Association between noninvasive hepatic fibrosis indices and heart failure mortality.

Heart failure (HF) is a chronic progressive clinical condition characterized by structural or functional impairment of the ventricles. The prevalence of HF is increasing, and it continues to be a significant cause of morbidity and mortality. Liver involvement during fibrosis in HF patients is associated with adverse clinical outcomes. Our study aimed to evaluate the relationship between noninvasive liver fibrosis indices and HF mortality. Our study was a retrospective analysis of patients with HF admitted to our hospital between 2017 and 2023. Patients were followed up until 2024 and divided i

Acetyl-CoA acyltransferase 2 palmitoylation drives liver fibrosis by inducing hepatic stellate cell ferroptosis.

Hepatic fibrosis is a major driver of mortality in metabolic dysfunction-associated steatotic liver disease (MASLD)-previously known as non-alcoholic fatty liver disease (NAFLD). While hepatic stellate cell (HSC) activation and myofibroblast accumulation are central to fibrogenesis, the regulatory mechanisms remain incompletely understood. Acetyl-CoA acyltransferase 2 (ACAA2), a pivotal enzyme in fatty acid oxidation, has been implicated in lipid metabolism but has not been investigated as a therapeutic target in MASLD. Here, we show that ACAA2 upregulation in HSCs exacerbates hepatic fibrosis

Modulation of SIRT1/PPARγ pathways and tight junction proteins by nicotinamide riboside under chronic variable stress.

Chronic stress disrupts homeostasis, leading to major health problems such as liver damage, intestinal barrier dysfunction, and impaired glucose metabolism. Although current treatments, including anxiolytics, sedatives, antidepressants, and beta blockers, are effective, their adverse effects emphasize the need for safer alternatives. Nicotinamide riboside (NR), a precursor of nicotinamide adenine dinucleotide (NAD+), plays a central role in energy metabolism and oxidative stress regulation; elevated NAD + levels have been associated with reduced risk of chronic diseases such as obesity and typ

Engineered Perfusable Hepatic Fibrosis Model via Embedded Sacrificial Bioprinting Recapitulates Stiffness-Driven Fibrogenesis.

Hepatic fibrosis, as the common pathological endpoint of chronic liver diseases, is characterized by a self-perpetuating vicious cycle comprising extracellular matrix (ECM) driven liver tissue stiffening and sustained hepatic stellate cell (HSC) activation. Although existing studies have simulated fibrotic microenvironments using 2D models with tunable matrix stiffness or static 3D cultures, these models lack engineered hepatic sinusoidal vasculature and dynamic mechanical stimulation within 3D ECM contexts. This study employed embedded sacrificial bioprinting to construct functional liver sin

Trigonelline regulates glycolysis and energy metabolism during hepatic fibrosis via Glut-1-HIF-1α axis: Focusing the interaction of macrophages and HSCs.

BACKGROUND: Reprogramming of aerobic glycolysis occurs with HSCs activation and is associated with regression of liver fibrosis. Trigonelline (TRG), a plant alkaloid extracted from Trigonella foenum-graecum L seeds, has a variety of pharmacological effect. PURPOSE: The current study investigated the hepatoprotective effect and mechanism of TRG against hepatic fibrosis by regulating glycolysis. METHODS: Anti-hepatic fibrosis effects of TRG were detected in thioacetamide (TAA)-induced hepatic fibrosis mice. HSCs or LX-2 were stimulated with TGF-β or conditioned medium (CM) from LPS-stimulated

Celastrol Targets Hsc70-Bim Interaction as a Novel Senolytic to Extend Lifespan and Mitigate Organ Fibrosis.

Senolysis holds promise for geroprotection but is limited by efficacy and safety; here we show that Celastrol, a pentacyclic triterpenoid, surpasses benchmark agents ABT-263 and fisetin in senolytic potency and elucidate its mechanism and a prodrug strategy to improve safety. Using stress- and replication-induced senescent cells, we demonstrate that Celastrol selectively triggers intrinsic apoptosis-evidenced by viability assays, Annexin V/PI, cleaved caspase-3 and blockade by the pan-caspase inhibitor Z-VAD-FMK-while ferroptosis is excluded by specific inhibitors. Proteomic, co-immunoprecipit

Metabolic Dysfunction-Associated Steatotic Liver Disease and Peripheral Arterial Disease: Associations and Treatment Considerations.

INTRODUCTION: Metabolic dysfunction-associated steatotic liver disease (MASLD) and peripheral arterial disease (PAD) are highly prevalent conditions that increase cardiovascular risk. This review aims to summarize current evidence on the association between MASLD and PAD, with a particular focus on clinical studies. A critical appraisal of this association will enhance understanding of MASLD as a multi-system disease and provide potential therapeutic implications. METHODS: A literature search was performed using the PubMed database. RESULTS: Both MASLD and PAD are multifactorial diseases tha

Background Liver Fibrosis as a Microenvironmental Risk Factor for Intrahepatic Recurrence in Colorectal Liver Metastases.

