In Situ Visualization of Tumor and Drug-Induced Liver Injury with AIEgens, Polarity-Responsive, Membrane-Targeting Fluorescent Probes.

Dynamic changes in the polarity of the cellular microenvironment play a pivotal role in drug-induced liver injury (DILI) and tumors, serving as a crucial biomarker for evaluating cell membrane integrity and functional status. Precise monitoring of polarity changes is essential for elucidating the underlying mechanisms of injury. However, the real-time and specific visualization of local polarity remains a considerable challenge. Herein, polarity-sensitive fluorescent probes Probemem-1 and Probemem-2 were successfully developed. The structures adopted a D-π-A configuration, where a triphenylami

Hepatic-Renal Cascade Clearable Aminopeptidase N Probe for Rapid Detection of Early-Stage Liver Disease.

Early liver disease is a prolonged process with vague symptoms and evolving pathological causes, leading to a delayed diagnosis and poor therapeutic outcomes. The development of rapid and effective approaches for liver disease detection in both preclinical research and clinical diagnosis is in high demand yet remains challenging. Herein, we report an activatable fluorogenic probe (CyP-A) that is cleared via a hepatic-renal cascade for monitoring of hepatocyte injury and tissue fibrosis. After systemic administration, CyP-A preferentially accumulates in the liver, where it undergoes APN-mediate

Structure-Guided Design, Synthesis, and Evaluation of Novel 6-Methoxyquinoline-4-carboxamide Derivatives as First-in-Class Nano-Molar B3GAT3 Inhibitors for the Treatment of Hepatocellular Carcinoma.

Beta-1,3-glucuronosyltransferase (B3GAT3) is overexpressed in hepatocellular carcinoma (HCC) and correlates with poor prognosis, serving as a desirable anti-HCC target. Based on our micromolar lead TMLB-C16 (IC50 = 6.22-6.53 μM, KD = 3.96 μM), 60 novel derivatives were designed via systematic structure-activity relationship optimization. The optimized candidate TMLB-G9 is the first nanomolar B3GAT3 inhibitor bearing a 6-methoxyquinoline-4-carboxamide scaffold. It exhibits stronger B3GAT3 binding affinity (KD = 0.72 μM) than both TMLB-C16 and natural substrate UDP-GlcUA (KD = 1.32 μM). TMLB-G9

Hepatitis E virus genome plasticity: a marker of chronicity and therapy failure?

Hepatitis E virus (HEV) is a globally distributed zoonotic pathogen, spread by contaminated food and blood products, and a leading cause of acute viral hepatitis. Recent data indicate that the HEV genome contains a highly heterogeneous hypervariable region (HVR) that can tolerate extensive genomic rearrangements, especially among immunocompromised patients. These rearrangements can increase HEV replication in tissue-culture and have further been observed in HEV-infected patients. Insertions comprise either duplications of virus-derived sequences, or alternatively, host-derived sequence fragmen

Magnetic field exposure and liver disease: from current research to future directions.

As both an environmental factor and a therapeutic modality, magnetic field (MF) exposure exerts a significant regulatory effect on liver physiology and pathology. This review systematically synthesizes the current body of evidence regarding the modulatory roles of MFs in liver diseases (LDs) and elaborates on the underlying potential mechanisms. Initially, the biological basis of the interaction between MFs and hepatic tissue is delineated. Subsequently, preclinical evidence is consolidated, which demonstrates that MFs with specific parameter settings exhibit protective or therapeutic potentia

Hepatic Orosomucoid 2 aggravates demyelination and drives microglial DAM-like activation.

INTRODUCTION: Demyelination is a central pathological feature of white matter injury (WMI). Emerging evidence suggests that hepatokines act as systemic regulators of brain pathology, raising the possibility that liver-derived signals contribute to WMI. Here, we investigated whether hepatokine Orosomucoid 2 (ORM2) aggravates demyelination through liver-brain communication by modulating disease-associated microglia (DAM)-like activation in WMI. METHODS: ORM2 expression was assessed in the cuprizone (CPZ) demyelination model using RNA sequencing, quantitative PCR, western blotting, and immunoflu

Host defense peptide hepcidin 1 and iron overload differentially modulate energy, lipid, and hepatic-biliary metabolic pathways in gilthead seabream (Sparus aurata) blood.

