Architectural Regulation of T-Lymphocyte Function in Hepatocellular Carcinoma: Current Evidence and Translational Opportunities.

Hepatocellular carcinoma (HCC) develops within a chronically injured liver in which anti-tumor immunity is governed not only by immune-cell abundance and checkpoint signaling, but also by the spatial, mechanical, and multicellular organization of T lymphocytes. This review reframes T-cell architecture as a regulatory layer operating across molecular, cellular, and tissue scales. Cytoskeletal remodeling, immunological synapse geometry, mechanotransduction, vascular positioning, stromal exclusion, and tertiary lymphoid structure (TLS) organization collectively influence T-cell activation thresho

Developing a Molybdenum(IV) Agent Based on a Specific N-Donor Residue in Human Serum Albumin IB Subdomain: Multi-Targeted Inhibition of Growth and Metastasis in Hepatocellular Carcinoma.

To achieve the targeted inhibition of hepatocellular carcinoma (HCC) growth and metastasis, we optimized a series of molybdenum (Mo) compounds to obtain a Mo(IV) 2-acetylquinoxaline thiosemicarbazone compound (Mo5) with remarkable cytotoxicity, and constructed a Mo5-human serum albumin (HSA) complex delivery system (Mo5-HSA). Structural analysis revealed that Mo5 binds to the IB subdomain of HSA, where His146 replaces a Cl ligand of Mo5 and coordinates with the Mo center. The Mo5-HSA complex exhibited improved inhibitory efficacy against HCC growth and metastasis, enhanced targeting ability an

Discovery of Potent, Selective DNMT1 PROTAC Degraders: Simultaneous Blockade of Catalytic and Non-Catalytic Functions for Liver Fibrosis Therapy.

Liver fibrosis is a progressive pathological condition driven by chronic hepatic injury and currently lacks effective therapies. DNA methyltransferase 1 (DNMT1) promotes fibrogenesis through both catalytic and non-catalytic mechanisms. To simultaneously target these dual functions, we designed and synthesized a series of DNMT1-targeting PROTAC degraders based on DC-517. Our optimization campaign culminated in compound B7, which emerged as the most potent agent, mediating ubiquitin-proteasomal degradation of DNMT1. In vitro, B7 reduced global DNA methylation and altered readouts associated with

In situ CAR-M engineered by liposomal mRNA nanomedicine remodels fibrotic microenvironment for ameliorating liver fibrosis.

Liver fibrosis is a severe complication of diverse chronic liver diseases characterized by a dysregulated hepatic microenvironment, and effective clinical therapeutic strategies for this condition remain lacking to date. Chimeric antigen receptor macrophages (CAR-Ms) hold considerable therapeutic potential for liver fibrosis via antigen-specific recognition and efferocytotic activity, yet their clinical translation is largely hindered by the sophisticated ex vivo preparation procedures. Herein, we report an in situ CAR-M engineering strategy that uses a phosphatidylserine (PS)-incorporated, ma

Lycium barbarum polysaccharides and melatonin alleviate cadmium-induced hepatic metabolic injury associated with alterations in PI3K/AKT signaling.

Lycium barbarum polysaccharides (LBP), bioactive macromolecules derived from Lycium barbarum, exhibit metabolic regulatory and hepatoprotective activities. Cadmium (Cd) exposure is a major environmental concern associated with hepatic metabolic dysfunction and liver injury; however, the potential protective role of LBP against Cd-induced hepatic dysfunction and the effects of its combined administration with melatonin (MT) remain incompletely understood. In this study, we investigated the protective effects of LBP, alone or in combination with MT, in a mouse model of chronic Cd exposure. Cd ex

Aqueous extract of Artemisia capillaris Thunb. herbal tea alleviates liver injury in MASLD mice and lipotoxicity in HepG2 cells by inhibiting apoptosis and promoting autophagy.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a chronic metabolic disorder characterized by diffuse hepatic steatosis. Excessive lipid accumulation induced by a high-fat diet can trigger hepatocyte apoptosis and impair autophagic flux, thereby aggravating liver injury. Artemisia capillaris Thunb. herbal tea contains multiple bioactive constituents with hepatoprotective, choleretic, and lipid-regulatory activities. Therefore, this study aimed to evaluate the protective effects of Artemisia capillaris Thunb. herbal tea aqueous extract (ACTE) against MASLD-associated hepatic

Probiotic bacterial ghosts enhance oral astaxanthin delivery to alleviate non-alcoholic fatty liver.

