Single Cell Analysis Reveals the Presence of Novel Intermediate Cells in Both Mice and Patients With Severe MASLD.

The landscape of cellular abnormalities occurring during metabolic dysfunction-associated steatotic liver disease (MASLD) is not completely clarified. We investigated cell heterogeneity involved in progressive MASLD at single cell resolution in mice fed AMLN. Single cell RNA sequencing (Sc-RNAseq) and spatial proteomics were applied to decipher cell populations and genes/pathways guiding MASLD progression. We identified 32 clusters, including hepatocytes (HEPs), hepatic stellate cells (HSCs), endothelial cells (ENDOs), Kupffer cells (KCs), and immune cells. HEPs clusters changed across disease

Therapeutic modulation of the gut-liver axis with faecalibacterium prausnitzii in metabolic dysfunction-associated steatohepatitis.

BACKGROUND: Faecalibacterium prausnitzii (F. prausnitzii) has attracted interest in recent research regarding its possible involvement in metabolic dysfunction-associated steatotic liver disease (MASLD). However, their causal relationship is still unclear. METHODS: Microbial biomarkers of gut microbiota associated with obesity-related phenotypes were identified from the GMrepo database. Information was retrieved on specific metabolites and host genes related to F. prausnitzii from the GutMGene database. Subsequently, a high-fat diet (HFD) was administered to C57BL/6 J mice for 16 weeks. The in

Awareness of metabolic dysfunction-associated steatotic liver disease (MASLD) among primary care providers in the four most populous U.S. cities.

INTRODUCTION AND OBJECTIVES: Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most common chronic liver disease but remains widely under-recognized in primary care. The 2023 shift from "nonalcoholic fatty liver disease" to MASLD emphasized metabolic dysfunction as a driver of disease but introduced new communication and educational challenges for primary care providers (PCPs). We aimed to assess PCPs' awareness, risk assessment, and management practices related to MASLD in the four most populous U.S. cities. MATERIALS AND METHODS: A cross-sectional online survey was con

Patients' Experience of Stigma as the Hidden Burden of Metabolic Dysfunction-Associated Steatotic Liver Disease: A Descriptive Qualitative Study with Thematic Analysis.

Background/Objectives: Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most prevalent liver disease globally, closely associated with obesity and metabolic syndrome. Despite its clinical significance, patients frequently experience stigmatization from society, healthcare professionals, and family, which may exacerbate psychological distress and hinder effective care. This study aims to explore how patients living with MASLD experience and interpret different dimensions of stigma. By examining stigma across healthcare, social, and personal contexts, the study offers insi

Human FGF1ΔHBS Gene Therapy as Treatment for Metabolic Dysfunction-Associated Steatohepatitis in ApoE-KO Mice.

The prevalence of metabolic dysfunction-associated steatohepatitis (MASH) is rising worldwide. hFGF1ΔHBS, a variant of human fibroblast growth factor 1 with three substitutions in its heparin-binding sites, was previously shown by our group to ameliorate fatty liver. However, hFGF1ΔHBS also significantly modulates systemic metabolism, making it unclear whether its hepatic benefits arise from direct liver-specific actions. Additionally, its poor pharmacokinetic profile underscores the need for alternative delivery strategies. Here, we employed adeno-associated virus serotype 8 under the thyroxi

Developmental and Seasonal Changes in Lipid Droplets and Fatty Acid Composition in the Ovary and Liver of Female Spotted Scat (Scatophagus argus).

The spotted scat (Scatophagus argus) is a fish species widely distributed in Indo-Pacific waters. In China, females are preferred for aquaculture due to their faster growth rates. Reproduction in teleost fish requires substantial energy, primarily in the form of lipids and fatty acids, which are closely linked to reproductive success and ovarian development. In this study, female spotted scat broodstock were sampled bi-monthly from May 2019 to March 2020 to examine annual ovarian development, lipid droplet accumulation, and fatty acid composition in the liver and ovaries. Body weight, total le

Loss of peroxisomal membrane proteins PEX13 and PEX14 disrupts fatty acid oxidation and drives lipid imbalance.