BACKGROUND: Colorectal cancer liver metastases (CRLMs) are a frequent pattern of recurrence, yet the prognostic impact of background liver fibrosis remains unclear. This study aimed to quantitatively assess hepatic fibrosis histologically and determine its association with intrahepatic recurrence after resection. METHODS: Patients who underwent hepatic resection for CRLM between 1999 and 2022 were retrospectively included in this study. Background liver fibrosis was evaluated histologically and quantified via whole-slide imaging. Fibrosis was analyzed as a continuous variable using Cox propor

Associations of relative fat mass with metabolic dysfunction-associated steatotic liver disease and liver fibrosis: evidence from the U.S. NHANES 2017-2023.

BACKGROUND: Relative fat mass (RFM) is a simple anthropometric indicator of body fat. Although its association with hepatic steatosis has been examined in previous studies, its predictive value for the more severe stages of liver disease, specifically advanced hepatic fibrosis (AHF) and cirrhosis, has not been systematically evaluated. This study aimed to explore the associations between RFM and the presence of MASLD, AHF, and cirrhosis. METHODS: We conducted a cross-sectional study using data from the National Health and Nutrition Examination Survey (NHANES) 2017-2023. Logistic regression mo

Regulation of Keratin Chemical Modifications: Potential Molecular Mechanisms in Proliferative Diseases.

Keratin, a core structural protein in epithelial cells, is essential for maintaining epithelial tissue integrity. Numerous studies have confirmed its critical role in proliferative disorders, including lung/liver cancer, idiopathic pulmonary fibrosis (IPF), and hepatic fibrosis (HF). Post-translational modification (PTM) regulates protein activity, and keratin undergoes phosphorylation, acetylation, and methylation-modifications that modulate fibrosis and cancer progression by regulating relevant signaling pathways. However, how these modifications reshape keratin's structure and function in t

Data-driven classification of metabolic-associated steatotic liver disease subtypes predicting hepatic and extrahepatic progression.

BACKGROUND: Metabolic-associated steatotic liver disease (MASLD) is a heterogeneous condition with highly variable outcomes. We aimed to distinguish the subtypes of MASLD that were associated with varying risks of hepatic and extrahepatic outcomes. METHODS: An innovative multi-task deep least absolute shrinkage and selection operator (LASSO) algorithm was developed for feature selection in the discovery cohort ( n = 1111, 87.6% [973/1111] of biopsy-proved MASLD), followed by clustering analysis in MASLD. Validation was performed in 6172 individuals who undertook health check-ups (MASLD: 43.9%

Circulating Thrombospondin-2 Levels Reflect Hepatic Fibrosis Severity and Renal Dysfunction in a General Population: Uku Study.

BACKGROUND: Serum thrombospondin-2 (TSP-2) plays a role in various biological processes, including collagen and fibrin formation. Preclinical studies have implicated TSP-2 in liver fibrosis and renal impairment. However, the clinical significance of serum TSP-2 levels in relation to liver and renal function in the general population remains unclear. This study aimed to investigate the association between serum TSP-2 levels and the severity of hepatic fibrosis or renal dysfunction. METHODS: A cross-sectional study was conducted in Uku Town, Nagasaki Prefecture, Japan, in 2019. A total of 225 p

Metabolic dysfunction-associated steatotic liver disease and heart failure with preserved ejection fraction: A state-of-the-art review.

Metabolic dysfunction-associated steatotic liver disease (MASLD) has rapidly become the leading cause of chronic liver disease and cirrhosis worldwide, driven by the global surge in metabolic disorders such as obesity, diabetes, hypertension, and dyslipidemia. In parallel, heart failure with preserved ejection fraction (HFpEF) has surpassed heart failure with reduced ejection fraction (HFrEF) as the predominant form of heart failure, particularly in individuals with metabolic comorbidities. Mounting evidence points to a significant overlap in the pathophysiological underpinnings of MASLD and H

Oncogenic Signaling Activation and Potential Biomarkers in Congestive Hepatopathy Revealed by Proteomic Analysis.

AIM: Fontan surgery improves outcomes in univentricular heart disease; however, some patients develop severe Fontan-associated liver disease (FALD). Even during adolescence, hepatocellular carcinoma occurs in some patients with FALD, although the mechanism remains unclear. In this study, we conducted proteomic analyses of liver tissues obtained using laser capture microdissection (LCM) and serum samples to investigate FALD-related peripheral liver oncogenic factors and explore potential biomarkers. METHODS: Ten-week-old mice underwent sham surgery, and age-matched mice underwent partial ligat

Performance of fibrosis-4 and NAFLD fibrosis score for screening liver fibrosis in Indian individuals with type 2 diabetes: a prospective cross-sectional study.

BACKGROUND: Blood-based non-invasive tests (NITs) in patients with T2DM/MASLD help in assessing the stage of hepatic fibrosis, but there is sparse data in the Indian population. METHODS: Fibrosis-4 (FIB-4) and NAFLD fibrosis score (NFS) were calculated for all individuals with T2DM with MASLD and compared to liver stiffness measurement (LSM) values obtained by vibration-controlled transient elastography (VCTE) with cut-offs of ≥9.7 kPa (rule-in cut-off for advanced fibrosis) and <6 kPa (as a rule-out cut-off for significant fibrosis). RESULTS: The prospective study included 789 patients (238
