Rapamycin Reverses the Hepatic Response to Diet-Induced Metabolic Stress That Is Amplified by Aging.

Aging is associated with increased susceptibility to metabolic stress and chronic liver disease, yet the interactions between age and metabolic stressors and the potential for ameliorating interventions remain incompletely understood. Here, we examined the hepatic response of young (7-month-old) and old (25-month-old) C57BL/6 male mice to a 9-week high-fat diet (HFD) and assessed whether rapamycin, a well-established pro-longevity intervention, could mitigate age-exacerbated effects. While both age groups developed metabolic-associated steatohepatitis (MASH), older mice displayed more severe h

Impact of bariatric surgery on the gut microbiota and metabolomic profile of the enterohepatic axis in an experimental model of obesity.

Obesity is a complex metabolic disease associated with alterations in the gut microbiota and the metabolism of fatty acids and bile acids. Bariatric surgery can modify this axis, contributing to benefits beyond weight loss, and the response may differ between surgical techniques. This study investigates this differential impact in an experimental model of obesity. Both surgical procedures reduced body weight, although gastric bypass (GB) induced a more sustained weight loss and deeper metabolic changes. GB significantly decreased butyrate levels while increasing bile acids and the abundance of

A high fermentable fiber Western diet reduces indole levels.

Changes in gut microbiota composition due to diet impact health. Fiber-rich diets promote beneficial microbiota and reduce the risk of metabolic diseases, while low-fiber, calorie-dense diets are linked to dysbiosis and increased disease risk. This study examines the effects of a Western diet (WD) and explores dietary fiber supplements as potential modifiers of those effects. 10-week-old C57Bl/6J male mice were fed control (low-fat) or WD (high-fat, high-sucrose) containing 0% fermentable fiber (FF) or WD supplemented with 20% FF (fructooligosaccharides, FOS; guar gum, GG, or pectin, Pec). Multi-omics analysis of the gut-liver axis revealed the effects of different exercise interventions in NAFLD mice.

BACKGROUND: Growing evidence indicates that gut microbiota may play a potential role in NAFLD. Exercise was recognized as an effective intervention for NAFLD, but the effects of these exercise modalities on gut microbiota in NAFLD remain unclear. RESULTS: C57BL/6J mice were divided into a normal diet control group (NC) and a high-fat diet (HFD) group. After 8 weeks on the HFD, the mice were further divided into three groups: HFD, HFD + AE (HAE), and HFD + RE (HRE), and treated for 12 weeks. Body weight and liver weight of the mice were measured, and samples (blood, liver, colon, and feces) wer

High-intensity interval training induces lactate dehydrogenase B lactylation to inhibit hepatic lipogenesis to alleviate metabolic dysfunction-associated fatty liver disease.

BACKGROUND: De novo lipogenesis (DNL) is an important cause of lipid deposition in metabolic dysfunction-associated fatty liver disease (MAFLD), and recent studies report that lactate is a major substrate for DNL in MAFLD. High-intensity interval training (HIIT) has a powerful regulatory effect on lactate metabolism, but whether it can alleviate MAFLD by regulating lactate metabolism has not been reported. METHODS: High-fat diet (HFD)-induced MAFLD mice were used as a model and underwent performed 8 weeks of HIIT. Serum and liver tissues were collected and analysed by biochemical assay, histo

Inflammatory Factors Derived From Metabolic Dysfunction-Alcoholic Fatty Liver Disease: Inducers of Anxiety and Spatial Memory Impairment.

Metabolic dysfunction-associated steatotic liver disease (MASLD) has emerged as a global epidemic, with growing evidence suggesting its adverse impact on brain function. However, the underlying mechanisms linking hepatic metabolic dysfunction to neurodegeneration remain unclear. In this study, we systematically investigated the liver-brain axis by integrating genetic epidemiology, experimental neuroscience, and transcriptomics techniques. Two-sample Mendelian randomization (MR) analysis revealed a potential causal relationship between MASLD and cognitive decline. These findings were validated

Liver Metabolomic Profiling Reveals Distinct Signatures Between Steatosis and Metabolic Dysfunction-Associated Steatohepatitis.

