Dengue warning signs persisting post-48h demand monitoring to avert severe complications.

INTRODUCTION: Dengue virus infection remains a major public health challenge in tropical regions, with heterogeneous progression to severe disease. METHODS: We conducted a prospective longitudinal study of 362 laboratory-confirmed dengue patients, stratified by sex and the presence of warning signs. Serial hematological, hepatic and renal markers were collected over 96h (at 24h, 48h, 72h, and 96h after symptom onset) to analyze their dynamic evolution. RESULTS: Patients with warning signs persisting beyond 48h exhibited progressive hematological and hepatic deterioration, unlike those withou

In Silico Analysis of Potential Vaccine Antigens for the Treatment of Crimean-Congo Hemorrhagic Fever Virus (Cchfv).

BACKGROUND: Crimean-Congo hemorrhagic fever virus (CCHFV), identified by the World Health Organization(WHO) as a potential epidemic threat, is a tick-borne Nairovirus primarily transmitted by Hyalomma marginatum species. Since its first detection in the 1940s, CCHFV has spread to multiple regions worldwide and remains a major public health concern due to its high fatality rate and expanding geographic distribution. The virus can be transmitted through tick bites or contact with infected individuals, and no licensed vaccine is currently available to prevent infection. METHODS: In this study, t

Despite good intentions, the regulation on in vitro diagnostic medical devices (IVDR) in Europe could impact negatively on preparedness and response for the next pandemic.

Currently, there is concern and uncertainty in the European and North American markets for in vitro diagnostics regarding the regulation of in vitro diagnostic tests. In the European Union, starting from May 2022, the regulation on vitro diagnostic medical devices (IVDR) has replaced the directive on in vitro diagnostic medical devices (IVDD). The IVDR, while written with the good intentions to ensure patient safety and health while supporting innovation and transparency, has resulted in uncertainty, instances of disruption of diagnostic development, and concerns related to pandemic preparedne

In silico design of a multi-epitope vaccine targeting DENV-1 and DENV-3.

The co-infection of DENV-1 and DENV-3 during endemic outbreaks can be potentially fatal and complicate the diagnostic process. In our study, we have focused on the development of a multiepitope vaccine against DENV-1 and DENV-3 co-infection, utilizing non-structural protein 1 (NS1) and envelope protein (E) as key antigens. B cell and T cell epitopes were predicted for their immunogenicity, antigenicity, and ability to elicit an IFN-γ response. The final construct showed predicted stability (Instability Index: 30.63), antigenicity (0.5509), non-allergenicity, and hydrophilic character (GRAVY: -

Evaluation of polyclonal antibodies raised in rabbits against dengue NS1 antigen.

Dengue, a vector-borne disease caused by the dengue virus (DENV), is spread by Aedes mosquitoes like, Aedes aegypti and Aedes albopictus. In the absence of antiviral drugs or vaccines, dengue prevention is centered on mosquito vector control. Laboratory confirmation of DENV infection is essential to offer the appropriate clinical care. The non-structural protein-1 (NS1) antigen of the virus, in its secretory form, is being employed as a diagnostic marker to identify active dengue infection in humans and mosquito vectors. Commercial dengue NS1 ELISA kits use monoclonal antibodies to detect the

The economic burden of dengue: a systematic literature review of unit costs for non-fatal episodes treated in the formal healthcare system.

BACKGROUND: Dengue, a vector-borne disease caused by the dengue virus, has emerged as a global public health concern, given the tenfold rise in reported cases over the last two decades. In light of the upcoming dengue interventions, country-specific cost-of-illness estimates are required to evaluate the cost-effectiveness of new interventions against dengue. This study aims to conduct an updated systematic review of dengue cost-of-illness studies, extracting the relevant data, and conducting regression analysis to explore potential factors contributing to the cost variations among countries. Temporal cytokine profiling of acute dengue, Zika, Chikungunya, and Mayaro virus infections in Northern Peru.

BACKGROUND: Arboviruses such as Dengue virus (DENV), Chikungunya virus (CHIKV), Zika virus (ZIKV), and Mayaro virus (MAYV) are major causes of acute febrile illness (AFI) in Peru. However, their overlapping clinical symptoms complicate differential diagnosis. Identifying immunological markers like cytokine profiles could facilitate differential diagnosis and improve understanding of their immunopathogenesis. This study aimed to describe the cytokine responses (IL-2, IL-6, IL-10, TNF-α, IFN-γ) associated with each infection. METHODS: A cross-sectional study was conducted from June 2020 to April

Innovative application of a traffic-prediction spatio-temporal graph convolutional network for dengue disease forecasting.