Astaxanthin (AXT) is a promising natural bioactive compound for the nutritional intervention of non-alcoholic fatty liver (NAFL), however, its poor stability and limited oral bioavailability restrict its practical application. In this study, a novel probiotic-derived oral delivery system, designated AXT@LPGs, was developed by loading AXT into Lactiplantibacillus plantarum bacterial ghosts (LPGs) to enhance its efficacy against NAFL. In vivo experiments demonstrated that LPGs significantly improved the intestinal accumulation and retention of AXT. Pharmacokinetic evaluation further revealed tha

Identification of candidate antimicrobial peptides against the mandarin fish ranavirus using peptidomics analysis.

Viral diseases severely restrict the sustainable development of aquaculture, with Ranavirus exhibiting a broad host range and high pathogenicity but limited control options. Antimicrobial peptides (AMPs) are regarded as ideal antiviral candidates owing to their broad-spectrum bactericidal and antiviral properties. In this study, healthy and MRV-infected mandarin fish were used. Peptidomics analysis revealed 896 peptide fragments across skin (33), gill (289), liver (155), and head kidney (419). Based on APD, CAMPR3, and DBAASP, 58 candidate AMPs were identified and classified into α-helix (9),

Apigenin Enhances Hepatic Lipophagy through Regulating Lipid Droplet Size via the AKT-FHL2/PPARγ-CIDEC Pathway.

Large lipid droplets (LDs) resist lipophagy-mediated degradation due to steric hindrance, contributing to hepatic lipid accumulation in nonalcoholic fatty liver disease (NAFLD). Apigenin (API), a flavonoid abundant in fruits and vegetables, improves NAFLD, but its mechanism remains unclear. In this study, we found that API notably reduced LD size, thereby facilitating subsequent lipophagy. Transcriptomic analysis and validation revealed that API downregulated cell death-inducing DFF45-like effector C (CIDEC), a key promoter of LD fusion. Mechanistically, API suppressed CIDEC transcription by i

P3-1 Alleviates NAFLD by Regulating the Liver-Gut Axis and PPARα/RXRα Pathway in Mice.

Plant-derived peptides show hepatoprotective biological functions. An octapeptide (Gly-Phe-Cys-Asp-Arg-Met-Pro-Leu), termed P3-1, was isolated from macadamia nuts for the first time. This study evaluated the ameliorative effect of P3-1 on nonalcoholic fatty liver disease (NAFLD) and clarified the molecular basis of this action. The results showed that P3-1 attenuated hepatic pathological damage, improved lipid metabolism and liver function indices, and enhanced antioxidant capacity in NAFLD mice. In addition, P3-1 enhanced intestinal barrier function, thereby improving hepatic metabolic functi

Renal-Clearable Fluorogenic Nanoprobes for Molecular Imaging of Hepatic Drug-Metabolizing Cytochrome P450 2C19 Enzyme.

Despite the fact that cytochrome P450 2C19 (CYP2C19) is an essential xenobiotic-metabolizing enzyme in many drug metabolisms, the real-time and dynamic monitoring of its activity remains challenging due to the lack of highly specific activity-based probes with translational applicability. Herein, we report a library of O-alkylated hemicyanine derivatives and screen the optimal probe (CyB) with favorable sensitivity and specificity toward CYP2C19. CyB was subsequently selected to construct a dual-responsive nanoprobe (CyBPPD) that could be activated by hepatic CYP2C19 and glutathione, triggerin

Refinement of Structural Alerts for Hepatic Steatosis.

With the growing focus upon application of new approach methodologies (NAMs) within regulatory risk assessment of chemical substances, there is a clear need for human-relevant in silico models to predict toxicity associated with chemical exposure. This work describes the development of an in silico profiler for hepatic steatosis, a condition that results from the excess accumulation of fat within the liver. Toxicological data for chemicals were compiled from the literature, and expert judgments were made regarding their potential to induce steatosis. This list was used to test the 214 rules de

TRIP13 Promotes Aflatoxin B1-Induced Hepatocellular Carcinoma Progression by Inhibiting Ferroptosis.

