Gut microbiota shaping fatty liver disease progression.

The gut-liver axis is an important regulator of homeostasis, and its dysregulation has been implicated in the pathogenesis of metabolic dysfunction-associated steatotic liver disease (MASLD). In this review, we summarize current knowledge on both compositional alterations of the gut microbiome in MASLD and the functional consequences of these changes. We first discuss changes across major microbial domains, including bacteria, archaea, eukaryota, and bacteriophages, and then focus on microbiota-derived metabolites, such as short-chain fatty acids, trimethylamine and its derivative trimethylami

C. butyricum Combined With Resmetirom Provides Therapeutic Benefits in MASH Associated With Gut-Liver-Brain Axis Modulation.

BACKGROUND AND AIMS: Resmetirom has emerged as a major advance in MASH pharmacotherapy as the first approved liver-targeted THR-β agonist; however, variable therapeutic efficacy and adverse responses may limit its clinical benefits. This study investigated whether C. butyricum could provide additional therapeutic benefits when combined with resmetirom and attenuate nausea/vomiting-like responses in association with changes across the gut-liver-brain axis. METHODS: In this study, we established a high-fat diet-induced MASH mouse model and administered C. butyricum, resmetirom, or their combina

Sub‑chronic PFOA exposure at human‑relevant increasing doses in male mice: Role of stress‑induced gut barrier disruption in shaping intestinal and hepatic responses under high-dose conditions.

In humans, dietary intake is thought to be the main route of exposure to perfluorooctanoic acid (PFOA). The gastrointestinal tract is the primary entry point for this highly persistent and bioaccumulative environmental pollutant; however, its fate and effects within the digestive system remain largely unknown. Combining biology-based approaches with mass-spectrometry chemical imaging, our study investigated the impact of 90-day oral exposure to PFOA at a dose relevant to highly exposed human populations on an intact versus stress-impaired gut barrier in adult male mice. A ten-day corticosteron

Ultrasound-defined fatty liver and the risk of progression from preclinical to clinical obesity: a prospective cohort study.

BACKGROUND: According to the 2025 international consensus published in The Lancet Diabetes & Endocrinology, obesity can be conceptualized as a progressive disease with two stages: preclinical obesity, characterized by excess adiposity without established organ dysfunction, and clinical obesity, defined by the presence of obesity-related organ damage. However, determinants of the transition from preclinical to clinical obesity remain poorly understood. Fatty liver, a common manifestation of metabolic dysfunction, may represent an early indicator of this progression. METHODS: We analyzed 18,955

Metabolic Dysfunction-Associated Steatohepatitis and the Progression of Cognitive Decline in Older Thai People With HIV.

INTRODUCTION: Metabolic dysfunction-associated steatohepatitis (MASH) is more common in people with HIV and associated with systemic inflammation and extrahepatic complications, including cognitive impairment. While cross-sectional studies suggest an association between MASH and worse cognitive outcomes, its impact on longitudinal cognitive trajectories in people with HIV remains unclear. METHODS: This prospective study included people with HIV aged≥50 years from HIV-NAT, Bangkok, Thailand, between 2015 and 2024. MASLD and MASH were identified at baseline using the FibroScan-AST (FAST) score.

Metabolic benefits of ketohexokinase inhibition in individuals with MASLD: a randomized placebo-controlled crossover trial.

High fructose consumption, particularly from sugar-sweetened beverages, contributes to cardiometabolic disease. Here we study the metabolic effects of 6-week pharmacological inhibition with PF-06835919 of ketohexokinase, the enzyme that catalyses the first committed step of fructose metabolism, in a randomized placebo-controlled crossover trial in 15 participants with metabolic dysfunction-associated steatotic liver disease. The primary outcome was hepatic insulin sensitivity, measured by insulin-stimulated suppression of endogenous glucose production. Secondary outcomes included fat distribut

Temporal regulation of activin, follistatin, and growth differentiation factor 15 in the liver and circulation during high-fat diet-induced fatty liver disease.

Metabolic dysfunction-associated steatotic liver disease (MASLD) develops over time, yet most hepatokine studies are cross-sectional. We investigated the temporal regulation of the activin-follistatin axis and growth differentiation factor 15 (GDF15) in high-fat diet (HFD)-induced fatty liver disease. Mice were fed an HFD or normal chow for 8, 20, or 30 weeks. Plasma, hepatic mRNA, and hepatic protein were measured by ELISA, real-time PCR, and immunoblotting. HFD increased body and liver weights and hepatic triglyceride content throughout. A central finding was discordance between compartments

Metabolomic and lipidomic analyses reveal phosphatidylcholine-lysophosphatidylcholine axis alterations in sorafenib-persistent hepatocellular carcinoma.

