA) is the primary lethal toxin found in poisonous mushrooms of the genus Amanita; accidental ingestion can lead to severe liver injury, and there is currently no specific antidote. Due to its significant anti-inflammatory and antioxidant activity, Sulforaphane (SFN) has attracted increasing attention for its role in preventing liver injury. However, it remains unclear whether SFN can alleviate α-AMA-induced liver injury. This study aimed to elucidate the protective effects of SFN against α-AMA-induced liver injury and its potential molecular mechanisms. METHODS AND

[Extreme values of alpha-fetoprotein in a patient with autoimmune hepatitis].

We present the case of a 29-year-old woman with a 5-year history of liver function test abnormalities who consulted for polyarthritis, fever, and jaundice lasting one month. Initial studies showed predominantly direct hyperbilirubinemia and elevated transaminases, alkaline phosphatase and gamma-glutamyl transferase. Viral hepatitis was ruled out by negative serology and toxic hepatitis by absence of exposure to hepatotoxic drugs. The antibody panel showed elevated antinuclear antibodies and positive anti-Ro titers. She also presented elevated polyclonal immunoglobulin G levels. Given the chron

Preliminary Structural Characterization of an Anti-Inflammatory Polysaccharide from Lysimachia christinae and Its Preventive Effect on Lipopolysaccharide-Induced Acute Liver Injury in Mice.

An acidic heteropolysaccharide, named LCP-90-3, with a relative molecular weight of 14.02 kDa, was isolated and purified from the traditional heat-clearing Chinese medicine, Lysimachia christinae Hance. The monosaccharide residue analysis indicated that LCP-90-3 was composed of glucuronic acid, mannose, and arabinose in a molar ratio of 79.2: 3.6: 17.2. Linkage analysis further indicated that LCP-90-3 was mainly composed of 1,4-GlcpA, 1,4,6-GlcpA, 1,2-Araf, T-Manp, 1,3-Araf, and 1,4-Manp residues. Furthermore, LCP-90-3 demonstrated potent anti-inflammatory activity by significantly inhibiting

Hepatoprotective Effects of Flavonoids Against Acetaminophen Toxicity.

Acetaminophen (APAP) is a common analgesic and antipyretic agent; however, overdose can cause severe liver injury requiring transplantation or resulting in death. Currently, N-acetylcysteine (NAC) is the standard first-line treatment for APAP-induced liver injury (AILI). There is ongoing interest in exploring complementary hepatoprotective compounds, such as plant-derived flavonoids and their mechanisms of action to improve our understanding and management of AILI. This review aims to identify key flavonoids studied to date in the setting of AILI and their proposed mechanisms. A systematic sea

Corticosteroid Exposure in Autoimmune Hepatitis: Early Toxicity, Cumulative Burden and Associations With Long-Term Patient-Reported Outcomes.

BACKGROUND: Autoimmune hepatitis (AIH) requires corticosteroid-based induction therapy, but prolonged exposure is common in practice. AIMS: This study quantified corticosteroid utilization patterns and associations with adverse outcomes from a patient perspective. METHODS: A cross-sectional patient-reported survey recruited AIHA network members with physician-confirmed AIH (N = 563). Corticosteroid exposure was quantified using annual duration thresholds (≥ 90 days/year at ≥ 5 mg/day prednisone-equivalent). Early side effects (within 6 months; n = 491) and long-term metabolic complications (

Polystyrene nanoplastics potentiate cadmium-induced pyroptosis and apoptosis in primary duck hepatocytes: differential responses to N-acetylcysteine.

INTRODUCTION: Nanoplastics (NPs) can act as carriers for heavy metals, and co-exposure to NPs and co-pollutants may produce greater toxicity than single exposure. However, the combined toxicity of nanoplastics and heavy metals in poultry liver cells remains poorly understood. METHODS: Primary duck embryonic hepatocytes (PDEH) were used as an in vitro model. Cells were exposed to polystyrene nanoplastics (PS-NPs, 80 nm, 10 μg/mL) and/or cadmium chloride (CdCl2, 4 μg/mL) for 12 h. Pyroptosis- and apoptosis-associated endpoints were assessed by brightfield microscopy, PI staining, SEM, flow cyto
