Acute Fulminant Cerebral Edema Caused by Coxsackievirus A6 Infection: A Case Report.

In recent years, Coxsackievirus A6 (CV-A6) has gradually replaced Enterovirus 71 (EV-71) and Coxsackievirus A16 (CV-A16) as the main pathogen causing hand foot mouth disease (HFMD) in China. This article reports a fatal case of HFMD caused by CV-A6, leading to fulminant cerebral edema and cardiopulmonary arrest. A 6-year-old boy was admitted with a chief complaint of "fever for 1 day, two episodes of seizures". On admission, the patient exhibited unresponsiveness, no spontaneous breathing, bilateral fixed and dilated pupils, complete muscle weakness, and loss of muscle tone. The brain computed

Validation of important genes and bioinformatics investigation of the relationship between ferroptosis and CVA16 infection.

Coxsackievirus A16 (CVA16) is a major pathogen linked to hand, foot, and mouth disease (HFMD), and effective therapies and vaccines are lacking. Ferroptosis, an iron-dependent form of cell death, may play a role in CVA16 infection and remains underexplored. We analyzed microarray datasets (GSE71673) from the Gene Expression Omnibus (GEO) and ferroptosis-related genes (FRGs) from FerrDb. A Venn diagram revealed 37 ferroptosis-related differentially expressed genes (FR-DEGs), and Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were conducted on these genes. Protein

Short-term effects of meteorological factors on hand, foot, and mouth disease in Zhengzhou, China.

Hand, foot, and mouth disease (HFMD) demonstrates distinct seasonal patterns in high-incidence regions, highlighting the growing interest in the relationship between meteorological factors and disease transmission. Nevertheless, findings across different geographic areas remain highly heterogeneous. Data on HFMD cases in Zhengzhou City from 2009 to 2023 were obtained from the Zhengzhou Center for Disease Control and Prevention, while meteorological data were sourced from the National Meteorological Data Sharing Service System. Distributed lag nonlinear models (DLNM) was employed to quantify th

Population-Level Impact of the Enterovirus A71 Vaccination Program on Hand, Foot, and Mouth Disease: Ecological Time-Series Study.

BACKGROUND: Hand, foot, and mouth disease (HFMD), a common childhood illness caused by various enteroviruses, poses a significant public health threat in the Asia-Pacific region, where severe cases associated with enterovirus A71 (EV71) are a major concern. The EV71 vaccination program was introduced in China in late 2016. Although randomized controlled trials have established the robust efficacy and safety of these vaccines, assessing their real-world performance remains crucial. Subsequent studies have evaluated its real-world effectiveness in several provinces, including Zhejiang and Guangd

Engineering of cell line assembled enteric organoid for enterovirus infection.

The non-polio-enteroviruses are ubiquitous pathogens infecting over a billion people in the world. An alarming number of enterovirus-associated acute flaccid paralysis, encephalitis, hand, foot, and mouth disease, conjunctivitis and diarrhoea cases are reported worldwide. Despite their clinical significance, vaccine development has been hindered due to lack of suitable in vitro models for preclinical investigations. The present study was undertaken to develop a cell line assembled organotypic model of human intestine for replication of enteroviruses. An enterovirus specific PSGL1 receptor was

Standardized neutralizing antibody assay for Coxsackievirus A10 in clinical trial: A framework for multivalent hand, foot, and mouth disease vaccine evaluation.

Enteroviruses (EVs) are the major etiological agents of hand, foot, and mouth disease (HFMD), which could lead to severe complications and even fatalities. Owing to the alternating or co-circulation of multiple EV serotypes and the lack of cross-protection, global institutions are pursuing the development of multivalent HFMD vaccines. Notably, the advancement of innovative multivalent HFMD vaccines into the clinical stage highlights the urgent need for a standardized platform to assess immunogenicity. In accordance with international regulatory guidelines, a standardized neutralizing antibody

CBX4 facilitates EV71 replication by SUMOylation and stabilizing 3D polymerase.

Enterovirus 71 (EV71) is a primary etiological agent of hand-foot-mouth disease (HFMD) in children under 5 years of age and can cause severe neurological disorders even death. Therefore, elucidating the infection mechanism and pathogenicity of EV71 is essential for developing more effective and targeted therapies to prevent and control EV71-associated diseases. Here, we initially reported that the SUMO E3 ligase CBX4 is important for EV71 replication. Furthermore, we found that CBX4 interacts with the EV71 3D polymerase, and overexpression of CBX4 significantly extends the half-life of 3D, whe

Epidemiological shifts in coxsackievirus A-induced hand, foot, and mouth disease in China, 2008-2024.

INTRODUCTION: Hand, foot, and mouth disease (HFMD) is primarily caused by enterovirus A71 (EV-A71) and coxsackievirus A (CV-A) serotypes. The introduction of the inactivated EV-A71 vaccine in China in 2016 significantly reduced EV-A71-related severe and fatal HFMD cases but did not confer cross-protection against other enteroviruses. Epidemiological data from China show that while the overall incidence of HFMD has declined since 2018, there has been a significant shift in the pathogen spectrum, with non-EV-A71 enteroviruses particularly CV-A6 and CV-A10 emerging as the predominant serotypes.

Systematic Proteomic Characterization of EV-A71-Infected Mice Identifies Dynamic Molecular Changes and Therapeutic Targets.

Enterovirus A71 (EV-A71) is recognized as the primary causative agent of hand, foot, and mouth disease (HFMD) and is prevalent worldwide. However, the precise pathogenic mechanisms of EV-A71 remain unclear, and specific drugs targeting it have yet to be successfully developed. To explore the mechanisms underlying EV-A71 pathogenesis and to identify potential therapeutic opportunities, we performed a comprehensive proteogenomic characterization of muscle tissues from BALB/c mice infected with EV-A71, integrating transcriptomic, proteomic, and phosphoproteomic analyses. Our results showed that p

Clinical Utility of Small Extracellular Vesicles as Liquid Biopsy for Oral Mucosal Disease Diagnostics: Emerging Perspectives.

Some diseases affecting the oral mucosa can be life-threatening and/or associated with life-threatening complications. Conventional diagnostic methods for most oral mucosal diseases are usually employed at a fully established disease state. All these peculiarities usually result in late diagnosis, poor prognosis, poor treatment outcomes, and reduced overall survival rates, hence the need for novel methods for the early detection of these disease conditions. Small extracellular vesicle (sEV)-based diagnosis carries great potential for early diagnosis of oral mucosal diseases, as sEVs reflect th