Hepcidin 1 (a host defense peptide with antimicrobial and iron-regulating properties) and iron overload are relevant in fish physiology; however, the in vivo effects of their increase in healthy fish are poorly characterised. This study evaluated the effects of intraperitoneal injections of hepcidin 1, iron (iron-dextran), and sequential treatment (iron and hepcidin 1) on the blood metabolome of gilthead seabream (Sparus aurata) after 10 days. Multivariate analyses revealed weak metabolomic changes in all three conditions. In all treatments, 15 chemical superclasses were conserved, with the pr

Endoscopic ultrasound in hepatology: A European Society of Gastrointestinal Endoscopy (ESGE) Technical and Technology Review.

BACKGROUND: Endoscopic ultrasound (EUS)-guided techniques are increasingly applied in hepatology, including for tissue acquisition, vascular access, pressure measurement, and selected therapeutic interventions. However, adoption remains heterogeneous because of variability in technical approaches, devices, outcome definitions, operator expertise, and the quality of available evidence. METHODS: This European Society of Gastrointestinal Endoscopy (ESGE) Technical and Technology Review summarizes the current evidence on selected EUS-guided techniques in liver disease and portal hypertension, inc

Dengue virus disease phenotypes in C57BL/6N Ifnar1-/-Ifngr1-/- mice.

Dengue virus (DENV) remains a major global health problem, and experimentally tractable animal models are essential for investigating severe dengue pathogenesis and antiviral or vaccine strategies. However, whereas interferon receptor-deficient mouse models are used, the disease phenotype in C57BL/6N Ifnar1-/- Ifngr1-/- double-knockout (DKO) mice has not been sufficiently characterized. Here, we generated and characterized C57BL/6N-based DKO mice deficient in both type I and type II interferon receptors (Ifnar1-/- Ifngr1-/-). We first assessed susceptibility to DENV-2 NGC infection in wild-typ

APASL-ACLF without previous decompensation is immunologically distinct from EASL-CLIF-ACLF with previous decompensation.

BACKGROUND AND AIMS: Definitions of acute-on-chronic liver failure (ACLF) differ, with a focus on extrahepatic organ failures on the basis of previously compensated or decompensated cirrhosis in EASL-CLIF-ACLF versus hepatic failure in patients without previous decompensation in APASL-ACLF. Here, we explored whether patterns of systemic inflammation and immune cell alterations differ between these types of ACLF. METHODS: Consecutive patients with acute decompensation, EASL-CLIF- or APASL-ACLF were recruited from two liver centres. Patterns of systemic inflammation were characterized by Olink

Assessment and Reversibility of Hepatic Abnormalities in Heart Failure: Liver in Heart Failure.

Hepatic abnormalities in non-Fontan heart failure result from congestive hepatopathy and hypoperfusion-related ischemic injury, compounded by neurohormonal activation and systemic inflammation. Assessment of liver disease severity is challenging because standard liver biochemistries, serum fibrosis markers, severity score models, elastography, and imaging modalities have limited diagnostic accuracy in congestive hepatopathy, and hepatic fibrosis is often heterogeneous on histology in this population. Management focuses on optimization of cardiac function through medical therapy or mechanical c

A live-cell medium-throughput assay for quantifying lipid accumulation in hepatocytes.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a global health burden contributing to the high prevalence of associated metabolic diseases such as cardiovascular disease and thus global mortality rates. MASLD is diagnosed by ≥ 5 % lipid accumulation in the liver, and candidate therapies are often evaluated for lipid lowering effects. Current cell-based lipid imaging approaches rely on fixed-cell staining with dyes such as BODIPY 493/503, limiting the ability to dynamically monitor lipid accumulation in living hepatocytes. To gain a better understanding of the effect of dif

Microvasculopathy in spinal muscular atrophy: The endothelium as a target of SMN deficiency and an amplifier of disease.