BACKGROUND AND AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD), the most common chronic liver disease worldwide, is closely linked to obesity and metabolic syndrome. The reason some patients with MASLD develop metabolic dysfunction-associated steatohepatitis (MASH) and which metabolic changes in the liver are linked to its progression are unclear. METHODS: A non-targeted metabolomics analysis was performed on liver samples from 106 Finnish patients with severe obesity (71 females, mean age ± SD: 48.6 ± 8.7 years, body mass index: 41.6 ± 5.2 kg/m2) selected for laparosco

Distinct fibrin clot characteristics in individuals with severe obesity and metabolic liver disease: 2-year follow-up after bariatric surgery.

BACKGROUND: Obesity predisposes individuals to metabolic dysfunction-associated steatotic liver disease (MASLD), the hepatic manifestation of metabolic syndrome. Studies in mice suggest that fibrin deposition in adipose tissue and liver promotes obesity and MASLD, but whether similar mechanisms are linked to metabolic disease in humans is incompletely studied. OBJECTIVES: This study determined the relationship between plasma fibrin clot characteristics and hepatic fibrin deposition with obesity and MASLD severity and examined changes in fibrin measures 2 years after bariatric surgery. Amelioration of hepatic steatosis in male obese rats by high-protein diet is dependent upon protein source.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is characterized by triglyceride accumulation and insulin resistance. Currently, weight loss remains the primary strategy for reducing liver fat. High-protein diets (HPDs) may improve metabolism, lower body weight, and reduce hepatic fat; however, the effects of specific protein sources are largely unknown. This study examines the effects of HPDs from animal and plant protein sources, separately and in combination, on hepatic steatosis and MASLD-related metabolic pathways. Obese fa/fa Zucker male rats received HPDs (35% kcal from

Effects of chitosan supplementation versus placebo on liver fat and metabolic parameters in patients with non-alcoholic fatty liver disease: a randomized controlled trial protocol.

BACKGROUND: Nonalcoholic fatty liver disease (NAFLD) is a common chronic liver disorder affecting about one-third of the global population. It encompasses a spectrum from simple steatosis to nonalcoholic steatohepatitis (NASH) and may progress to fibrosis and cirrhosis. Chitosan oligosaccharide (COS) has recently emerged as a potential therapeutic agent with antioxidant and anti-inflammatory properties; however, most evidence comes from animal studies. Further clinical research is therefore warranted to evaluate its efficacy in humans. METHOD: This double-blind, randomized, placebo-controlled

Eicosapentaenoic and Docosahexaenoic Acid Levels in Mouse Tissues After Intake of Echium and Ahiflower Oils Rich in Stearidonic and α-Linolenic Acids.

Omega-3 fatty acids (n-3 FA), particularly eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), have been consumed aiming to reduce cardiovascular disease. Fish oil is the main dietary source of EPA and DHA but has limitations that have stimulated interest in sustainable alternatives such as Echium ( Echium plantagineum ) and Ahiflower ( Buglossoides arvensis ) oils, both rich in α-linolenic acid (ALA) and stearidonic acid (SDA), precursors of EPA. However, the ability of these two oils to increase EPA and DHA levels in different tissues remains unclear. Thus, this study aimed to invest

Cross-organ communication based on non-coding RNA networks leads to cognitive dysfunction in mice.

BACKGROUND: Beyond regulation of mRNAs by individual ncRNA types, like miRNAs, circRNAs or lncRNAs, their interplay as competitive endogenous RNAs (ceRNAs) appears pivotal in diseases. ceRNAs are promising biomarkers in diagnosing diseases as well as in understanding off-target drug mechanisms or developing specific therapeutics. Many diseases are influenced at different molecular levels and across multiple organs, making cross-organ multi-omics interaction studies necessary to holistically understand diseases and their regulation. In our study we applied this approach to the cognitive dysfunc

Longitudinal changes in vibration-controlled transient elastography in pediatric fatty liver disease.

OBJECTIVES: There is a lack of research describing changes in vibration-controlled transient elastography (VCTE) parameters in pediatric metabolic dysfunction-associated steatotic liver disease (MASLD). We hypothesized that VCTE measurements would be positively associated with changes in anthropomorphic and biochemical parameters in pediatric MASLD over time, making VCTE a useful tool to monitor MASLD in children. METHODS: This was a retrospective chart review of children under 18 with MASLD evaluated at Boston Children's Hospital from 2015 to 2023. Demographic, biochemical, and anthropometri