Aflatoxin B1 (AFB1) is a potent foodborne hepatocarcinogen that contributes substantially to the global burden of hepatocellular carcinoma (HCC), yet the molecular mechanisms underlying AFB1-driven HCC progression remain incompletely understood. In this study, we identified thyroid hormone receptor-interacting protein 13 (TRIP13) as an important contributor to AFB1-related HCC progression and investigated its involvement in ferroptosis regulation. TCGA-LIHC, GSE25844, and single-cell RNA sequencing data (GSE166635) revealed that TRIP13 was significantly upregulated in AFB1-related HCC, enriche

Lactarius hatsudake Tanaka Polysaccharides Inhibit Hepatocellular Carcinoma by Modulating the Gut Microbiota and Its Metabolites.

We previously reported that Lactarius hatsudake Tanaka polysaccharide (LHP) can inhibit hepatocellular carcinoma, but the mechanism remains unclear especially gut microbe-mediated antitumor immunity. This research found that LHP and fecal microbiota transplantation were associated with reduced tumor growth. The ratio of CD8+/CD4+ T cells and levels of interferon-γ were increased following LHP intervention in the tumor, spleen, and blood. Additionally, in vivo and in vitro microbiome analyses indicated that LHP treatment significantly increased beneficial bacteria such as Bifidobacterium animal

Engineering a Galactosylated Zein-Orientin Nanoplatform for NAFLD Alleviation via Lipid Metabolic Modulation and Gut Microbiota Remodeling.

Nonalcoholic fatty liver disease (NAFLD) is characterized by excessive hepatic lipid accumulation, and the existing pharmacological alternatives are constrained by considerable side effects and probable toxicity. Orientin (Ori), a natural flavonoid with hepatoprotective potential, is restricted by poor solubility and low bioavailability. Herein, a galactosylated zein nanoplatform (Ori@Gal-Zein) was engineered via the Maillard reaction, resulting in uniform spherical nanoparticles (163.3 ± 2.6 nm) with high loading capacity of Ori (7.89 ± 0.14%) and gastrointestinal stability. A GSH and β-Galactosidase Sequential-Activated Dual-Locked Fluorescent Nanoprobe for Precise Liver Cancer Diagnosis and Intraoperative Navigation.

Accurate intraoperative navigation for solid tumors is often hindered by the nonspecific activation of imaging probes in non-target tissues. In this work, a near-infrared dual-locked fluorescent nanoprobe HCy-M@MSN@PEG was developed that can be sequentially activated by glutathione (GSH) and β-galactosidase (β-Gal), enabling highly specific imaging of liver tumors. This nanoprobe is constructed by encapsulating a β-Gal-responsive small-molecule probe (HCy-M) within PEGylated mesoporous silica nanoparticles (MSN@PEG). Upon exposure to the first biomarker GSH, the nanoprobe undergoes rapid disas

Hyperoside Derived from Actinidia arguta Bee Pollen Alleviates Zearalenone-Induced Hepatotoxicity and Oxidative Stress in Piglets via the PI3K/AKT Signaling Pathway.

Zearalenone (ZEA) is a prevalent mycotoxin inducing hepatotoxic effects. Hyperoside (HYP), a flavonoid derived from Actinidia arguta bee pollen, exhibits hepatoprotective properties against ZEA; however, the molecular mechanisms underlying this protection remain incompletely characterized. This study integrated in vivo piglet models, in vitro porcine hepatocytes, network pharmacology, and molecular docking to characterize the molecular targets of HYP. The results demonstrated that HYP significantly attenuated ZEA-induced hepatic lesions and oxidative stress. Computational analyses pinpointed t

Scavenger Receptor Class B Type 1 Mediated Transport Enables Delivery Across Capillarized Liver Sinusoids in Metabolic Dysfunction-Associated Steatohepatitis.

Liver fibrosis in metabolic dysfunction associated steatohepatitis (MASH) lacks effective therapies, in part because therapeutic agents poorly penetrate the capillarized sinusoidal endothelium. As fibrosis progresses, liver sinusoidal endothelial cells (LSECs) become defenestrated and restrict trans-sinusoidal exchange, thereby limiting drug access to hepatic stellate cells (HSCs) within the perisinusoidal space. Here, we developed a lipoprotein mimetic liposomal nanocarrier, SV@SRB-Lip, to exploit scavenger receptor class B type 1 (SR-B1)-associated transendothelial transport across capillari