Peroxisomal disorders arise from severe peroxisome dysfunction and are frequently linked to liver pathology. Metabolic dysfunction-associated steatotic liver disease, which affects up to 38% of adults worldwide, has driven extensive efforts to identify genes that contribute to its development. In the present study, we investigated the role of two peroxisomal membrane proteins, PEX13 and PEX14, by performing single and dual small interfering RNA-mediated knockdowns in a liver cell line, HUH-7. Steatosis was induced using free fatty acids, and changes in lipid-metabolism gene expression were ass

Ketogenic Diet and Exercise Improve Peripheral Neuropathy in a Mouse Model of Metabolic Syndrome.

UNLABELLED: Metabolic syndrome (MetS) is a growing health concern that increases risk for peripheral neuropathy (PN), defined by nerve degeneration and sensory dysfunction. Although lifestyle strategies, like ketogenic diet (KD) and exercise, are known to mitigate MetS, their efficacy in maintaining nerve health or reversing established PN is less clear. First, in a maintenance paradigm, we tested whether long-term KD could maintain nerve health in mice, as opposed to a Western-style high-fat diet (HFD) that induces MetS PN. Second, in an intervention paradigm of mice with established MetS PN,

β-Hydroxybutyrate upregulates hepatic histone β-hydroxybutyrylation modification, promotes the expression of PPARα, and alleviates the hepatic steatosis in MASLD.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) stands as the most widespread chronic liver disorder globally. Histone β-hydroxybutyrylation (Kbhb)-a novel post-translational modification of histones driven by β-hydroxybutyrate (BHB)-has recently been recognized as a key epigenetic modulator. Our study aimed to explore how BHB influences the expression of hepatic lipid metabolism-associated genes in MASLD, and to determine whether histone Kbhb acts as the mechanistic mediator underlying these effects. METHODS: For in vivo experiments, db/db mice fed a high-fat die

Glyphosate-Induced Metabolic and Immune Modulation in Hepatoma Cells: Identification of Key Genes as Diagnostic and Therapeutic Targets Using an In Silico Systems Biology Approach.

Glyphosate, one of the most widely used herbicides worldwide, has raised significant concerns regarding its potential involvement in hepatotoxicity and molecular changes associated with liver cancer biology. These concerns highlight the need to better understand its underlying molecular mechanisms in hepatoma cells. Emerging evidence suggests that glyphosate exposure may increase the risk of liver cancer and chronic liver disease. However, the precise molecular alterations and promising biomarkers associated with glyphosate-induced hepatic toxicity and disease remain largely unexplored. In thi

Improvement of glucose homeostasis during leptin treatment does not alter the intestinal microbiome in male diabetic UC Davis type-2 diabetes mellitus rats.

Gut dysbiosis contributes to type 2 diabetes mellitus (T2DM) progression according to the preclinical evidence. Alterations in gut microbiome, energy metabolism, and barrier function were observed in individuals with obesity and insulin resistance. However, it remains unclear whether therapeutic interventions can reverse these alterations. This study aimed to evaluate whether improvements of glucose homeostasis resulting from leptin administration can lead to changes in colonic epithelial metabolism and barrier function in male UC Davis type 2 diabetic mellitus (UCD-T2DM) rats. Male UCD-T2DM r

The many pathways driving liver inflammation in MASH.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most prevalent chronic liver disease worldwide, affecting one-third of the global population. Most patients exhibit simple steatosis, whereas up to 20% develop metabolic dysfunction-associated steatohepatitis (MASH), potentially culminating in liver cirrhosis and hepatocellular carcinoma. Diverse parallel mechanisms contribute to the development of MASH, which are fueled by hepatic lipotoxicity, intestinal dysbiosis, and pro-inflammatory diets shaping innate and adaptive immune responses. Moreover, adipose tissue is drivin