Metabolic reprogramming and lipid remodeling are increasingly recognized as key processes underlying carcinogenesis and cancer treatment failure. The liver, as a central hub for lipid metabolism and homeostasis, is particularly susceptible to such alterations. Accordingly, hepatocellular carcinoma (HCC) exhibits extensive metabolic dysregulation and lipid remodeling, contributing to resistance to systemic therapies, including sorafenib. This study characterized histopathological and metabolic signatures across HCC carcinogenesis stages using a diethylnitrosamine/thioacetamide-induced rat model

Bariatric surgery may alleviate MASLD by restoring mitochondria-lipid droplet structural and functional integrity through suppression of the Mitochondrial Fission Factor-Perilipin 5 axis.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is one of the most prevalent chronic liver diseases worldwide, characterized primarily by excessive lipid droplet (LD) accumulation in hepatocytes. Bariatric surgery, particularly sleeve gastrectomy (SG), represents the most effective intervention for reversing MASLD, although the underlying mechanisms remain incompletely understood. This study aimed to investigate the mitochondrial and lipid droplets (LDs) related molecular mechanisms by which SG ameliorated MASLD. Using a high-fat diet (HFD)-induced mouse model of MASLD, combin

Designing PEDOT:PSS hydrogels for bioelectronic interfaces.

Conductive hydrogels have emerged as promising materials for wearable and implantable bioelectronic interfaces because they combine tissue-like mechanical properties with efficient signal transduction. Among them, poly(3,4-ethylenedioxythiophene):poly(styrene sulfonate) (PEDOT:PSS) has attracted significant attention owing to its dual ion-electron conductivity, biocompatibility, and solution processability. However, practical bioelectronic applications require hydrogel electrodes to simultaneously achieve high conductivity, mechanical robustness, strong adhesion, self-healing capability, envir

Non-pharmacological treatment of MASLD.

Non-pharmacological treatment of metabolic dysfunction-associated steatotic liver disease (MASLD) aims at modifying lifestyle patterns that promote metabolic dysfunction and hepatic fat accumulation. These include unhealthy dietary habits, which are characterised by excess energy intake and a qualitatively poor diet that is high in ultra-processed food, saturated fat, free sugar, red and processed meat and/or sweetened beverages, and low in fibre. In addition, sedentary habits, and sleep and circadian rhythm disruption have been identified as detrimental lifestyle factors. Lifestyle interventi

Evaluation of Stigma Toward Fatty Liver Disease.

BACKGROUND AND AIMS: A recent Delphi statement proposed an adapted term from metabolic dysfunction-associated fatty liver disease (MAFLD), metabolic dysfunction-associated steatotic liver disease (MASLD), substituting "fatty" with "steatotic", as the former was claimed to be stigmatizing. In this cross-sectional study, we aimed to investigate this and understand stigma and awareness among fatty liver disease patients. METHODS: A semi-structured interview consisting of questions was carried out with patients to get an understanding of their knowledge of fatty liver disease and their opinions o

Chlorpyrifos and its metabolite 3,5,6-trichloro-2-pyridinol elicit MASLD-related lipid metabolic perturbations in HepG2 cells.

Metabolic dysfunction-associated steatotic liver disease (MASLD) has been increasingly linked to environmental chemical exposure, yet the metabolic consequences of pesticide parent-metabolite co-exposure remain poorly understood. HepG2 cells were exposed for 48 h to chlorpyrifos (CPF, 10 mg/L), its major metabolite 3,5,6-trichloro-2-pyridinol (TCP, 1.25 mg/L), or their combination (CTCP). Network toxicology was applied for hypothesis generation, followed by integrated lipidomics and transcriptomics analyses. Under the selected exposure conditions, TCP induced greater lipid-droplet accumulation

The Evolution of Pediatric MAFLD Research (2014-2023): A Decade-Long Bibliometric Analysis of Emerging Trends.

BACKGROUND: Metabolic dysfunction-related fatty liver disease (MAFLD) has emerged as the predominant chronic liver disorder among children and adolescents. Like in adults, pediatric MAFLD encompasses a disease spectrum progressing from isolated steatosis to inflammatory changes, fibrotic development, and ultimately, cirrhosis. Despite increasing recognition of MAFLD as a major pediatric health issue, current literature lacks a systematic quantitative evaluation of research trends, leading to knowledge gaps in this field. To address this limitation, a comprehensive bibliometric analysis was per
