Real-world safety of dengue vaccines in Brazil: A pharmacovigilance study.

BACKGROUND: The expansion of dengue vaccination strategies in Brazil, including the introduction of the Butantan dengue vaccine (DV), has reinforced the importance of post-marketing safety surveillance. We aimed to characterize reporting patterns of adverse events following immunization (AEFIs) and immunization errors associated with dengue vaccines in Brazil. METHODS: We conducted a nationwide retrospective observational study using records of AEFIs associated with dengue vaccines reported to the Brazilian Ministry of Health through June 2026. AEFIs were classified as serious or non-serious

First laboratory-confirmed fatal dengue virus infection and its implications for national arboviral surveillance in Liberia.

INTRODUCTION: Dengue virus (DENV) is an emerging public health threat in Africa, where its impact is underestimated due to nonspecific clinical manifestations and restricted diagnostic capabilities. In Liberia, laboratory-confirmed cases of DENV were initially recorded in an urban febrile surveillance study (2018-2020), yet severe or fatal outcomes had not been previously reported. This paper reports the first laboratory-confirmed fatal dengue case and its implications for national surveillance and policy in Liberia. CASE REPORT: On February 2, 2026, a 38-year-old Liberian man arrived at J. J

How accurate are effective reproduction number estimates for mosquito-borne diseases in urban settings?

The time-varying effective reproduction number, Rt, characterizes the dynamics of infectious disease transmission at a given point in time. For mosquito-borne diseases, there has been growing interest in estimating Rt for spatially targeted vector control interventions. However, systematic evaluations of Rt estimation accuracy for mosquito-borne diseases are lacking: a concerning phenomenon given that errors in Rt estimation at fine spatial scales could be amplified by human mobility-driven transmission and high rates of asymptomatic cases. We systematically evaluated the accuracy of popular R

[Nucleoside analogues in etiotropic therapy of hantavirus fevers].

Hantavirus fevers (HF) are caused by pathogenic orthohantaviruses and are classified into two clinical forms: hemorrhagic fever with renal syndrome (HFRS), reported in Eurasia, and hantavirus pulmonary syndrome (HPS), reported in the Americas. HF are characterised by an acute course with predominant involvement of internal organs: HFRS is mainly associated with kidney damage, while HPS is characterised by severe lung injury. Both diseases may also affect the central nervous and endocrine systems. In addition, hantavirus fevers are marked by severe complications during the acute phase and a pro

Wolbachia releases for dengue control: why the evidence supports a pause and independent re-evaluation.

BACKGROUND: Aedes aegypti is the primary vector of dengue and other arboviruses whose global incidence continues to rise. In response, releases of Aedes aegypti infected with the wMel strain of Wolbachia pipientis have been promoted as a sustainable, transmission-blocking strategy to curb arboviral disease. OBJECTIVES: This study was undertaken to reassess the real-world performance of wMel-Wolbachia interventions, evaluate their epidemiological impact and durability under diverse field conditions. METHODS: Evidence from peer-reviewed publications, program documentation, and routine surveill

Hantavirus infection in emergency radiology: Imaging manifestations, pathophysiology, and infection control considerations.

In April 2026, several cases of severe respiratory illnesses were reported aboard the cruise ship MV Hondius during its voyage from Argentina. Subsequent laboratory testing identified Andes virus (ANDV), a hantavirus subtype, with rare but documented human-to-human transmission. Although hantavirus infections are uncommon, both hantavirus pulmonary syndrome (HPS) and hemorrhagic fever with renal syndrome (HFRS) are associated with substantial morbidity and mortality. Their rapid progression and potential need for intensive respiratory support continue to pose important challenges for emergency

Quantitative and qualitative differences in anti-Envelope IgG-secreting cells distinguish subclinical and hospitalized dengue in a Cambodian pediatric cohort.

Dengue virus (DENV) infection can result in outcomes ranging from subclinical disease to life-threatening illness, yet the antibody features mediating protection in humans remain poorly defined. Dengue virus envelope protein (E) forms homodimers on the virion surface and elicits both neutralizing and non-neutralizing antibodies from IgG-secreting cells (IgG-SC). How the developing antibody response at the monoclonal level contributes to protection is unknown. We profiled single IgG-SC responses in a Cambodian pediatric cohort by measuring affinity and serotype cross-reactivity of secreted anti

How climate change affects mosquito-borne infectious disease: The different trajectories of malaria and dengue.

As climate change increases temperatures, additional warmth combines with rainfall and humidity to enhance conditions for mosquito populations to thrive. Greater spread of Anopheles and Aedes vectors increases the risk of transmission of the world's most prevalent mosquito-borne infectious diseases, malaria and dengue. Yet the impacts of climate drivers on disease transmission are complex: Relationships are nonlinear, effects are seasonal, and there is substantial local variability. Further complexity is added by the many nonclimate factors that influence disease transmission, including moveme

The Social Amplification of Secondary Risk Perceptions: Online Media Attention, Interpersonal Discussion, and Dengue Vaccination Willingness in Singapore.

