Vitamin A-spermine conjugated lipid nanoparticles for efficient nucleic acid delivery in ocular and hepatocellular cells.

Lipid nanoparticles (LNPs) currently represent the predominant delivery strategy for clinically approved nucleic acid drugs. However, the inherent limitations in targeting, low lysosomal escape efficiency, and potential immunogenic risks pose significant challenges to the broader applications of LNP technology. Vitamin A and spermine are endogenous small molecules that all have shown potentials for low toxicity and efficient drug delivery. In this study, we constructed a series of novel VA-linker-Sper lipids through the conjugation of vitamin A and spermine. With the helper lipid dioleoylphosp

Ginsenoside RG3 and cantharidin synergistically suppress the progression of hepatocellular carcinoma via targeting the PRMT1-SREBF1 axis-mediated lipid metabolism.

BACKGROUND: Ginsenosides, such as ginsenoside RG3, demonstrate antitumor potential in hepatocellular carcinoma (HCC) and are often combined with cantharidin (CTD) in traditional Chinese medicine to achieve synergistic effects while mitigating CTD’s toxicity. However, the precise molecular mechanisms underlying this synergy remain elusive. METHODS: The progression of HCC was assessed using a series of in vitro assays, including CCK-8 for cell viability, EdU staining for proliferation, wound healing for migration, and transwell assay for invasion. The antitumor efficacy and hepatotoxicity were a

Delivery of disulphiram by copper-doped mesoporous silica nanoparticles in combination with doxycycline for treating colorectal cancer liver metastasis.

Colorectal cancer liver metastasis (CCLM) remains a major therapeutic challenge due to chemoresistance and dose-limiting toxicities of conventional therapies. This study developed a copper-doped mesoporous silica nanoparticle (DSF@Cu-MSNs-BSA) for co-delivering disulphiram (DSF) and doxycycline (DOX) to synergistically target CCLM. The nanocomposite was synthesised via Sol-gel co-condensation, surface amination, and albumin adsorption, achieving a high DSF loading capacity (12.69 ± 2.47% w/w) with sustained release. Physicochemical characterisation confirmed uniform spherical morphology (160.8

Bioactivity of Cyperus amuricus extracts against hepatocellular carcinoma and molecular docking analysis targeting the PI3K/AKT/mTOR pathway.

Cyperus amuricus (Cyperaceae) has exhibited potential anticancer activity against hepatocellular carcinoma (HCC), yet its molecular mechanisms and phytoconstituent interactions with oncogenic pathways remain underexplored. This study integrates in vitro cytotoxicity assays and molecular docking to evaluate the effects of C. amuricus fractionated extracts on HCC, focusing on PI3K/AKT/mTOR signaling axis. The ethyl acetate (EA) fraction selectively inhibited HepG2 cell proliferation (IC50 = 159.76 µg/mL) with minimal toxicity to normal fibroblasts. Apoptotic features-cell shrinkage, membrane ble

Impaired nitrogenous waste clearance promotes hepatocellular carcinoma.

In mammals, hepatic urea cycle enzymes (UCEs) convert ammonia, the toxic nitrogenous waste, into urea for excretion. In hepatocellular carcinoma (HCC), UCE expression is often heterogeneously repressed, but its role in tumorigenesis is unclear. We show that, as in patients, UCE expression is markedly reduced in multiple HCC mouse models, including those driven by oncogenic c-MET/β-catenin, leading to impaired ammonia clearance, altered amino acid metabolism, and increased pyrimidine synthesis. In contrast, UCE expression is largely preserved in c-MET/sgAxin1 tumors, allowing assessment of the

Integrative analyses of network pharmacology and bioinformatics reveal the synergistic antitumor effects of cantharidin and ginsenosides Rg3 on hepatocellular carcinoma.