Spinal muscular atrophy (SMA) is caused by deficiency of the ubiquitous survival motor neuron (SMN) protein. Degeneration of anterior horn motor neurons defines the disease, but many organs are affected, including heart, liver, kidney, and bone. The vascular compartment is among them: digital necrosis in infants with the fewest SMN2 copies, capillary rarefaction in SMA mice, and decreased von Willebrand factor with fibrinogen extravasation in human post-mortem spinal cord. Whether these reflect endothelial injury by SMN deficiency itself or capillary adaptation to atrophic, denervated tissue h

Advances in assessment systems and non-pharmacological intervention strategies for cancer-related fatigue following transarterial chemoembolization in hepatocellular carcinoma: a narrative review.

PURPOSE: Transarterial chemoembolization (TACE) is a primary treatment for intermediate to advanced hepatocellular carcinoma (HCC); however, cancer-related fatigue (CRF) is a prevalent and distressing symptom following TACE that significantly impairs patients' recovery and health-related quality of life. This review aimed to systematically summarize the current evidence on CRF screening, assessment, and non-pharmacological interventions specifically tailored to the post-TACE HCC population, and to propose an integrated, nurse-led clinical management framework. METHODS: A narrative review was

PAK1 Promotes Gluconeogenesis Via the ACSS2/KAT2A/PCK1 Complex to Attenuate Steatotic Liver Ischemia-Reperfusion Injury.

Ischemia-reperfusion (IR) injury is particularly severe in steatotic livers, substantially increasing the risk of graft failure. The growing use of steatotic grafts amid donor shortages has therefore heightened the clinical importance of this problem. The conserved serine/threonine kinase p21-activated kinase 1 (PAK1) regulates cell migration, growth, and redox homeostasis, but its role in steatotic liver IR injury remains unclear. To address this question, we examined the association between PAK1 expression in human steatotic liver grafts and clinical outcomes in 74 liver transplantation (LT)

A naturally occurring polyphenolic compound for liver cancer treatment.

As a polyphenolic compound naturally found in nature, piceatannol is an analogue of resveratrol. These compounds share a common structural motif but have various substituents. Resveratrol shows anti-inflammation and anti-tumor properties, but such biological activities of the active resveratrol metabolite - piceatannol are unclear. In this study, the anti-tumor properties of piceatannol were investigated through cell viability, colony formation, migration assay, detection of reactive oxygen species (ROS) and mitochondrial membrane potential (MMP) change, reverse transcription quantitative poly

Evidence Suggests Linoleic Acid Exerts Health Benefits Beyond Cardiovascular Disease: A Brief Review of Clinical and Observational Studies.

PURPOSE OF REVIEW: Questions challenging the healthfulness of diets high in linoleic acid (LA) and seed oils, which are major sources of this essential fatty acid, have recently arisen. These questions are especially relevant to adherents of plant-based diets as their LA intake is higher than the general population. Some maintain that LA increases inflammation and oxidative stress and for these reasons, despite its ability to lower circulating LDL-cholesterol levels when replacing saturated fat, does not lower cardiovascular disease (CVD) risk. Because most of the discussion about LA has focus

Passively Targeted Polymeric Nanocarrier Delivering STAT1 siRNA to the Liver Ameliorates Steatohepatitis and Fibrosis in the Thioacetamide Mouse Model.

Liver fibrosis is a major complication of chronic liver injury, driven in part by Signal Transducer and Activator of Transcription 1 (Stat1). This study evaluated passively liver-targeted polymeric nanocarriers delivering Stat1 siRNA (siSTAT1) to treat steatohepatitis and fibrosis in a thioacetamide (TAA) mouse model. Two polyplex nanocarriers with different core-corona ratios, EN15 and EN76, were tested in TAA-treated mice and activated human and murine hepatocytes. In vitro, both formulations enabled efficient cellular uptake and Stat1 silencing, reducing TAA-induced lipid accumulation. Thro

Effect of conditions and diseases associated with systemic inflammation on the plasma concentration of growth differentiation factor 15 (GDF15) in dairy cows.

Plasma growth differentiation factor 15 (GDF15) was analyzed in dairy cows during the transition period, following treatment with the endotoxin lipopolysaccharide (LPS), during heat stress, and in the presence of infectious diseases. Events associated with these conditions include systemic inflammation and reduced appetite. GDF15 may be relevant to these conditions given its discovery as an overexpressed cDNA in an activated immune cell line and its emergence as an anorexic signal. An i.v. LPS bolus to early lactating cows increased plasma GDF15 from a baseline concentration of 45.2 ± 7.7 pg/m