Drawing on a combined perspective integrating the social amplification of risk framework and secondary risk theory, we examined the ways in which people's online attention and interpersonal discussion shape their perceptions of secondary risks related to the dengue vaccine, as well as how these perceptions subsequently impact their willingness to receive the vaccine. Data were collected through an online survey (N = 1,001) using quota sampling methods. The findings indicated that interpersonal discussions had a positive connection to individuals' perceptions of both the severity and susceptibi

Safety and Immunogenicity of the Live-Attenuated Quadrivalent Dengue Vaccine V181 Compared With Butantan-DV Among Healthy Adults in Brazil: A Randomized, Double-Blind, Phase 2 Trial.

BACKGROUND: Dengue is caused by infection with 1 of 4 dengue virus (DENV) serotypes (DENV-1 to DENV-4). V181 and Butantan-Dengue Vaccine (Butantan-DV) are independently developed, single-dose, quadrivalent dengue vaccines based on the same attenuated parental strains. METHODS: In this randomized, double-blind, active comparator-controlled, multisite, phase 2 trial, we compared the safety and immunogenicity of V181 and Butantan-DV in healthy adults in Brazil. The primary immunogenicity objectives were to compare vaccine-induced neutralizing antibody geometric mean titers (GMTs) and seroconvers

Meta-analysis of Maternal and Fetal Outcomes of Crimean-Congo Hemorrhagic Fever during Pregnancy.

Crimean-Congo hemorrhagic fever (CCHF) during pregnancy is associated with severe maternal and fetal complications, but evidence remains limited to heterogeneous case reports. We synthesized patient-level data from published and unpublished cases identified through systematic database searches and direct collaboration with centers in CCHF-endemic areas. We harmonized patient-level clinical, laboratory, obstetric, treatment, and outcome data for 55 pregnancies. The maternal mortality rate was 28.8% (15/52), and fetal loss occurred in 57.4% (31/54) of cases. Maternal mortality rates increased wi

A small molecule inhibits Ebola virus entry through glycoprotein stabilization.

Ebola virus (EBOV) causes outbreaks with high fatality rates, yet potent small-molecule inhibitors targeting EBOV glycoprotein (GP) remain limited. Here, using DNA-encoded chemical libraries, we screen 4.73 billion compounds against EBOV GP and, through stereochemical optimization, identify MWAC-3634. This compound binds GP with high affinity (Kd=40.7 nM), potently neutralizes infection (IC₅₀ = 0.65 nM), shows no detectable cytotoxicity, and exhibits favorable pharmacokinetics in mice. This potency substantially exceeds that of known small-molecule inhibitors targeting EBOV GP. Cryo-EM at 2.54

The circSNX6 restricts dengue virus replication in association with PARP1 and ferroptosis.

Dengue virus (DENV) infection poses a persistent global health challenge with no specific antiviral therapy. Although circular RNAs (circRNAs) regulate antiviral immunity, their roles in DENV infection remain largely unexplored. Here, transcriptome sequencing of peripheral blood mononuclear cells (PBMCs) from dengue patients across disease phases and severities identified 15 circRNAs consistently dysregulated across all clinical stages, enriched in ferroptosis, glutathione metabolism, and endocytosis pathways. Among these, hsa_circ_0031607 (circSNX6) was the sole circRNA linked to all three pa

Characteristics and outcomes of dengue infection in kidney transplant recipients: an integrative review.

Dengue is an arbovirus infection caused by the dengue virus, common in tropical and subtropical areas. It is mainly transmitted by bites from infected Aedes aegypti and Ae. albopictus mosquitoes, but donor-derived cases have been reported. In kidney transplant recipients with dengue, immunosuppressive therapy can affect their immune response and disease presentation. This study analyzed the characteristics and outcomes of kidney transplant recipients diagnosed with dengue. An integrative review was conducted, including case reports, series, case-control, and cohort studies. The PubMed/Medline

Early Action Review of Detection, Notification, and Response Timeliness during Cross-Border Bundibugyo Virus Disease Outbreak, Uganda, 2026.

Bundibugyo virus disease (BVD), an Ebola virus species with no licensed vaccine or therapeutic, reemerged in May 2026 as a cross-border outbreak in Uganda and the Democratic Republic of the Congo. During a 2-day workshop, July 8-9, 2026, we conducted an early action review of the outbreak response using the 7-1-7 framework (7 days to detect, 1 day to notify, 7 days to complete early response actions) to assess timeliness and identify bottlenecks and enablers across 9 response pillars. Uganda declared its outbreak on May 15, 2026; by July 8, the country had recorded 20 confirmed cases (15 impor

Molecular confirmation of transfusion-transmitted dengue virus infection following platelet transfusion in the Chinese mainland.

BACKGROUND: Dengue virus (DENV) is considered an emerging threat to blood safety. While transfusion-transmitted DENV (TT-DENV) cases have been documented, TT-DENV has never been reported in Chinese mainland. In this study, a retrospective investigation was conducted after two patients developed dengue fever (DF) following apheresis platelet (PLT) transfusion from the same healthy donor. METHODS: Blood samples from the recipients and the donor were tested for DENV RNA by nucleic acid testing (NAT). Nonstructural protein 1 (NS1) antigen and anti-DENV immunoglobulin M/G (IgM/IgG) antibodies were