Cantharidin (CTD) has been traditionally used in Traditional Chinese Medicine (TCM) and exhibits pronounced antitumor activity, particularly in hepatocellular carcinoma (HCC). However, its preclinical translation has been substantially constrained by dose-limiting toxicity. Inspired by traditional Mylabris and Ginseng combinations, we conducted an investigation into the synergistic efficacy of CTD and Ginsenoside-Rg3 (Rg3), one of the most prevalent and bioactive ginsenosides, in combating HCC. We find that CTD/Rg3 co-treatment synergistically suppressed HCC cell proliferation, migration, and

Elemene Augments the Effects of Anti-PD-1 Immunotherapy on Hepatocellular Carcinoma by Regulating the miR-130a-5p/SPP/MHC-I Axis.

Immune checkpoint inhibitors, particularly PD-1 inhibitors, constitute the cornerstone of first-line treatment for hepatocellular carcinoma (HCC). However, suboptimal overall response rates persist alongside dual challenges: immune-related toxicities and immunosuppressive tumor microenvironment. Combination therapies represent a pivotal strategy to overcome these limitations. In recent years, traditional Chinese medicine has gained significant attention. Elemene, a small-molecule compound derived from Curcuma wenyujin, has garnered attention for its immunomodulatory potential and demonstrates

Atorvastatin regulates hepatic transcriptome PXR dependently but distinct from pregnenolone 16α-carbonitrile and does not induce PXR-mediated liver steatosis.

Activation of the pregnane X receptor (PXR) by various drugs promotes liver steatosis. Yet, atorvastatin, a widely prescribed statin and a PXR ligand, is considered hepatically safe, even in patients with metabolic dysfunction-associated steatotic liver disease. To reveal mechanistic differences between atorvastatin and steatosis-promoting PXR ligands we investigated whether atorvastatin elicits distinct transcriptional and epigenomic responses, and whether these underlie its favorable hepatic safety. Mice were treated orally for four days with atorvastatin, pregnenolone 16α-carbonitrile (PCN;

Efficacy and safety of first-line EGFR-TKI combined with chemotherapy versus EGFR-TKI monotherapy in patients with EGFR-mutant non-small cell lung cancer and liver metastases: A propensity score-matched analysis.

INTRODUCTION: Liver metastasis (LM) confers a poor prognosis in patients with epidermal growth factor receptor (EGFR)-mutant non-small cell lung cancer (NSCLC). Although EGFR tyrosine kinase inhibitors (TKIs) are the standard first-line treatment, the optimal approach for this high-risk subgroup remains uncertain. This study compared the efficacy and safety of EGFR-TKI monotherapy versus EGFR-TKI combined with chemotherapy in EGFR-mutant NSCLC patients with LM. PATIENTS AND METHODS: We retrospectively analyzed 98 patients with stage IV EGFR-mutant NSCLC and LM treated between 2020 and 2025. F

Entinostat suppresses hepatocellular carcinoma metastasis by upregulating AZGP1 through histone acetylation.

Hepatocellular carcinoma (HCC) has a high mortality rate, primarily driven by metastasis. The role of the histone deacetylase inhibitor (HDACi) entinostat in this process remains controversial, limiting its clinical application. This study aims to define entinostat's function and mechanism in HCC metastasis. We employed in vitro models, transcriptomic sequencing, chromatin immunoprecipitation-qPCR, and an orthotopic mouse model to assess the effects of entinostat on epithelial-mesenchymal transition (EMT), invasion, and tumor growth. Our findings demonstrate that entinostat potently prevented

Hepatocellular carcinoma (HCC) is one of the most common cancers and the third leading cause of cancer-related death worldwide. The prognosis is poor, with a median survival of 12-15 months in patients with advanced-stage disease. Early diagnosis and the development of new, more effective therapeutic strategies are needed to address the challenges posed by this malignancy. Although immune checkpoint inhibitors have replaced multikinase inhibitors as first-line therapy, sorafenib continues to represent a valuable option for patients with contraindications to newer treatments. Based on genome-wi
