Influence of Heart Function on the Assessment of MASLD Using Indocyanine Green Clearance.

INTRODUCTION: The incidence of MASLD is rapidly increasing while the accuracy of noninvasive tests for MASLD remains limited. Indocyanine green clearance is a dynamic liver function test established for the evaluation of liver disease but is limited by the major influence of systemic circulation. We aimed to assess the role of Indocyanine green clearance in the evaluation of MASLD, together with cardiac function measurements. METHODS: We conducted a prospective controlled study of 77 patients who underwent metabolic and bariatric surgery (MBS) and 25 controls who underwent elective cholecystec

Two Sides of a Coin: Molecular, metabolic, and Phenotypic Convergence in Pediatric Undernutrition and Obesity.

PURPOSE OF REVIEW: This narrative review juxtaposes the metabolic and molecular consequences of pediatric under- and overnutrition, highlighting the similarities and differences between these two nutritional states occurring simultaneously in different parts of the world. RECENT FINDINGS: Numerous biological changes in pediatric acute undernutrition and obesity have been linked to elevated risks of chronic metabolic disorders. We summarize recent evidence on pathophysiological pathways and outcomes common to both conditions. Despite etiological divergence, early-life nutritional imbalances con

Indole-3-carbinol ameliorates ER stress-mediated hyperleptinemia in western diet-fed apoE-/- mice.

BACKGROUND: Endoplasmic reticulum (ER) stress during overnutrition causes leptin resistance in obese animals and humans. ER stress induces the activation of the unfolded protein response, which disrupts the leptin signaling pathway, accelerating atherosclerosis development and its complications. OBJECTIVE: Indole-3-carbinol (I3C) improves metabolic dysfunction in diet-induced obesity; however, its role in protecting against ER stress-induced hyperleptinemia remains unclear. Herein, we explored whether dietary I3C alleviates ER stress in apolipoprotein E-deficient (apoE-/-) mice fed a western

Gestational PFHxS exposure at environmentally relevant levels is associated with adult hepatic steatosis and PERK/SREBP1 pathway activation.

Perfluorohexane sulfonate (PFHxS), an increasingly detected environmental pollutant, poses potential metabolic health risks, yet its developmental toxicity at environmentally relevant doses remains poorly understood. Here, we investigated the long-term effects of PFHxS exposure on the hepatic development using a mouse model with oral administration of 0.03 and 0.3 μg/kg/day throughout pregnancy. Our findings demonstrated that gestational PFHxS exposure induced persistent hepatic oxidative stress and was associated with the activation of the endoplasmic reticulum stress response (PERK/eIF2α/ATF

Cannabidiol and cannabigerol ameliorate steatotic liver disease via phosphocreatine buffering and lysosomal restoration.

BACKGROUND AND PURPOSE: Cannabidiol (CBD) and cannabigerol (CBG) are non-psychoactive phytocannabinoids with emerging therapeutic potential in metabolic dysfunction-associated steatotic liver disease (MASLD). However, the molecular mechanisms underlying their beneficial effects remain incompletely understood. In this study, we assessed the metabolomic and lipidomic impact of CBD and CBG in a mouse model of diet-induced obesity and MASLD. EXPERIMENTAL APPROACH: Male C57Bl/6 mice fed on a high-fat diet for 14 weeks were treated for 4 weeks with daily intraperitoneal CBD, CBG or vehicle. Assessm

LYPLAL1 rare loss-of-function variants in humans and deletion in human hepatoma cells protect against MASLD.

Genetic variants near LYPLAL1 are associated with Metabolic dysfunction-Associated Steatotic Liver Disease (MASLD) in humans, but their impact on LYPLAL1 function is unknown. We identified LYPLAL1 loss-of-function variants from UK BioBank (UKBB) whole-exome sequencing data that had AlphaMissense or GPN-MSA scores in the top 20% of LYPLAL1 variants for being disruptive. We aggregated these variants and carried out burden analysis for effects on MRI Proton Density-Fat Fraction (MRI-PDFF) and ICD-based MASLD in UKBB. Rare loss-of-function LYPLAL1 variants were associated with reduced MRI-PDFF and

Effects of Pancreatic Resection on Liver Fat Content and Amino Acid, Lipid, and Glucose Metabolism: A Prospective 1-Year Follow-up Study.