Dengue fever, a vector-borne disease, is a major public health challenge. Accurate prediction methods that can better reflect the complexity of the outbreak are essential for dengue prediction and vector control. In this study, we introduce an adapted Spatio-Temporal Graph Convolutional Network (STGCN), originally developed for traffic forecasting, to predict weekly dengue cases in nine countries in South and Central America from 2014 to 2022. In this approach, we use environmental and socio-economic data in addition to climate data and historical dengue case information to capture complex tra

Animal models illuminate dengue immunopathogenesis to guide vaccine and therapeutic developments.

Dengue virus (DENV) infection affects over 390 million people annually, and its severe forms, including dengue hemorrhagic fever (DHF) and dengue shock syndrome (DSS), occur as a result of antibody-dependent enhancement (ADE), T-cell dysregulation, and a cytokine storm. This review integrates the results of human immunopathology with those reported by animal models, consolidating the findings in a systematic manner to validate these mechanisms. Research on AG129 interferon receptor-deficient mice consistently demonstrates ADE-mediated viremia and vascular leakage. However, humanized NSG-BLT mi

Dengue in children and adolescents with onco-hematological disease: a case series.

Dengue is a viral disease transmitted mainly by the Aedes aegypti mosquito. Argentina has seen a significant increase in epidemics in recent years. Pediatric patients with oncohematological diseases are particularly vulnerable due to their immunosuppression, which can predispose them to severe cases. This report presents five clinical cases of pediatric patients with oncohematological diseases who developed dengue infection, illustrating the variability in its clinical presentation. Symptoms were heterogeneous, ranging from mild forms to severe cases with liver failure, bleeding, and the need

Bridging the knowledge gap: Thai parents' perspectives on dengue infection and its vaccination and the need for targeted promotion.

BACKGROUND: Dengue infection is endemic in Thailand, with children being the most impacted group. Various measures, including the dengue vaccine (recently recommended in Thailand since 2017 for Dengvaxia and 2023 for Qdenga), have been implemented to prevent and manage the disease. However, the rate of infection among Thai children is still high. This study explored Thai parents' knowledge and attitudes toward dengue infection and its preventive vaccine, guided by the 4C Model for vaccine hesitancy. METHODOLOGY: This cross-sectional study employed a questionnaire survey, utilizing five invest

Epidemiological and digital syndromic surveillance data on dengue, chikungunya, and SARI in Brazil.

Vector-borne diseases, such as dengue and chikungunya, along with air-borne diseases like influenza and COVID-19, are prone to epidemics, which increases the demand for real-time outbreak monitoring. Developing such systems requires harmonized datasets for calibration and validation. In this study, we created a dataset containing official disease notifications for dengue, chikungunya, and severe acute respiratory infections (SARI) across Brazilian states for over a decade. The dataset integrates Google Trends search data for each disease and all associated symptoms, organized by the correspond

An mRNA vaccine encoding the Ebola virus glycoprotein induces high neutralizing antibody titers and provides strong protection against lethal infections in mouse models.

Ebola virus (EBOV) is the causative agent of Ebola disease (EBOD), a viral hemorrhagic fever with a notably high case fatality rate. Current treatments for EBOD are limited to monoclonal antibodies or two licensed viral vector vaccines, a recombinant vesicular stomatitis virus (rVSV)-vectored vaccine or an adenovirus and modified vaccinia Ankara regimen. However, comparisons of protection, efficacy, and durability with alternative nucleotide platforms remain understudied. Here, we evaluated the immunogenicity of an mRNA vaccine expressing the EBOV glycoprotein (GP) in parallel with rVSV- and D

Measuring real-time disease transmissibility with temperature-dependent generation intervals.

Accurate real-time estimation of the effective reproduction number (Rt) is critical for infectious disease surveillance and response. In vector-borne diseases like dengue, temperature strongly influences disease transmission by affecting generation times. However, most existing Rt estimation methods assume a fixed generation interval, leading to biased estimates and unreliable assessments of transmission risk in settings with fluctuating temperatures. In this study, we proposed and evaluated a novel framework to estimate a temperature-dependent reproduction number (td-Rt) that dynamically upda

Sociodemographic determinants of willingness to adopt surveillance technology and mosquito control training for dengue in Malaysia.

Dengue fever remains a major public health concern in Malaysia, where rapid urbanization and ineffective vector control measures contribute to persistent outbreaks. While drone-based mosquito surveillance and mobile applications offer innovative solutions, public acceptance and adoption remain uncertain. Additionally, community-based mosquito control training is crucial, yet participation is often influenced by sociodemographic factors. This study examines determinants influencing willingness to adopt a dengue surveillance mobile application and engage in mosquito control training programs. A