UNLABELLED: Individuals who undergo pancreatic resection are at increased risk of developing hepatic steatosis. Glucagon is a key regulator of hepatic glucose, amino acids, and lipid metabolism, and the change in circulating glucagon is suggested to contribute to the pathogenesis of postoperative steatotic liver disease. Here, we aimed to elucidate hepatic and metabolic changes induced by pancreatic resection. Fifty individuals scheduled to undergo pancreatic surgery were recruited and evaluated by blood samples and a liver biopsy obtained during surgery. One year after surgery, 21 eligible pa

Early fecal metabolomic profiling for predicting acute graft-versus-host disease following allogeneic hematopoietic stem cell transplantation.

Acute graft-versus-host disease (GVHD) is a major cause of morbidity and mortality after allogeneic hematopoietic stem cell transplantation (allo-HSCT). Reliable early, noninvasive biomarkers for GVHD prediction remain limited. We conducted targeted fecal metabolomic profiling using liquid chromatography-tandem mass spectrometry in 77 allo-HSCT recipients at two time points: before conditioning (S1) and immediately after conditioning but prior to donor T-cell engraftment (S2). We assessed changes in metabolite profiles and their association with subsequent GVHD development. A multivariate logi

APOE4 drives widespread changes to the hepatic proteome and alters metabolic function.

Apolipoprotein E (APOE) is essential for lipid homeostasis and has been extensively studied in Alzheimer's disease (AD). Individuals carrying an APOE4 allele have an increased risk of AD and exhibit deficits in energy metabolism, including glucose utilization and mitochondrial dysfunction. While the role of APOE in the liver is well characterized, the impact of APOE genotype on hepatic health and metabolism remains poorly understood. We sought to investigate this using young APOE3 and APOE4-targeted replacement mice and isogenic-induced pluripotent stem cell (iPSC)-derived hepatocyte-like cell

Maternal docosahexaenoic acid and eicosapentaenoic acid supplementation: effects and mechanisms on lipid metabolism in the offspring.

Environmental factors, such as nutrition, hormones, and metabolites, which are present in early stages of life, have long-lasting effects throughout an organism's lifespan, and an abnormal nutritional environment throughout gestation and lactation may significantly increase the possibility that offspring will develop chronic metabolic disorders. The important nutrients docosahexaenoic acid (C22:6n-3, DHA) and eicosapentaenoic acid (C20:5n-3, EPA), which are essential long-chain omega-3 polyunsaturated fatty acids, contribute to proper neurological and retinal development and exhibit both anti-

Immunometabolism of IRF4 in adipose, muscle, and immune cells influences obesity and MASLD.

Interferon regulatory factor 4 (IRF4) connects immunity and metabolism in immune cells and parenchymal cells of metabolic tissues. In immune cells, IRF4 is a metabolic rheostat that promotes glycolysis to support macrophage polarization and increase oxidative phosphorylation to enable T cell proliferation and memory, while limiting transcription of pro-inflammatory programs. IRF4 also dictates cell-autonomous responses in adipocytes and muscle cells, including transcriptional regulation of lipolysis in white adipocytes, thermogenesis in brown adipocytes, and glucose and amino acid metabolism i

Roles of mitochondrial complexes in non-alcoholic fatty liver disease.

Non-alcoholic fatty liver disease (NAFLD) is increasingly recognized as a mitochondrial-driven metabolic disorder, yet the specific contributions of individual mitochondrial respiratory chain complexes remain poorly defined. In particular, inconsistent alterations in complexes I-V have been reported across different NAFLD models, representing a critical knowledge gap. Here, we systematically reviewed in vivo and in vitro studies to evaluate changes in mitochondrial complexes I-V during NAFLD progression. Overall, NAFLD is commonly associated with reduced complex activity, impaired mitochondria
